Starting from (−)-quinic acid, the title compound was synthesized in seven chemical steps and an overall yield of 35−38%. The route of the improved Gilead synthesis was not changed. However, significant improvements in each step led to a doubled overall yield, a 30% reduction in the number of unit operations, and an excellent quality (≥99%) of the resulting epoxide. A highly regioselective method for the dehydration of a quinic acid to a shikimic acid derivative and for the reduction of a cyclic ketal was found. Alternatively, the title compound was synthesized in six chemical steps and 63−65% yield from commercially available (−)-shikimic acid. Compared to the optimized quinic acid route, the production time was reduced by about 50%. The quality of epoxide produced from either natural product was equivalent. Therefore (−)-shikimic acid is the preferred raw material. The absolute configuration of the epoxide was determined by X-ray single crystal structure analysis and it was demonstrated that the epoxide was stereoisomerically pure.
Several approaches are utilized to incorporate rhodium-con-plexes, RhCp units are attached, as in Co,RhCp(CO),(ptaining building blocks in alkyne-bridged trinuclear clusters. Es sind hochmolekulare Rhodiumcluster bekannt I), und rhodiumhaltige Mehrkernkomplexe wurden schon intensiv fur Katalysestudien eingesetzt2). Letzteres war auch fur uns der AnlaD, rhodiumhaltige Cluster, speziell heterometallische, zu untersuchen. Solche Verbindungen waren noch vor kurzem in relativ kleiner Zahl bekannt 3), wobei wesentliche Syntheseverfahren auf Stone4) und Shriver 51 zuruckgehen. Wir haben bisher die Clusterexpansions-Reagenzien Cp-Rh(CO)* (Cp = C5H5 oder C,Me,) zum Aufbau vier-und funfkerniger Cluster benutzt 6 --8 ) . Fur Reaktivitgtsstudien erschienen uns jedoch Dreikerncluster, speziell solche rnit Rh(C0)3-Baueinheiten, vorteilhafter. Wir fuhrten deshalb die in dieser Arbeit beschriebenen Studien zur Gewinnung alkinverbruckter Hetero-Rhodium-Cluster durch. AusgangskomplexeDas Einbringen von Rh(COh-Gruppen verlangt entweder geeignete Vorstufen zum Metallaustausch oder einfache Rh(CO),-haltige Komplexe, an die andere Organometall-Baueinheiten anzubringen sind. Fur den ersteren Zweck erwies sich der Heterometallcluster 1') als geeignet. Fur den letzteren Zweck hofften wir auf die Reaktivitat der labilen Zweikernkomplexe 3, die nach Horvath'') in Gegenwart von iiberschussigem CO und Alkin rnit den stabilen Vierkernclustern 2 im Gleichgewicht stehen. Wir synthetisierten daher aus Co2Rh2(C0)12111 und MeC = CMe bzw. MeC = CPh die von Horvath") noch nicht beschriebenen Cluster 2a und b.Fur den alternativen Weg des Ankondensierens von RhCp-Einheiten muDten alkinverbruckte Zweikernkomplexe ausgewahlt werden, wobei sich zeigte, daI3 solche rnit terminalen Alkinen besser geeignet sind. Als am leichtesten zugangliche Zweikernkomplexe boten sich dann 4 a und b12) an. Daraus lieBen sich durch Metallaustausch rnit NaMoCp(C0)3 die Hetero-Zweikernkomplexe 5a 131 und 5 b gewinnen. Die Ausbeuten an Zweikernkomplexen 1 Me 3a: R = Me, 3b: R = Ph H\ 4a: R = Me, 4b: R = COOMe 2a: R = Me, 2b: R = Ph 5a: R = Me, 5b: R = COOMe waren gut, als Nebenprodukte fielen alkylidinverbriickte CoMo2-Cluster an (siehe exp. Teil), die vermutlich auf die intermediare Umwandlung von CO~(CO)~(K-HC = CR) in C O~( C O )~(~~-C -CH2R) 14) zuriickgehen. Cluster mit Rh(CO)3-BaueinheitenAls einzigen neuen Cluster rnit Rh(C0)3-Baueinheit erhielten wir das thermisch mal3ig stabile 6 durch Metallaustausch einer Fe(CO),-Gruppe 15). The expansion of vinylidene-bridged FeCoz and RuCoz clusters 1 a -d, 2d with (cyclopentadieny1)rhodium building blocks 3a, b mainly yields alkyne-bridged FeCozRh Fur die Komplexchemie terminaler Alkine gilt allgemein, dalj der Alkinligand leicht der Tautomerisierung zum Vinylidenliganden