A organização de diferentes genes de resistência da cultivar Ouro Negro de feijoeiro-comum (Phaseolus vulgaris) à ferrugem, antracnose e mancha-angular foi estudada com o auxílio de marcadores moleculares. Uma população de 154 linhas endogâmicas recombinantes (RIL's) obtidas do cruzamento entre as cultivares Ouro Negro e Rudá foram inoculadas com sete raças fisiológicas de Uromyces appendiculatus, três de Colletotrichum lindemuthianum, e quatro de Phaeoisariopsis griseola. Amostras de DNA de cada uma das RIL's foram amplificadas via PCR utilizando 70 diferentes primers. A análise da segregação da resistência à ferrugem, antracnose e mancha-angular na população de 154 RIL's revelou diferentes modos de herança para a resistência a cada uma das raças fisiológicas. A análise de ligação genética revelou que os diferentes genes de resistência à ferrugem e à antracnose estão no mesmo grupo de ligação. Os genes de resistência à mancha-angular também foram mapeados juntos, porém em outro grupo de ligação. Verificou-se neste trabalho que a utilidade dos marcadores RAPD, previamente identificados como ligados a genes de resistência do feijoeiro a doenças foi restrita. Apenas cinco dos 38 marcadores moleculares testados foram validados na população de RIL's como ligados aos genes de resistência à ferrugem e à antracnose. Três novos marcadores (OBA16(669) e OBA16(583) a 10,4 cM em acoplamento e OAD9(3210) a 13,9 cM em repulsão) ligados ao bloco gênico de resistência da cultivar Ouro Negro à mancha-angular foram identificados.
Foram inoculadas 17 cultivares de feijoeiro-comum (Phaseolus vulgaris) recomendadas para plantio em Minas Gerais com cinco patótipos (6, 7, 9, 10 e 12) de Uromyces appendiculatus e cinco (63.55, 31.23, 63.39, 31.55 e 63.23) de Phaeoisariopsis griseola. Foi conduzido um ensaio separadamente para cada um dos patótipos dos dois patógenos, totalizando 10 ensaios. Nos ensaios de ferrugem foi avaliada a freqüência de infecção (FI) e estimado o tamanho médio das pústulas (TMP) e, nos ensaios de mancha-angular foram avaliados os sintomas utilizando uma escala de notas variando de 1 a 9. A maioria das cultivares se mostrou suscetível a pelo menos três dos cinco patótipos de ferrugem. As cultivares mais suscetíveis foram Roxo 90 e Meia Noite e a mais resistente foi a Ouro Negro, a qual apresentou proteção completa aos cinco patótipos testados. Com relação à mancha angular, além do Roxo 90, as cultivares Carioca MG, Pérola, Carioca 1030 e Aporé, extensivamente plantadas em Minas Gerais, foram altamente suscetíveis. As cultivares Ouro Negro e EMGOPA 201-Ouro apresentaram genes de resistência a diferentes patótipos e a combinação desses genes em uma nova cultivar resultaria em resistência a todos os patótipos testados.
A identificação de fontes de resistência é uma etapa básica em programas de melhoramento que visam o desenvolvimento de cultivares resistentes a doenças. Neste trabalho, foram inoculadas, sob condições controladas, oito linhagens de feijoeiro-comum (Phaseolus vulgaris) desenvolvidas pelo Departamento de Agricultura dos Estados Unidos, com quatro raças conhecidas de Uromyces appendiculatus coletadas no estado de Minas Gerais, Brasil. A cultivar Ouro Negro foi utilizada como padrão de resistência e US Pinto 111 (suscetível universal) como padrão de suscetibilidade. Foi avaliada a freqüência de infecção (FI) e estimado o tamanho médio das pústulas (TMP). As linhagens BelMiDak-RMR-12 e BelMiDak-RR-3 destacaram-se como as mais resistentes, apresentando, para todas as raças, apenas pústulas (com diâmetro < 300mim) na face abaxial da folha. As linhagens BelDakMi-RMR-10 e BelNeb RR-2 apresentaram baixa FI média e foram resistentes a quatro e três raças, respectivamente. A linhagem BelDak-RR-2, apesar de sua resistência a todas as raças, apresentou uma FI média relativamente alta. As linhagens BelMiNeb-RMR-3 e BelDakMi RR-7 também foram resistentes às quatro raças testadas, porém apresentaram alta FI média e algumas pústulas com diâmetro maior que 300 mim. A linhagem BelDakMi-RMR-11 foi resistente às raças 45, 46 e 49 e moderadamente resistente à raça 52. A cultivar Ouro Negro foi resistente às quatro raças, com um nível de resistência igual às melhores linhagens norte americanas.
