The interactions between maize, sorghum, wheat and rice plants and Herbaspirillum seropedicae were examined microscopically following inoculation with the H. seropedicae LR15 strain, a Nif(+) (Pnif::gusA) mutant obtained by the insertion of a gusA-kanamycin cassette into the nifH gene of the H. seropedicae wild-type strain. The expression of the Pnif::gusA fusion was followed during the association of the diazotroph with the gramineous species. Histochemical analysis of seedlings of maize, sorghum, wheat and rice grown in vermiculite showed that strain LR15 colonized root surfaces and inner tissues. In early steps of the endophytic association, H. seropedicae colonized root exudation sites, such as axils of secondary roots and intercellular spaces of the root cortex; it then occupied the vascular tissue and there expressed nif genes. The expression of nif genes occurred in roots, stems and leaves as detected by the GUS reporter system. The expression of nif genes was also observed in bacterial colonies located in the external mucilaginous root material, 8 days after inoculation. Moreover, the colonization of plant tissue by H. seropedicae did not depend on the nitrogen-fixing ability, since similar numbers of cells were isolated from roots or shoots of the plants inoculated with Nif(+) or Nif(-) strains.
RESUMO -(Inter-relações entre a anatomia vegetal e a produção vegetal). É realizada uma revisão de literatura que procura aproximar a anatomia vegetal do contexto da produção vegetal. RESUMO -Neste trabalho quantificaram-se as principais alterações histológicas ocorridas em cultivares de capim-elefante (Pennisetum purpureum Schumach.), em três estádios de desenvolvimento. A degradação dos tecidos foi avaliada após incubação em líquido ruminal de bovinos. As porcentagens de tecidos presentes em colmo, quilha, limbo e bainha foliares foram determinadas. A quilha e o colmo apresentaram maior proporção de tecido lignificado, enquanto o limbo foliar, maior quantidade de tecido epidérmico e tecido vascular não-lignificado. O tecido parenquimático foi encontrado em menor proporção na bainha foliar, principalmente pela presença do aerênquima, a partir da segunda coleta. A proporção de tecido lignificado aumentou com a maturidade do vegetal, sendo mais acentuado em colmos e limbos. Entre as principais alterações, destaca-se a grande área de degradação encontrada na bainha foliar, mesmo com o envelhecimento dos tecidos. Isto foi associado à presença do aerênquima encontrado nos estádios de desenvolvimento mais avançados.Os estômatos favoreceram a penetração dos microorganismos nos tecidos mais internos da folha (mesofilo). O espessamento e a lignificação da parede celular ocorreram com o envelhecimento das plantas, acompanhado de redução na área de degradação dos tecidos.Palavras-chave: anatomia vegetal, degradação de tecidos, forrageira, lignina, parede celular, ruminante Quantitative Anatomy and in vitro Tissue Degradation in Elephant Grass(Pennisetum purpureum Schumach.) Cultivars ABSTRACT -In this research, the main histological changes that occurred in cultivars of elephant-grass (Pennisetum purpureum Schumach.) at three development stages were determined. The tissue degradation was evaluated after incubation in cattle ruminal fluid. The percentage of tissues presented in the stem, keel, leaf limbo and sheath were determined. The keel and the stem presented greater proportion of lignified tissues, while the leaf limbo presented higher amount of epidermal and non-lignified vascular tissues. The parenchymal tissue was found in minor proportion on the leaf sheath, mainly by the presence of aerenchyma, from the second collection. The proportion of lignified tissues increased with the grass maturity, being more accentuated in stems and limbo. Among the main changes, stand out the greater degradation area found for leaf sheath, even with the tissue become older. This was associated with the presence of aerenchyma found in the advanced development stages. The stomata propitiated intrude of the microorganisms in the most internal leaf tissues (mesofilo). Summary• The chemical composition and structure of the layers of the external coat and the three-dimensional organization of the gel during the swelling process are reported here for seeds of Magonia pubescens .• Histochemical-light and scanning electron microscopy (SEM) were used in conjunction with CP-MAS (cross polarization -magic angle spinning: solid state) 13 C NMR spectroscopy to analyse the seed coat layers.• M. pubescens seeds extrude a gel from the second of five regions of the external seed coat when immersed in water. This gel, visualized by SEM, had an extruded cylindrical, laminated structure. Staining revealed two anionic components in the cells of the second layer; the cytoplasm showed the presence of carbohydrates containing vicinal hydroxyl groups and at least some of these carbohydrates were acidic polysaccharides. Part of the β -linked glucan present in this layer was cellulosic material. Phenolic compounds were not detected in the second layer, but existed in other layers. Lignin and/or tannins were shown in the cell walls of the three innermost layers by CP-MAS 13 C NMR spectroscopy.• The solid-state NMR technique had the advantage of being more sensitive and quantitative compared with light microscropy and might prove to be a useful tool in investigating cell components.
