Different extraction procedures were employed to characterise the polyphosphate granules in autotrophic planktonic organisms, the green microalgae Chlorella vulgaris and the cyanobacterium Synechocystis sp. strain PPC 6803. The effectiveness of these methods was assessed using epifluorescence microscopic analysis of DAPI stained specimens as well as by electron spectroscopic imaging. The results clearly indicate that NaOH and hot water treatment followed by filtration of the extracts are suitable to obtain a cell free suspension of intact polyphosphate granules without hydrolysing the polymers. The methods described are useful to gain physiological information on the phosphorus status of autotrophic planktonic organisms.

Various freshwater, estuarine and coastal stations of the Southern Baltic Sea were comparatively studied to evaluate pelagic bacterial performance. Inner coastal waters (so-called bodden or lagoons) are highly productive systems and dominate the coast of the Southern Baltic Sea. Due to high nutrient loads up to the 1990s in combination with an enhanced primary production, increased amounts of particulate (POC) and dissolved organic carbon (DOC) accumulated in these waters. In the Darß-Zingst bodden chain, POC < 16 and DOC < 13 mg C l -1 , and C:N ratios of 9 to 11 in particulate matter were measured in winter and spring samples. Due to high POC concentrations, the average ratio of DOC:POC was 1.1:1, which is very low compared to other aquatic systems. Bacterial abundance and activities were rather high and reached 24 × 10 6 ml -1 and 18 µg C l -1 h -1 , respectively. Although 2 of the 3 investigated freshwater systems were classified as eutrophic, the highest measured POC concentrations, bacterial abundance and production were much lower (1.6 mg C l -1 , 11 × 10 6 bacteria ml ) than in the bodden. In contrast to that, the DOC load was as high as in these inner coastal waters (<12 mg C l -1 ). The coastal stations of the Baltic Sea, classified as mesotrophic, were not severely loaded with organic matter and bacteria (POC < 0.8, DOC < 5.5 mg C l -1 , bacteria < 3 × 10 6 ml -1). Bacterial production again was lower than at all other stations; however, levels did reach an exceptional 4.6 µg C l -1 h -1, which is comparable to values of the freshwater systems. Compared to the other investigated marine and freshwater systems, the bodden were heavily loaded with organic matter, especially particulate organic matter (POM). The origin of this material is assumed to be mainly autochthonous as it is known not to be transported by rivers into these estuaries. Although dissolved inorganic nitrogen (DIN) concentrations were high at least in winter, POM was of poor quality; this was reflected by high C:N ratios and a low contribution of phytoplankton carbon to POC. However, this is particularly surprising, because nitrogen should be readily available at all bodden sites by resuspension from the sediment caused by frequent winds in these very shallow systems of < 2 m depth. Paleolimnological techniques were used to identify environmental changes in and around Lake Dudinghausen (northern Germany) over the past 4800 yr. Diatom-inferred total phosphorus (DI-TP) changes identify four phases of high nutrient levels (2600–2200 BC, 1050–700 BC, 500 BC–AD 100 and AD 1850–1970). During these high DI-TP phases, fossil pollen, sediment geochemistry and archaeological records indicate human activities in the lake catchment. Although the same paleo-indicators suggest increased human settlement and agriculture activity during the late Slavonic Age, the Medieval Time and the Modern Time (AD 1000–1850), DI-TP levels were low during this period. In the sediments, iron and total phosphorus were high from ∼AD 100 to 1850, likely due to increased inflow of iron-rich groundwater into the lake. Increased iron input would have lead to a simultaneous binding and precipitation of phosphate in the upper sediment and overlying water column. As a result, anthropogenic impact on Lake Dudinghausen was masked by these phosphorus-controlling processes from AD 1000 to 1850 and was not evident by means of DI-TP. In accordance with fossil pollen, sediment geochemistry and limited archaeological records, DI-TP levels were low from AD 100–1000. Groundwater levels likely rose during this period as the climate gradually changed toward colder and/or moister conditions. Such climate change likely led to reduced settlement activities and forest regeneration in the catchment area. Our results are concordant with similar studies from central Europe which indicate rapid decreasing settlement activities from AD 100 to 1000.

The cyanobacterial diversity in the pelagic of a shallow estuary at the Southern Baltic Sea has been investigated by a combination of classical morphological data and a polymerase chain reaction (PCR)-based molecular approach. The aim of the study was to investigate possible changes in the composition of the cyanobacterial community along the salinity and nutrient gradients. For this purpose partial gene sequences of cyanobacterial 16S rDNA and of two functional genes (ggpS- salinity tolerance marker, isiA- iron starvation marker) were amplified and compared with total community DNA. Random distribution of ggpS genotypes along the salinity gradient suggests that synthesis of the osmolyte glucosylglycerol is not restricted to higher salinity sampling sites. Most of the isiA sequences formed a new homogenous cluster in a phylogenetic tree, which indicates that the indigenous cyanobacterial community comprises a group of unknown species. Minimum iron concentrations, which can activate isiA transcription in model cyanobacteria, occurred at a few sampling sites with high phytoplankton biomass and moderate salinity. Nevertheless, isiA expression could be detected at all sampling sites, which indicated restricted iron supply to cyanobacterial phytoplankton in summer.