unterliegt I), Dies wurde mehrfach fur einkernige,) und zweikernige Komplexe3) gezeigt, und wir konnten an zahlreichen Dreikernclustern die Umwandlung des p,-Alkin-in den p,-Vinyliden-Liganden beobachten4). Mit der Tautomerisierung ist im Falle der Cluster auch eine Verschiebung der Reaktivitat der C,-Einheit vom terminalen Alkin-C-Atom" zur vinylidenischen CHR-Gruppe6) verbunden.Wir sind bemuht, diesen Spezialfall einer metallkontrollierten organischen Reaktivitatssteuerung auszunutzen und die Bedingungen fur die Bevorzugung der p,-Vinyliden-bzw. p3-Alkin-Form auszuloten. Beim Studium entsprechender Silylalkin-Silylvinyliden-Umlagerungen unter Silylgruppenwanderung') wurden wir dabei auf die Moglichkeit aufmerksam, daD in Vierkernclustern wieder die Alkin-Form gegenuber der Vinyliden-Form bevorzugt ist.Wir wandten uns daher noch einmal den vinylidenverbruckten Basis-Clustern 1 und 24) zu rnit der Absicht, sie zu Vierkernclustern zu expandieren und so weiteres Material zur Alkin-Vinyliden-Tautomerie zu gewinnen. Nach einigen Vorversuchen erwiesen sich dann die Rhodiumverbindungen 3 als gute Expansionsreagenzien, die den Einstieg in die hier beschriebenen Reaktionen und Produkte eroffneten. Cluster-ExpansionenDie drei Cluster l b -d lieljen sich mit 3a durch Erhitzen in Toluol in die Vierkerncluster 4 b -d uberfuhren. Die Ausbeuten waren maljig bis gering, die Produkte thermisch und oxidativ recht stabil. Mit der Cluster-Expansion ging regelma13ig eine Umlagerung des Vinyliden-zum Alkinliganden einher. Auf die gleiche Weise gingen l a und b rnit 3b in die Cluster 5a und b uber, und das exemplarisch untersuchte RuCo,-System 2d bildete mit 3 a analog den Vierkerncluster 6 d.
Durch Umsctzung von alkinvcrbriickten Dicobalt-Komplcxen mit Fe2(CO), bzw. Fe3(C0)12 wurden alkin-und vinylidenverbriickte Cluster des allgemeinen Typs FeCo2(C0)9(p3-L) zuganglich. Auf diese und die homologen RuCo&luster liekn sich die Reaktionstypen Metallaustausch und Alkin-Vinyliden-Umlagerung anwenden. Kombination beider Reaktionstypen in der geeigneten Reihenfolge lieferte insgesamt 16 neue Hetero-Dreikerncluster mit p3-Alkin-Liganden und 29 entsprechende Verbindungen mit p,-Vinyliden-Liganden. Es kamen interne Alkine R2Cz rnit R = Me, Et, Ph und terminale Alkine HC2R mit R = H, Me, I-Bu, Ph zum Einsatz; letztere waren auch die Vorlaufer der entsprechenden Vinyliden-Liganden C2HR. Die entstehenden Metallgeruste waren vom Die bereits sehr umfangreiche Chemie metallorganischer Alkinkomplexe erlebt in den letzten Jahren eine Renaissance. Die zunehmende Leistungsfahigkeit strukturanalytischer Methoden hat es moglich gemacht, die ungewohnlichen Reaktivitaten des Alkinliganden und die zahlreichen Umwandlungsmoglichkeiten der Komplexgeruste zu verfolgen. Fur Cluster rnit Alkinliganden bedeutet dies regioselektive Reaktionen'), Auf-und Abbausequenzen im anorganischen" wie im organi~chen~) Molekulteil sowie Geriist~mwandlungen~). Weitere Herausforderungen in der Cluster-Alkin-Chemie bestehen darin, Stereo-Diskriminierung durch Einfuhrung von Chiralitat anzustreben sowie die oft fur Komplexsynthesen zu reaktiven terminalen Alkine systematisch einzubringen. Beides ist nicht nur von theoretischem Intere~se~,~), sondern eroffnet auch neue Reaktionswege',').Wir haben durch Uberdachung von RUCO~(CO),~ unter sehr milden Bedingungen einen Zugang zur MehrzentrenAnbindung von Acetylen und terminalen Alkinen gefunden'), wodurch prochirale RuC~~(CO)~(p~-L)-Cluster (L = Alkin oder Vinyliden) erhaltlich wurden. Wir erwarteten, da13 mit den verschiedenen Metallaustausch-Methoden") daraus chirale RuCoM-Cluster entstehen sollten. Da entsprechende FeCo2(C0)9(p3-L)-C1uster bisher nur unbequem und unsystematisch zuganglich waren, suchten wir auch fur sie nach einer gezielten Synthese als Voraussetzung fur den anschlieBenden Metallaustausch.
