The endophytic bacteria belong to a larger group of microorganisms that have their life-cycle partly or entirely inside the plant and are located in intra and inter-cellular spaces or in the vascular tissue. These bacteria can be found colonizing aerial parts or roots. This review aims to analyze the colonization strategies of endophytic bacteria through interaction with plants, as well as to highlight the metabolic influence of these organisms in plant tissues, which result in physiological and biochemical changes. Depending on the different mechanisms used internally to colonize a plant, these microorganisms are called obligate, facultative, or passive endophytes. Phytostimulation, biofertilization and biological control are mechanisms that result in the development of the plant through the production of plant hormones, bioavailability of nutrients and antagonistic action to phytopathogens, respectively. The association between endophytic bacteria and plants features important benefits such as significant increases in growth, plant biomass, length of roots, dry matter production, and grain yield. Studies show that there is a great diversity of endophytic bacteria colonizing plant structures that result in several benefits to the host plant.
Os nematóides são encontrados em quase todos os ambientes do mundo; aproximadamente 10% são parasitas de plantas podendo causar danos em todas as partes vegetais, sendo as raízes seu alvo principal. Os prejuízos financeiros decorrentes da atividade destes parasitas nos diferentes cultivos foram estimados em cerca de R$ 400 milhões anuais. São conhecidas mais de 90 espécies do gênero Meloidogyne; no entanto, as que causam danos econômicos importantes são: M. incognita, M. javanica, M. arenaria e M. hapla. As bactérias com capacidade de causar doenças aos fitonematóides possuem diferentes modos de ação e podem ser encontradas no solo, nos tecidos das plantas hospedeiras e nos próprios nematóides. As principais bactérias estudadas para controle biológico dessa praga são aquelas da rizosfera com capacidade de invadir os tecidos internos das plantas, ou seja, endofíticas facultativas, como Bacillus spp. e Pseudomonas spp., além das bactérias parasitas obrigatórias de nematóides, como o gênero Pausteria. Essa revisão destaca as principais bactérias com potencial de controle de fitonematóides, como B. subtilis, B. firmus e B. thuringiensis as quais têm demonstrado grande eficiência no controle destes parasitas. Os estudos também destacam o potencial de bactérias do gênero Pseudomonas com efeito nematicida principalmente contra os nematóides formadores de galhas (Meloidogyne spp.). Dentre as bactérias parasitas, Pausteria penetrans possui grande potencial de aplicação no controle biológico de fitonematóides, especialmente quando utilizada em conjunto com práticas culturais. Currently the major challenge of humanity is focused on population growth/ increase through agricultural production in order to meet the demand for food. Overtime, different pests have emerged, with some being of great importance. Among these pests, the nematodes are noted for attacking leguminous plants, grasses, citrus, and other fruits. The main pest species are of the genus Heterodera, Meloidogyne, Pratylenchus, and Globodera. These nematodes cause losses up to 100 %, preventing agriculture of certain areas. Financially, about $100 billion annual damage is caused by nematodes. These amount to 90 % of the yield of cotton, yams, beans, and soybeans, and in citrus damage is estimated at 14 % of production. Alternative methods of control are being studied, and in this context, a bacterium of the genus Bacillus has prominence and importance. Besides Bacillus subtilis, some by pesticides are already marketed for the control of nematodes, such as Bioarc ® the basis of Bacillus megaterium, Bio Zeid ® the basis of Trichoderma album, and also using the brown alga, Ascophyllum nodosum (Algaefol ® ), among others. The objective of this chapter is to report the use of different Bacillus species and some biopesticides used to control nematodes of agricultural importance.
