Due to the strong bitter taste, sacha inchi seeds are usually consumed after roasting, which also contributes to the elimination of antinutrients. Sacha inchi plants fully adapted to cultivation under sub-tropical climate conditions were produced in southeastern Brazil. Our main goal was to evaluate the effect of dry heating (roasting) on the antinutrient content of these seeds. We also investigated the effects of the applied roasting treatments on the antioxidant activity, proximate composition and oxidative stability of the seeds. To the best of our knowledge, this is the first report on antinutrients of sacha inchi seeds cultivated under sub-tropical conditions, outside their native tropical environment. Except for saponins, which are not heat-labile compounds, the contents of all assessed antinutrients continually reduced with the increase in roasting temperature. Roasting improved antioxidant activity and phenolic content in the seeds at the highest temperature. Oxidation changes occurred in the seed oil, and they increased with temperature. However, maximum peroxide value was within the acceptable consumption limits. As a conclusion, roasting treatments can be applied to minimize the antinutrient potential in sacha inchi seeds. Knowledge on the composition and proper processing of sacha inchi cultivated under sub-tropical conditions may support future efforts focused on the development of new production areas.

RESUMOA literatura tem relatado a ocorrência de variação somaclonal em plantas derivadas de cultura de tecidos. Neste trabalho, foi estudada a influência do número de subcultivos na indução de variantes em bananeira, cultivar Pacovan (Musa spp., grupo AAB). Ápices caulinares foram introduzidos e multiplicados in vitro, utilizando-se no estabelecimento o meio de cultura MS com adição de 2,5 mg.L -1 de BAP e nos subcultivos subseqüentes, o meio MS com 4,0 mg.L -1 de BAP para indução de brotações laterais. Foram obtidas gerações com diferentes números de subcultivos (3, 4, 5, 6, 7, 8 e 9), e comparadas com mudas obtidas de rizomas. Observaram-se as plantas obtidas, no total de 426, em condições de campo no Vale do Açu (RN). Para cálculo da porcentagem de variantes somaclonais, avaliaram-se as seguintes caracterís-ticas morfológicas: estatura da planta, coloração e forma das folhas e conformação do cacho. Foram consideradas como variantes apenas as plantas cujos perfilhos também mostravam o mesmo tipo de variação. Constatou-se a ocorrência de variação somaclonal nas mudas provenientes dos tratamentos a partir de cinco subcultivos, atingindo-se o valor de 5,8% de variantes no nono subcultivo. O aumento da porcentagem de variação somaclonal com o do número de subcultivos indica o cuidado que as biofábricas devem ter em relação a esse parâmetro, desenvolvendo protocolos de micropropagação específicos para cada cultivar a ser comercializada.Palavras-chave: produção de mudas, Musa spp, in vitro, variação somaclonal. ABSTRACT SOMACLONAL VARIATION EVENT ON MICROPROPAGATED PACOVANBANANA SEEDLING (Musa spp. AAB group)The occurrence of somaclonal variation for plants in developed from tissue culture is well documented in the literature. The present study evaluated the influence of numbers subculture in the induction of variants in Pacovan banana tree (Musa spp., AAB group). Apex stems were introduced and multiplied in vitro using culture media MS, with addition of 2.5 mg.L -1 of BAP. In subsequent subcultures, MS with 4.0 mg.L -1 of BAP was used to induce side buds. Results showed that plants were regenerated with different numbers of subculture (3, 4, 5, 6, 7, 8, and 9). These data were compared with seedlings from developed rhizomes. All the plants, were observed at field conditions in Vale do Açu, Rio Grande do Norte State. The great interest for heliconias by the external market has placed this crop in a prominent position in the flower production chain. The small offer of healthy seedlings, however, the expansion of this activity. The presence of endophytic microorganisms such as Pseudomonas sp. represents an obstacle even for the evolution of heliconia tissue culture studies. Stem apices of Heliconia rauliniana were inoculated and analyzed in vitro to investigate the presence of endophytic microorganisms. An antibiotic analysis was also run for posterior microbiological control. The presence of Pseudomonas sp. and Klebsiella sp. in stem apices confirmed infection. Antibiotics such as chloramphenicol, cefotaxime and the association of both (chloramphenicol + cefotaxime) at rates of 50, 150, 250 and 500 mg L -1 , were applied to the MS medium and supplemented with 3.5 mg L -1 of 6-BA. After a 50 day-period, 500 mg L -1 of cefotaxime was the most efficient form to control