RESUMO -(Anatomia comparada das folhas de espécies de Peperomia (Piperaceae) -I. Ontogênese do tecido aqüífero e dos estômatos). A maioria das espécies de Peperomia apresenta tecido que reserva água na face adaxial de suas folhas. Esse tecido pode variar em espessura, determinando maior ou menor suculência à folha. A fim de se determinar a origem de estruturas anatômicas relevantes ao grupo, o presente trabalho descreveu o desenvolvimento do tecido especializado na reserva de água, bem como a ontogênese dos estômatos de P. catharinae Miquel, P. emarginella (Sw.) C.DC., P. quadrifolia (L.) Kunth e P. rotundifolia (L.) Kunth. Para tanto, foram observadas folhas jovens do ápice caulinar e do primeiro nó subseqüente, em secções transversais e paradérmicas. As técnicas utilizadas foram as usuais para microscopia fotônica. Todas as espécies apresentaram tecido especializado na reserva de água, originado de divisões periclinais das células protodérmicas. A ontogênese estomática é mesoperígena, dando origem a estômatos anisocíticos, tetracíticos e estaurocíticos.Palavras-Chave -Peperomia, epiderme múltipla, ontogênese estomática ABSTRACT -(Comparative leaf anatomy of Peperomia (Piperaceae) -I. Ontogenesis of the aquiferous tissue and the stomata). Most species of Peperomia presents a water reservoir tissue on the adaxial surface of the leaf. This tissue can vary in thickness determining more or less succulence to the leaf. On purpose of determining the origin of important anatomic structures for this group, the present work describes the development of the water reservoir specialized tissue, as well as the stomata ontogenesis for P. catharinae Miquel, P. emarginella (Sw.) C.DC., P. quadrifolia (L.) Kunth and P. rotundifolia (L.) Kunth. Young leaves from the stem apex and the first node were observed in cross and paradermic sections. The techniques were the usual for light microscopy. All the species presented the water reservoir specialized tissue originated from periclinal divisions of protodermal cells. The stomata ontogenesis is mesoperigenous and it gives rise to anisocytic, tetracytic and staurocytic stomata.
O objetivo do presente trabalho foi determinar a composição química e a digestibilidade das folhas (lâminas e bainhas) e caules de cultivares das espécies Axonopus scoparius e Axonopus fissifolius, em dois estádios de desenvolvimento. O delineamento experimental foi inteiramente casualizado com dois tratamentos (seis e vinte semanas de crescimento) e cinco repetições. Para determinação da cinética de digestão, utilizou-se a técnica do saco de nylon, e a composição bromatológica foi determinada através da técnica do infra-vermelho (NIRS). A degradação dos tecidos foi observada histologicamente após a incubação de pequenas porções de caule e folhas em líquido ruminal de bovinos. Observou-se que, com a maturidade, os cultivares de A. scoparius apresentaram maior queda na taxa de degradabilidade efetiva, a qual foi relacionada com o espessamento da parede celular e aumento da área de tecido lignificado. O teor de proteína bruta foi o parâmetro mais afetado com o envelhecimento das espécies. A maior área de tecidos lignificados, encontrada nas frações foliares (limbo, quilha e bainha) de A. fissifolius, resultou em menores taxas de digestibilidade para os cultivares dessa espécie, em relação aos de A. scoparius. O cultivar "verde" de A. scoparius apresentou maior suscetibilidade à degradação ruminal, quando jovem, comparado ao cultivar "roxo", enquanto os de A. fissifolius apresentaram características similares ante à degradação ruminal. O caule foi a fração cuja degradação decresceu em maior proporção quando comparado às frações foliares.
-(Comparative anatomy of leaves and stems of Axonopus scoparius (Flügge) Kuhlm. and Axonopus fissifolius (Raddi) Kuhlm. (Poaceae)). Several species in the genus Axonopus are commonly used as forage milk cattle to feed in southern Brazil. However, little is known about the anatomy of these plants. In this work, the structural features of leaves and stems of Axonopus scoparius (Flügge) Kuhlm. and A. fissifolius (Raddi) Kuhlm. are described. The samples were collected in Itajaí, SC and processed for light and scanning electron microscopy histological studies. The Kranz structure was found in both species. The species could be distinguished by the presence of bulliform cells in the leaf blade upper epidermis of A. fissifolius and the absence of these cells in A. scoparius and by the presence of unicellular macrohairs in the leaf blade upper epidermis of A. scoparius and the absence of these strutures in A. fissifolius.Key words -Axonopus fissifolius, Axonopus scoparius, forage, Kranz, plant anatomy RESUMO -(Anatomia comparativa de folhas e caules de Axonopus scoparius (Flügge) Kuhlm. e Axonopus fissifolius (Raddi) Kuhlm. No presente trabalho foram descritas as características estruturais de folhas e caules das espécies Axonopus scoparius (Flügge) Kuhlm. e A. fissifolius (Raddi) Kuhlm. As amostras foram coletadas em Itajaí, SC e processadas para estudo histológico ao microscópio de luz e eletrônico de varredura. A estrutura Kranz foi evidenciada em ambas espécies. As espécies distinguem-se pela presença de células buliformes na epiderme da face adaxial da lâmina foliar de A. fissifolius e pela ausência destas células em A. scoparius e pela presença de tricomas longos, unicelulares na epiderme da face adaxial da lâmina foliar de A. scoparius e pela ausência destas estruturas em A. fissifolius.
