As a kind of novel hydrophobic drug carrier materials, the drug-loaded nanofibers were successfully fabricated using amphiphilic alginate derivative (AAD) and poly (vinyl alcohol) (PVA) by electrospinning method. The AAD was synthesized by amide linkage attachment of octylamine onto the carboxylic group of alginate, which was characterized by the means of elemental analysis, FT-IR spectrometer, 1 H NMR spectrometer and fluorescence measurement. Experimental results showed the amidation of alginate was successful, in which the degree of modification was 0.27 and the critical aggregation concentration in 0.15 mol/L aqueous NaCl solution was 0.43 mg/mL. The octyl groups of AAD could effectively make alginate chains flexible and enhance chain entanglements through the hydrophobic interactions between octyl groups. In contrast to sodium alginate (SA)/ PVA blend solutions, AAD/PVA blend solutions showed more excellent electrospinnability. Additionally, the different volume ratios of SA or AAD to PVA played an important role in the formation and morphology of the electrospun blend nanofibers. Although the amidation of alginate could not fundamentally alter the spinnability of alginate, the content of alginate in the blend nanofibers was appropriately increased. The new generated drug-loaded AAD/PVA (20/80) blend nanofibers exhibited relatively continuous and uniform nanofibers with the average diameter of 191.72 nm. Compared to the drug-loaded SA/PVA (20/80) blend nanofibers, the drug-loaded AAD/PVA (20/80) blend nanofibers presented a sustained release performance with the slow release of about 76 % k-cyhalothrin during the initial period of 10 h.

Glioblastoma is a common malignant tumor in the central nervous system with an extremely poor outcome; understanding the mechanisms of glioblastoma at the molecular level is essential for clinical treatment. In the present study, we used bioinformatics analysis to identify potential biomarkers associated with prognosis in glioblastoma and elucidate the underlying mechanisms. The result revealed that 552 common genes were differentially expressed between glioblastoma and normal tissues based on TCGA, GSE4290, and GSE 50161 datasets. Gene Oncology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment and protein-protein interaction (PPI) network were carried out to gain insight into the actions of differentially expressed genes (DEGs). As a result, 20 genes (CALB1, CDC20, CDCA8, CDK1, CEP55, DLGAP5, KIF20A, KIF4A, NDC80, PBK, RRM2, SYN1, SYP, SYT1, TPX2, TTK, VEGFA, BDNF, GNG3, and TOP2A) were found as hub genes via CytoHubba in Cytoscape and functioned mainly by participating in cell cycle and p53 signaling pathway; among them, RRM2 and CEP55 were considered to have relationship with the prognosis of glioblastoma, especially RRM2. High expression of RRM2 was consistent with shorter overall survival time. In conclusion, our study displayed the bioinformatic analysis methods in screening potential oncogenes in glioblastoma and underlying mechanisms. What is more is that we successfully identified RRM2 as a novel biomarker linked with prognosis, which might be expected to be a promising target for the therapy of glioblastoma.

Interpenetrating polymer network (IPN) is generally considered to be one of the most effective methods for imparting high-strength mechanical properties to hydrogel materials. To broaden the application of alginate hydrogel in skeletal muscle or muscle tissue engineering, the interpenetrating network technology and layer-by-layer (LBL) electrostatic assembly were proposed to fabricate the homogeneous alginate/polyacrylamide-chitosangelatin (Alg/PAAM-CS-GT) composite hydrogel scaffolds, using the hydroxyapatite/D-glucono-δ-lactone (HAP/GDL) complex as the gelling system. The effects of different components of alginate/polyacrylamide (Alg/PAAM) on the morphology, mechanical properties, swelling, degradability, and cytocompatibility of the hydrogel scaffolds were comparatively characterized. Experimental results showed that the as-prepared Alg/PAAM-CS-GT composite hydrogel scaffolds exhibited sharp edges and regular 3D structures. Their pore structure could be regulated by changing the content of PAAM in Alg/PAAM-CS-GT composite hydrogel scaffolds. And the filling of PAAM reduced the pores and thickened the pore walls, which obviously enhanced the mechanical properties of Alg/PAAM-CS-GT composite hydrogel scaffolds. FT-IR and thermogravimetric analysis showed that O‧‧‧N hydrogen bonding between COOH of SA and NH 2 of PAAM could be helpful to improve the thermal stability of the composite hydrogel scaffolds. The swelling and biodegradation measurements indicated that the swelling and degradation of Alg/PAAM-CS-GT composite hydrogel scaffolds could be effectively regulated by changing the component content of the composite hydrogel. In addition, the in vitro cytocompatibility analysis showed that the Alg/PAAM-CS-GT composite hydrogel scaffolds could support the growth of MC3T3-E1 cells and promote cell proliferation and differentiation. According to the in vitro cytocompatibility test results, the optimum PAAM content for the Alg/PAAM-CS-GT composite hydrogel scaffolds was 60%.