endophytic microorganisms in this species of heliconia. The chloramphenicol, although effective for endophytic microorganism control, was inadequate for in vitro growth due to its cytotoxicity to explants. Key words: tissue culture, microbiologic control, endophytics ESTABELECIMENTO IN VITRO DE Heliconia rauliniana (HELICONIACEAE)RESUMO: A acentuada procura por helicônias, principalmente por parte do mercado externo tem colocado o cultivo desse gênero de plantas em posição de destaque, dentre as atividades desenvolvidas no ramo da floricultura. A pouca oferta de mudas e a inexistência de mudas sadias prejudicam consideravelmente a expansão dessa cultura. A presença de microrganismos endofíticos, como a Pseudomonas sp., representa um empecilho no desenvolvimento de trabalhos de cultura de tecidos em helicônias. Ápices caulinares de Heliconia rauliniana foram inoculados in vitro e avaliados quanto aos tipos de microrganismos endofíticos e análise de antibiograma para posterior controle microbiológico. A presença de Pseudomonas sp. e Klebsiella sp. nos ápices caulinares estudados, comprova a presença dos endofíticos. Os antimicrobianos cloranfenicol, cefotaxima e a forma associada cloranfenicol + cefotaxima em doses 50, 150, 250 e 500 mg L -1 , foram aplicados ao meio MS, suplementado com 3,5 mg L -1 de 6-BA, como forma de controle. Após 50 dias de avaliação, a cefotaxima aplicada isoladamente, na dose 500 mg L -1 , foi a forma mais eficiente de controle dos endofíticos para esta espécie de helicônia. O cloranfenicol, apesar do controle efetivo dos endofíticos, mostrou-se inadequado ao cultivo in vitro, sendo cito-tóxico aos explantes. RESUMO O objetivo do presente trabalho foi avaliar, em condições de campo, características horticulturais de mudas propagadas in vitro de Sacha inchi (Plukenetia volubilisLConhecida pelos povos andinos a milhares de anos, a euforbiácea Plukenetia volubilis Linneo é nativa da região Amazônica, compreendendo os países do Peru, Colômbia, Venezuela e Brasil. Essa trepadeira perene se adapta a solos ácidos e argilosos, altitude de 0 a 1200m e tem melhor desenvolvimento em clima quente. As fl ores são unissexuadas e dispostas em infl orescências racemosas. Trata-se de uma planta monoica e, apesar de ser possível a autofecundação, há uma grande porcentagem de fecundação cruzada. A semente oval e de coloração marrom escura é a estrutura de maior interesse na planta. Por ser rica em proteínas, vitamina E, e principalmente em óleos poli-insaturados ômegas 3, 6 e 9, em maior concentração até que em outras oleaginosas, ela apresenta um grande potencial agro tecnológico e atualmente é largamente explorada por indústrias peruanas na produção de seu azeite. To evaluate the behavior of micropropagated heliconia seedlings during the acclimatization process using different substrates and shade levels, seedlings of H. bihai (Lobster Claw I) were planted in plastic trays using the substrates: washed sand, vermiculite (medium texture), and PlantMax ® Horticultura, and shade cloths with percentages of 0% (full sun), 30%, 40%, 50%, 60%, 70%, and 80% of shade in relation to full sun. Best results were obtained for washed sand and PlantMax substrates at shade conditions starting from 50%, especially at 70% and 80%. Pre-existing roots from in vitro rootings were not functional, not presenting secondary roots, and showing root tissue necrosis. Acclimatized seedlings developed new roots. This finding suggests that this plant can be acclimatized without going through a laboratory rooting stage. Key words: Heliconia, ex vitro culture, rooting ACLIMATIZAÇÃO DE MUDAS MICROPROPAGADAS DE Heliconia bihai (HELICONIACEAE)RESUMO: A cultura da helicônia é a que apresenta maior crescimento entre o cultivo de flores tropicais para a exportação. Avaliou-se o comportamento de mudas de helicônias micropropagadas no processo de aclimatização, em diferentes substratos e níveis de sombreamento. Foram plantadas mudas de H. bihai (Lobster Claw I) em bandejas plásticas contendo os diferentes substratos: areia lavada, vermiculita (textura média) e PlantMax ® Horticultura. Na avaliação de níveis de sombreamento foi utilizado o telado (sombrite), com ordem de sombreamento 0% (pleno sol), 30%, 40%, 50%, 60%, 70% e 80%. O desempenho foi superior nos substratos areia lavada e PlantMax ® Horticultura e nas condições de sombreamento a partir de 50 %, em especial 70% e 80%. As raízes pré-existentes, oriundas do enraizamento in vitro, mostraram-se não funcionais, sem a emissão de raízes secundárias e apresentando necrose do tecido radicular. As mudas aclimatizadas desenvolveram novas raízes, o que sugere que esta cultura pode ser aclimatizada sem que as plântulas passem pela fase de enraizamento em laboratório.
