RESUMO -O pinhão manso (Jatropha curcas L.) apresenta elevado potencial para produção de biocombustível; porém, existem poucos estudos relacionados à avaliação da qualidade de suas sementes. Assim, a pesquisa objetivou estudar os procedimentos para a condução do teste de tetrazólio e verificar a eficiência dos testes de tetrazólio e de raios X para a avaliação da viabilidade de sementes desta espécie. Para o teste de tetrazólio, foi comparado o pré-condicionamento entre papel e imersão em água, até que as sementes atingissem os teores de água de 30% e 40%. Após esta etapa, o tegumento das sementes foi retirado manualmente e os embriões foram imersos em solução de 2,3,5 trifenil cloreto de tetrazólio nas concentrações de 0,075% e 0,5%, por 120 minutos, a 40°C. Os resultados do teste de germinação foram confrontados com os resultados do teste de tetrazólio. A hidratação entre papel, até as sementes atingirem 30% de água e a coloração do embrião em solução 0,5% de tetrazólio, durante 120 minutos, no escuro, a 40ºC, produzem informações consistentes. As sementes de pinhão manso foram submetidas ao teste de raios X, na intensidade de 25 Kv durante 60s; posteriormente, foi realizado o teste de germinação, relacionando-se os resultados obtidos nesses dois testes. Concluiu-se que é possível utilizar o teste de tetrazólio e de raios X para avaliar a viabilidade das sementes de pinhão manso.Termos para indexação: análise de imagens, potencial fisiológico, oleaginosas. EVALUATION OF THE Jatropha curcas L. SEED VIABILITY BY TETRAZOLIUM AND X-RAY TESTSABSTRACT-The Jatropha curcas L. presents a great potential for fuel, but there are few studies to assess seed quality of this species. Therefore, this research aimed to study the performance of the tetrazolium salt test procedures and to verify the efficiency of the X-ray test to evaluate the seed viability of this species. Seeds were hydrated between paper towel and immersed in water to reach 30 and 40% m.c.. Following, seed coat was removed manually and seeds were immersed in 0,075 and 0,5% tetrazolium solutions for 120 min., respectively, at 40°C. Best results of tetrazolium test comprised the hydration between paper, until the seeds reach 30% m.c. and coloration in contact with a solution of 0,5%. The Jatropha curcas L. seeds were also submitted to X-ray test, in intensity of the 25Kv for 60s. Results of tetrazolium and X-ray tests were compared to germination and it was shown that both are efficient to assess Jatropha curcas L. seeds viability.
The X-ray image analysis technique is a precise method that enables examination of regions that are damaged and/or altered in seed and their location and extension. It is a non-destructive method allowing the X-ray treated seed to be submitted to quality physiological tests. The objective of this study was to use X-ray image analysis technique to identify mechanical and stink bug damages in soybean seed. Seeds from different batches of the BRS184 cultivar were sorted by X-ray test and then were tested for germination with the aim to relate the damage with possible seed viability loss. Alternatively, the tetrazolium test was carried out to be compared with the X-ray test. Damage severity and location were considered for the interpretive analysis of X-ray images. The results showed that the X-ray image analysis technique was effective in detecting mechanical and stink bug damage in soybean seed.
RESUMO -Condições climáticas desfavoráveis após a maturidade fisiológica em sementes de soja têm ocasionado problemas no seu potencial fisiológico, incluindo a ocorrência de danos por "umidade". Para a avaliação desses danos, a técnica de análise de imagens de raios X é aplicável para identificar, de maneira precisa, sua intensidade e localização. Assim, foi o objetivo desdte trabalho avaliar o efeito de diferentes condições de armazenamento na evolução dos danos por "umidade" em sementes de soja utilizando o teste de raios X e testes de potencial fisiológico. Foram utilizados três lotes do cultivar TMG113-RR, armazenados durante 8 meses em ambiente não controlado, câmara seca (50% UR e 20 o C) e câmara fria (65% UR e 10 o C). Periodicamente, a cada 2 meses, as sementes foram avaliadas por meio dos testes de germinação, envelhecimento acelerado, emergência de plântulas em campo e raios X. O delineamento experimental foi inteiramente casualizado em esquema fatorial 3x3x5 (lotes x ambientes x épocas) e as médias foram comparadas pelo teste de Tukey, a 5% de probabilidade. Observou-se que houve evolução dos danos por "umidade" e, conseqüentemente, diminuição do potencial fisiológico durante o período de armazenamento das sementes. A evolução do referido dano foi maior nas sementes armazenadas em ambiente não controlado e menor para as armazenadas em câmara fria.Termos para indexação: qualidade de sementes, Glycine max (L.) EVALUATION OF THE EVOLUTION OF "WEATHERING" DAMAGE AND REDUCTION IN VIGOR OF SOYBEAN SEEDS, TMG 113-RR CULTIVAR, DURING STORAGE, USING X-RAY IMAGES AND PHYSIOLOGICAL POTENTIAL TESTSABSTRACT -Adverse weather conditions after the physiological maturity of soybean seeds have caused problems in their