Prototype strain, M-25 , of parainfluenza type 4 (PI-4) virus was grown to high titers and passaged serially in Vero (Cercopithecus kidney cell line) cell cultures under fluid overlay containing low concentrations of trypsin. Infectivity titers in both primary cercopithecus kidney cell cultures (VK) and in Vero cell cultures under ordinary conditions (Vro) did not change essentially through passages in the trypsin-added Vero cell cultures (Vrt) from those of the original VK-grown virus. The 14th Vrt-passage virus was not transferable in Vro beyond two subpassages. Consequently, occurrence of adaptation to Vro during passages in Vrt can be excluded. Identification of V rt-grown virus as PI-4 was demonstrated by the cross neutralization and hemagglutination-inhibition tests performed among paramyxoviruses and b y several biological a nd physico-chemical tests. Other proteolytic enzymes , pancreatin and pronase, gave similar results as did trypsin, but hyaluronidase and lysozyme were ineffective. It was found that animal sera were inhibitory to the trypsin-effect mentioned above. Therefore, the Vrt system must be serum-free for obtaining reproducible results .
To assess the differential stimulus to central and intravascular osmoreceptors during recovery from thermal dehydration, we measured Na concentrations in cerebrospinal fluid ([Na]CSF) and plasma ([Na]p) continuously and compared these during simulated drinking by gastric water infusion (INF) in euhydrated and thermally dehydrated rats under anesthesia. Continuous measurement of [Na]CSF was obtained with a double-barreled Na electrode placed in the lateral ventricle. Continuous measurement of [Na]p was obtained from a flow cell Na electrode in an extracorporeal shunt. Measurements were made during 10 min of INF (2.5 ml/100 g body wt) into the stomach and during 20 min of recovery. Changes in [Na]CSF always lagged behind those in [Na]p and were quantitatively smaller after INF. The decrease in [Na]CSF occurred sooner in dehydrated than in euhydrated rats in response to the decrease in [Na]p (P < 0.01). These results suggest that water and/or Na movement between blood and CSF is accelerated during restitution from thermal dehydration, acting to prevent overhydration during the early phase of rehydration.

We have studied methods for testing the neurovirulence of live poliovaccine viruses by intraspinal inoculation into mice carrying the human poliovirus receptor gene (Tg mice). A comparison of the neurovirulence of Sabin type 3 vaccine virus and related viruses using the 50% paralysis dose determined after intraspinal inoculation into the Tg mice as an index revealed a close correlation between the results of the paralysis dose in Tg mice, the neurovirulence expressed by the histopathological lesions core in monkeys, and the temperature sensitivity of the viruses. The results of experiments in the Tg mice also showed a good correlation with the number of mutations at position 472 from U to C in the 5' noncoding region in the genomes of the viruses tested. These results strongly suggest that the neurovirulence test for oral poliomyelitis vaccine using the Tg mice is an excellent method and may be used in place of the test using monkeys.