Microsatellites were developed for Stylosanthes macrocephala, aiming at developing tools for studying the genetic diversity of this species. A total of 13 polymorphic microsatellite markers were isolated from a S. macrocephala enriched genomic library. The isolated microsatellites were characterized in 20 accessions of the S. macrocephala germplasm collection belonging to the Empresa Brasileira de Pesquisa Agropecuária (Embrapa) Cerrados. The number of alleles per locus varied from 2 to 11, with an average of 4 alleles per locus. The observed and expected heterozygosities ranged from 0 to 0.25 and 0.05 to 0.90, respectively. Cross-amplification of the S. macrocephala polymorphic microsatellites was evaluated in three other Stylosanthes species. The microsatellites reported herein are the first set of microsatellite markers developed for S. macrocephala and are potentially useful for further studies on genetic diversity, conservation and breeding of this species.
Microsatellite (SSR) markers were developed for the following tropical forage species, using accessions available from the plant genetic resources (PGR) collections held by EMBRAPA (Brazilian Agricultural Research Corporation): Brachiaria brizantha, B. humidicola, Panicum maximum, Paspalum spp., Stylosanthes capitata, S. guianensis, S. macrocephala, Calopogonium mucunoides and Centrosema spp. The markers were used to analyze population structure and genetic diversity, evolution and origin of the genetic variability in the center of origin, mating systems and genetic resources in EMBRAPA's germplasm bank. The results shed light on the amount of genetic variation within and between populations, revealed the need in some cases for further plant collection to adequately represent the species in PGR collections, allowed us to assemble core collections (subsets of the total collections) that should contain most of the available diversity and (in the case of the legumes) showed the need to avoid unwanted outcrossing when regenerating conserved material. The data will allow plant breeders to better select accessions for hybrid production, discriminate between genotypes and use marker-assisted selection in breeding programs. Our results will also underpin the construction of genetic maps, mapping of genes of agronomic interest and numerous other studies on genetic variability, population structure, gene flow and reproductive systems for the tropical forage species studied in this work. ResumenSe desarrollaron marcadores microsatélite (SSR) para las siguientes especies forrajeras tropicales, usando accesiones de germoplasma disponibles en las colecciones de recursos genéticos mantenidas por EMBRAPA (Empresa Brasileira capitata, S. guianensis, S. macrocephala, Calopogonium mucunoides y Centrosema spp. Los marcadores se usaron para analizar la estructura de las poblaciones y su diversidad genética; la evolución y el origen de la variabilidad gené-tica en los respectivos centros de origen; los sistemas reproductivos; y los recursos genéticos en el banco de germoplasma de EMBRAPA. Los resultados arrojaron luz sobre la magnitud de la variación genética dentro de y entre poblaciones; revelaron la necesidad, para algunas especies, de incrementar las colecciones de germoplasma y así obtener una adecuada representación de las especies en las colecciones de recursos genéticos; permitieron establecer colecciones núcleo (subconjuntos de colecciones grandes), representativas de la diversidad genética disponible en las respectivas colecciones completas; y mostró, para las leguminosas, el riesgo de cruzamientos no deseados al multiplicar semilla de las accesiones conservadas. La información obtenida facilitará a los fitomejoradores la selección de accesiones para la generación de híbridos, la distinción entre genotipos, y el uso de la selección asistida por marcadores en programas de fitomejoramiento.