Oryzophagus oryzae (Costa Lima 1936) (Coleoptera: Curculionidae)
The aim of this investigation was the assessment of toxicity of two new isolates of Bacillus thuringiensis, and the aqueous extract of Melia azedarach through in vivo assays in CF1 mice. Bt 1958-2, Bt 2014-2 and the BTh Thuricide 63 standard isolates were grown in liquid usual glicosed medium, and Cry proteins were purified by centrifugation on a sucrose gradient. The supernatant was autoclaved at 121º C, 15min. to maintain the exotoxins. Dehydrated leaves of M. azedarach were used to prepare a 10% aqueous extract. Mice were treated either orally or intraperitoneally with a whole bacterial suspension (1.10 10 UFC/mL), a culture supernatant or purified crystal protein (50 µg/mL), and with the plant extract (50 µg/mL). The stomachs of the mice were collected and observed in stereomicroscopy, and the stomach contents were analyzed in 10% SDS-PAGE. Results showed that none of the oral treatments were toxic to mice, but intraperitoneal bacterial suspensions were lethal to the animals 6 -24 hours after injection. In conclusion, the Cry proteins of the new B. thuringiensis isolates must be evaluated for their use as tools in the biotechnology field, since they do not show toxicity against mammals, intragastrically or peritoneally, just like the M. azedarach aqueous extract (10%), with those being indicated for the biological control of pest insects.KEY WORDS: Bacterium, mammalian, Meliaceae, toxicology. RESUMO TOXICIDADE VIA INTRAGÁSTRICA E INTRAPERITONEAL DE BACILLUS THURINGIENSIS E MELIA AZEDARACH, EM CAMUNDONGOS. O objetivo deste estudo foi a avaliação da toxicidade de dois novos isolados deBacillus thuringiensis e o extrato aquoso de Melia azedarach, através de ensaios in vivo em camundongos CF1. Os isolados Bt 1958-2, Bt 2014-2 e o isolado padrão BTH Thuricide 63 foram cultivados em meio usual glicosado, e as proteínas Cry foram purificadas por centrifugação em gradiente de sacarose. O sobrenadante foi tratado em autoclave a 121º C, 15 min para manter as exotoxinas. As folhas desidratadas de M. azedarach foram utilizadas para preparar um extrato aquoso a 10%. Camundongos foram tratados, via oral ou por via intraperitoneal, com a suspensão bacteriana (1.10 10 UFC/mL), o sobrenadante de cultura ou a proteína do cristal purificada (50 µg/mL), e com o extrato da planta (50 µg/mL). Os estômagos dos ratos foram coletados e observados em estereomicroscópio e os conteúdos estomacais foram analisados em SDS-PAGE a 10%. Os resultados mostraram que nenhum dos tratamentos orais foram tóxicos para os camundongos, mas, via intraperitoneal, as suspensões bacterianas foram letais para os animais entre 6 e 24 horas após a injeção. Em conclusão, as proteínas Cry dos novos isolados de B. thuringiensis devem ser avaliadas para sua utilização como ferramenta no campo da biotecnologia, uma vez que elas não mostram toxicidade contra mamíferos, intragástrica ou intraperitoneal, assim como o extrato aquoso (10%) de M. azedarach, podendo ser indicado para o controle biológico de insetos-praga.
Bacilos entomopatogênicos são bactérias observadas principalmente no solo, consideradas importantes agentes no controle biológico de doenças e pragas da agricultura. No entanto, existem poucos estudos que relatam a influência das práticas agrícolas nas populações de Bacillus spp. Essa pesquisa objetivou avaliar a freqüência de bactérias entomopatogênicas pertencentes ao gênero Bacillus obtidas de amostras de solo coletadas no ano agrícola de 2006/2007 de diferentes sistemas de cultivo de arroz irrigado da EEA-IRGA, Cachoeirinha (RS). Foram analisadas 36 amostras de solos, oriundas de três sistemas de plantio de arroz irrigado, pré-germinado (SPG), convencional (SPC), plantio direto (SPD) e de uma área sem plantio de arroz (C). Para tanto, um grama de solo foi diluído, pasteurizado e inoculado em Ágar Nutriente (AN), incubado a 30 °C, por 24 horas. Todas as unidades formadoras de colônias bacterianas (UFC) foram repicadas em meios seletivos e as espécies, B. thuringiensis, B. sphaericus e B. cereus, foram identificadas por microscopia de contraste de fase. Os resultados revelaram que não houve diferença significativa entre os sistemas de plantio analisados. Por outro lado, a frequência de Bacillus spp. variou entre as diferentes fases do plantio de arroz nos períodos amostrados. A irrigação foi o principal fator que contribuiu para a variação da frequência dos Bacillus em sistemas de plantio de arroz irrigado da EEA-IRGA.