physiological potential, including "weathering" damage. Through the image analysis technique (X-ray), the intensity and location of the damage can be identified. The objective of this study was to evaluate the effect of different storage conditions on the evolution of humidity damage in soybean seeds using the X-ray and physiological potential tests. Three batches of TMG113-RR cultivar were used, stored for eight months under three conditions: uncontrolled environment, dry chamber (50% RH and 20 o C) and cold chamber (90% RH and 10 o C). Every 2 months, the seeds were evaluated using the tests for germination, accelerated ageing, seedling emergence in the field and X-rays. The experimental design was completely randomized in a 3x3x5 factorial (batches x environment x storage) and the means were compared by the Tukey test at the 5% probability
RESUMO -Entre os problemas envolvidos na produção de sementes de soja destacam-se os danos por umidade, que resultam em perdas de germinação e de vigor. A técnica de análise de imagens, por meio do teste de raios X, é um método de precisão que possibilita examinar, com detalhes, a região danificada ou alterada, sua localização e extensão. Por ser um método não destrutivo, as sementes em análise podem ser submetidas a testes fisiológicos e, desta forma, é possível estabelecer as relações de causa e efeito. O presente trabalho teve como objetivo avaliar a eficiência da técnica de análise de imagens, por meio do teste de raios X, na identificação dos danos por umidade em sementes de soja, comparativamente ao teste de tetrazólio. Sementes de diferentes lotes de um mesmo cultivar (BRS 184) foram submetidas ao teste de raios X e, posteriormente, destinadas ao teste de primeira contagem de germinação, de forma a relacionar os danos com os possíveis prejuízos proporcionados às sementes. Paralelamente, foi realizado o teste de tetrazólio visando comparação com o teste de raios X. Para a análise interpretativa do teste de raios X, foram consideradas a severidade e a localização dos danos, juntamente com as fotografias digitais das plântulas ou sementes mortas resultantes do teste de primeira contagem de germinação. This precise method examines seeds individually using enlarged images in which damaged areas, as well their exact location and extension, can be found and examined in detail. The goal of this research was to study the efficiency of the image analysis technique, by the X-ray test, to identify weathering damage in soybean seeds, compared with the tetrazolium test. Seeds of different lots of soybean, cultivar BRS 184, were submitted to the X-ray test and, consecutively, used for the first count of the germination test, in order to determine the possible relationship between cause and effect. Simultaneously, the X-ray test was compared with the tetrazolium test. For the interpretation of X-ray test, the severity and location of weathering damage was considered. The results showed that image analysis, by the X-ray test, was efficient in detecting weathering damage in soybean seeds.
RESUMO-Em espécies multiplicadas comercialmente via semeadura indireta, como várias essências florestais, a utilização de sementes de alto potencial fisiológico é imprescindível para garantir o desenvolvimento e estabelecimento adequado das plantas no campo. Assim, diversas técnicas têm sido utilizadas para avaliar a integridade das sementes e, dentre elas, o teste de raios X. O objetivo no trabalho foi avaliar o potencial fisiológico das sementes de ipê roxo (Tabebuia heptaphylla), após separação por cor e tamanho, por meio dos testes de raios X e de germinação. As sementes testemunha (sem classificação). Em seguida, as sementes das diferentes frações, foram submetidas a avaliações da integridade física e viabilidade, por meio de teste de raios X e de germinação, respectivamente. Adotou-se delineamento experimental inteiramente casualizado, com seis tratamentos e cinco repetições, comparando-se as médias pelo teste de Scott-Knott (p ≤ 0,05). O teste de raios X é eficiente na avaliação da morfologia interna de sementes de ipê-roxo e sua relação com o potencial fisiológico. A coloração e o tamanho das sementes de ipê-roxo influenciam o potencial fisiológico das mesmas. Several techniques have been used to evaluate seed quality including the X-ray test. The objective of this study was to evaluate the physiological potential of Tabebuia heptaphylla seeds with X-ray and germination tests after grading by color and size. Seeds were classified using the Munsell color chart and screens, in light-yellow larger, light-yellow smaller, dark-yellow larger, dark-yellow smaller, mixture of light-yellow larger, light-yellow smaller and dark-yellow larger and control. , maintained in vitro for two time periods, namely P1, 6 weeks and P2, twelve weeks, and were statistically evaluated after each interval with respect to: height, fresh weight, number and length of roots, number of leaves, and pseudostem diameter, in addition to the rate of acclimatization in percentage terms. Treatment B100 presented the lowest level of development in the two periods, and 100 % survival in acclimatization.
