Entre as espécies do gênero Passiflora, a P. foetida L. apresenta a maior variabilidade genética e tem grande importância medicinal, pois é usada no tratamento de doenças como asma, icterícia, e na forma de emplastros, para as erisipelas e doenças de pele com inflamação. Portanto, são necessários estudos que visem a micropropagação e conservação. As sementes de P. foetida L. apresentam dormência e muitas vezes, levam alguns meses para germinar, produzindo mudas desuniformes e de baixo vigor. Neste sentido, a cultura de tecidos apresenta-se como uma forma alternativa a propagação. Assim, o objetivo do trabalho foi estabelecer e micropropagar P. foetida L., para formação de um banco de germoplasma. Para tanto, sementes foram escarificadas, desinfestadas e inoculadas em meio MS(½) sem reguladores de crescimento e cultivados por 66 dias. Explantes de hipocótilos obtidos de plantas germinadas in vitro, foram cultivados no mesmo meio suplementado com 1,0 mg L-1 de BAP. Na fase de estabelecimento, 45% dos explantes brotaram e formaram gemas axilares. 88,9% dos explantes de hipocótilo induziram brotação e 11,1% produziram calos. Plântulas regeneradas com 1,82 cm de altura, com raízes foram aclimatadas.

RESUMO:A espécie Passiflora foetida L.é uma trepadeira exótica cujas folhas e frutos possuem propriedades medicinais. No Brasil, a erva é utilizada na forma de extratos ou emplastros para erisipelas e doenças de pele com inflamação. O objetivo deste trabalho foi caracterizar cinco acessos de Passiflora foetida L. baseados em 17 descritores morfológicos O experimento foi conduzido em casa de vegetação em delineamento inteiramente ao acaso, com 15 repetições por genótipo. As variáveis analisadas foram cor da casca do fruto, formato do fruto, comprimento do fruto, diâmetro do fruto, massa fresca do fruto, comprimento do pedicelo, cor da polpa do fruto, espessura da casca, acidez titulável, sólidos solúveis, potencial hidrogeniônico, razão sólidos solúveis/ acidez titulável, comprimento da semente, diâmetro da semente, espessura da semente, massa fresca das sementes e o número de sementes por fruto. Os dados foram submetidos à análise de variância e teste de médias pelo teste Tukey a 5% de probabilidade. Houve diferença significativa pelo teste F a 5% de probabilidade, entre os genótipos para quase todas as características com exceção da espessura da casca, massa fresca do fruto, comprimento da semente e massa fresca das sementes. A característica que apresentou maior variação foi o teor de sólidos solúveis. E entre os genótipos, o acesso 5 foi o que apresentou as maiores médias pelo teste de Tukey a 5% de probabilidade, em todas as características analisadas, podendo ser utilizado em programas de melhoramento da espécie. The species Passiflora foetida L. é an exotic vine, which leaves and fruit have medicinal properties. In Brazil, the herbis used in the form of extracts or poultices for erysipelas and skin diseases with inflammation. The aim of this study was to characterize five Passiflora foetida L. accessions based on 17 morphological descriptors The experiment was conducted in a greenhouse in a completely randomized design with 15 replicates per genotype. The variables were the fruit skin color, fruit shape, fruit length, fruit diameter, fresh fruit, length of pedicel, flesh color of the fruit, skin thickness, acidity, soluble solids, hydrogen potential ratio soluble solids / acidity, seed length, seed diameter, thickness of seed, fresh seed and seed number per fruit. The data were submitted to analysis of variance and the means were compared by Tukey test at 5% of probability. There was a significant difference by F test at 5%,of probability among the genotypes for nearly all traits, except the thickness of the shell, fresh fruit mass, seed length and fresh weight of seed. Intraspecies or interspecies crossings transfer relevant alleles between plants. However, some interspecies crossings involving Passifl ora species impede ovule fertilization and the viable development of seeds. Thus, the purpose of this study was to verify the viability of interspecifi c crossings and monitor pollen tube development.The experiment had six species of Passifl ora in the reciprocal crossings. Histochemical tests aimed to evaluate the percentage of intraspecies or interspecies crossings that resulted in fruit development and pollen tube development. Ovule fertilization and fruit development occurred in determined directions of crossings when controlling the female or male genitor, but only one case of reciprocal crossing had success. In crossings with no fruit development, histological analysis showed that some callus developed in the stigma and style, confi rming unilateral and interspecies incompatibility in the genus Passifl ora to some species and some directions of crossings.

Cacti have a highly specialized stem that enables survival during extended dry periods. Despite the ornamental value of cacti and the fact that stems represent the main source of explants in tissue culture, there are no studies on their morpho-anatomical and cytological characteristics in Melocactus . The present study seeks to address the occurrence of cells with mixed ploidy level in cacti tissues. Specifically, we aim to understand how Melocactus stem tissue is organized, how mixoploidy is distributed when present, and whether detected patterns of ploidy change after long periods of in vitro culture. To analyze tissue organization, Melocactus glaucescens and Melocactus paucispinus plants that had been germinated and cultivated in vitro were analyzed for stem structure using toluidine blue, Xylidine Ponceau, Periodic Acid Schiff, ruthenium red, and acid floroglucin. To investigate patterns of ploidy, apical, medial, and basal zones of the stem, as well as, periphery, cortex, and stele (vascular tissue and pith) regions of the stem and root apexes from four- and ten-year old cultured in vitro were analyzed by flow cytometry. X-ray micro-computed tomography (XRµCT) was performed with fragments of stems from both species. The scarcity of support elements ( i.e. , sclereids and fibers) indicates that epidermis, hypodermis, and wide-band tracheids present in cortical vascular bundles and stele, as well as water stored in aquifer parenchyma cells along the cortex, provide mechanical support to the stem. Parenchyma cells increase in volume with a four-fold increase in ploidy. M. glaucescens and M. paucispinus exhibit the same pattern of cell ploidy irrespective of topophysical region or age, but there is a marked difference in ploidy between the stem periphery (epidermis and hypodermis), cortex, stele, and roots. Mixoploidy in Melocactus is not related to the age of the culture, but is a developmental trait, whereby endocycles promote cell differentiation to accumulate valuable water.

To increase coumarin production in medicinal plants, this research evaluated the elicitor effect of bacterial products (Dipel ® and Xentari ® ) and biofertilizer (Vairo) on bioactive production when applied to Mikania laevigata. To estimate the induction of antifungal substances, aqueous extract of M. laevigata leaves was used against Fusarium oxysporium and Sclerotinia sclerotiorum. In the trials, clones were prepared from a matrix plant, where 22mL of treatments were sprayed on each plant, totaling 288 plants. There were 3 repetitions of 3 plants and 8 treatments. The leaves were dehydrated for 48h at 45ºC and quantification of coumarin was performed in High Performance Liquid Chromatography. In the fungal assays, CFUs and conidiogenesis were evaluated. Data revealed 85% germination in M. laevigata clones. The coumarin quantification curve resulted in the linear equation: y = 9E, 7x + 914140, and the coefficient of determination was R2 = 0.9893. The results show that the mixture of Vairo and Xentari ® act as coumarin elicitors in M. laevigata, being recommended to collect the leaves between 2 and 7 days. Plant protection against F. oxysporium was also observed by inhibiting conidiogenesis.
