In den vorliegenden Untersuchungen wurden insbesondere mit chromatographischen Verfahren Phosphatide und daraus hergestellte Diglyceride aufgetrennt und anschließend bestimmt. Damit wurden Dünnschicht–und Gaschromatographie zur Analytik kombiniert. Die Wiederfindungsrate von Phosphatiden nach DC und Elution war hierbei von besonderem Interesse. Für Bezugsmessungen wurden die Phosphatidwerte nach Veraschung und die enzymatisch bestimmten Glyceringehalte zu wichtigen Parametern. Im weiteren wurde die Phospholipase‐C‐Spaltung von Phospholipiden und die Acetylierung daraus hergestellter Diglyceride überprüft. Die acetylierten Diglyceride wurden auf Kieselgel‐AgNO3‐Platten subfraktioniert. Die Untersuchungen an biologischem Material erfolgten bei Phosphatiden aus Seren und Eigelb und zum Teil auch an käuflichen Phosphatiden als Reinsubstanzen.
Die Positionsisomerie von Fettsauren in Phosphatidylcholin (PC) und Phosphatidylethanolamin (PE) von Membranen aus Schweineherz und Meerschweinchenherz wurde untersucht und das Phosphatidmuster erstellt. Zur spezifischen Freisetzung von Fettsauren aus sn-2 Position wurde Phospholipase A2 aus Schlangengift eingesetzt. Vor der A2-Spaltung wurden die membranstandigen Phospholipide mit ''C-Palrnitin-bzw. ''C-Olsaure im in vim-Experiment markiert. Neben Diacylglycerophosphatiden kommen in Herzmembranen auch Alkenylacylglycerophosphatide (Plasmalogene) vor. Letztere werden von Phospholipase A2 im Zweiphasensystem sehr vie1 schlechter gespalten und wurden nach dunnschichtchromatographischer Abtrennung und Umesterung der Fettsauren mittels G C charakterisiert. Das Fettsauremuster von PC und PE wurde im Vergleich zu den freigesetzten Fettsauren und den Lysoverbindungen charakterisiert. Bei Plasmalogenen wurden die den Kettlangen von C16 ,, und C I 8 entsprechenden Aldehyde als Dimethylacetale im G C neben den Fettsauremethylestern nachgewiesen. Die Alkenylethergruppierung scheint die Abspaltung der Fettsaure in sn-2 Position durch Phospholipase A2 negativ zu beeinflussen. The Analysis of Phospholipids from Cardiac Membranes by Phospholipase A,.Phospholipase A2 from snake venom has been employed to analyse the positional distribution of fatty acids in glycerolipids from guinea pig and pig cardiac membranes. It is known that phospholipase A2 hydrolyses the fatty acids in sn-2 position of 1,2 diacylglycerophospholipids. In cardiac membranes phosphatidylcholine and phosphatidylethanolamine contain along with diacyl esters the corresponding alkenylether analogues of phospholipids. In the present experiments the alkenylether moieties were slowly hydrolysed by phospholipase A2 from snake venom. We therefore separated by TLC lysophosphatides from fatty acids and unattacked phospholipids. The latter were the plasmalogens. The separated lipids were characterized by GLC. In some experiments the phospholipids were labelled with "C-fatty acids before they were hydrolysed by phospholipase A2.The alkenyl ether chain of plasmalogens seems to negativly influence the hydrolysis of fatty acids in sn-2 position of those phospholipids.
A) Linter diesem Thema fand vom 26. bis 28. September 1989 in Lorsch ein medizinhistorisches Friihmittelalter-Symposium statt, im Rahmen der diesjahrigen Jubilaumsfeierlichkeiten der Stadt Lorsch anlai3lich der vor 1225 Jahren erfolgten Griindung des Klosters Lorsch (gleichzeitig feiert Lorsch 25 Jahre Stadtrechte). Das Symposium wurdeunterstutzt durch die Firma Boehringer Ingelheimvom Magistrat der Stadt ausgerichtet und stand unter der Schirmherrschaft von Burgermeister Ludwig Brunneng~ber. Die wissenschaftliche Leitung hatte Gundolf Keil (Wiirzburg), der schon fur die wissenschaftliche Durchfiihrung des dreijahrigen Proiektes verantwortlich gezeichnet hatte, dasinitiiert durch eine Lorscher Burgerin, Frau Adelheid Platte, gefordert durch den Heimat-und Kulturverein Lorsch unter dem Vorsitz von Paul Schnitzer und finanziell abgesichert durch den Magistrat der Stadt Lorsch sowie die Firma Boehringer Ingelheimunter beispielhaftem Zusarnmenwirken von Privatinitiative und Wissenschaft, kommunalen Mitteln und Industrieforderung zur kritischen Edition des Lorscher A rzneibucbs fuhrte, das demge-maf3 im Zentrum des Symposiums stand. Die sensationelle Zuweisung des bedeutendsten medizinischen Manuskripts der Baniberger Staatsbibliothek (Codex Medicinalis I) zum Skriptorium des ehemaligen Lorscher Klosters durch den Munchner Palaographen Bernhard Bischoff wurde iiber langere Zeit von der Wissenschaftsgeschichte nur unzureichend zur Kenntnis genommen. Erst das zunachst lokalhistorisch motivierte Engagement der Stadt Lorsch hat dazu gefuhrt, das alteste bekannte Arzneibuch deutscher Provenienz im gesamten Umfang zu edieren, zu iibersetzen und durch ein Fachglossar zu erschliefien, um auf dieser Grundlage der Erforschung der friihmittelalterlichen Medizin-und Pharmaziegeschichte neue Impulse zu geben. Dlie Ursache fur die bisher relativ geringe Beachtung vorsalernitanischer Rezept-sammlungen mag man wohl in deren scheinbar schwachen Wirkungsgeschichte sehen: Spatestens nach dem 12. Jahrhundert und der Ubernahme arabischen sowie byzantinischen Wissens in die abendlandische Medizin und die sich immer mehr verselbstandigende Pharmazie wurden sie alle vom wirkungsgeschichtlich vie1 starkeren Antidotarium Nicolai iiberstrahlt, welches bekanntlich seinen Einflufi bis in die Moderne geltend machte. Das Lorscher Arzneibuch wurde um 795in der Benediktinerabtei konzipiert und geschrieben. Neben umfangreichen pharmakotherapeutischen Schriften (Rezeptsammlungen, Drogen-Austauschliste, Synonymen-Verzeichnis), diatetischen Traktaten (,Anthimus-Brief', Monatsregeln, Vier-JahreszeitenTrank) und isagogischen Elementen bietet es insbesondere durch seine medizinalpolitischen Programmschriften (,Verteidigung der Heilkunde', ,Kosmas-und-Damian-Verse') AnlaB zu einer eingehenden Betrachtung jener Umstande, welche die Weitergabe medizinischen Wissens im friihen Mittelalter behinderten, namlich vor allem jene christlich rnotivierte Medizinfeindlichkeit, mit deren Argumenten sich der Kompilator des Lorscher Arzneibuch auseinanderzusetzen hatte un...
