Borneol is a bicyclic monoterpenoid alcohol commonly used in traditional Chinese and Indian medicine. It is extracted from the essential oil of various medicinal plants. It has antibacterial, analgesic, and anti-inflammatory action proven in studies that used oral and intraperitoneal applications of this monoterpene in mice. The current study was designed to develop a topical gel formulation containing the monoterpene borneol using carbopol as gel base and to evaluate its stability. The prepared formulation was subjected to physical characterization and physical-chemistry assessment. The gel was prepared from carbopol and 5% of borneol. The prepared gel was subjected to pharmacotechnical tests such as its pH, viscosity, conductivity, spreadability, centrifugation, and accelerated stability with freezing-thaw cycle. The borneol was successfully incorporated into the carbopol formulation. Borneol gel (BG5) showed good stability after eight months of its development and after 12 days in the freeze-thaw cycle, not showing statistical difference in pH value, conductivity, and viscosity before and after test. Furthermore, the formulation showed a good spreadability. Therefore, it was concluded that the formulation could be very promising alternative for the topical or transdermal treatment of skin diseases.
RESUMO:O objetivo desse estudo foi realizar um ensaio toxicológico pré-clínico para analisar a toxicidade do chá das folhas de Morus nigra L. (Moraceae). A toxicidade subcrônica do chá (CFMn) foi avaliada durante 30 dias por via oral em ratos. Ao grupo controle foi administrado água, para comparação. Durante o período experimental foi avaliada a presença de sinais de toxicidade, variação do peso corporal, e o consumo de líquido e alimento. Ao final do experimento o sangue dos animais foi retirado para análise de parâmetros hematológicos e bioquímicos. Não foram observados mortalidade e sinais de toxicidade indicando baixa toxicidade da planta. Não houve alterações nos parâmetros hematológicos e bioquímicos. Nas condições do estudo, o CF-Mn pode ser considerado de baixa toxicidade, pois não produziu efeitos tóxicos nos animais tratados. . The subchronic toxicity of this tea (CF-Mn) was orally evaluated during 30 days in rats. The control group was given water for comparison. During the experimental period, signs of toxicity, body weight variation, and water and food consumption were assessed. At the end of the experiment, the blood of animals was removed for analysis of hematological and biochemical parameters. No mortality and no toxicity signs were observed, indicating low toxicity of the plant. There was no alteration in the hematological and biochemical parameters. Under the study conditions, CF-Mn can be considered of low toxicity since it did not produce toxic effects in treated animals. Palavras
Neoglaziovia variegata is a Bromeliaceae plant species widely found in Brazil with several pharmacological properties, including photoprotective activity. Although herbal-based active ingredients have been applied in cosmetic products, especially for skin treatment, its application in sunscreen formulations remains unexplored. The aim of this work is to evaluate the photoprotective effect of cosmetic formulations containing hydroalcoholic extract of N. variegata (Nv-HA). Initially, the phenolic and flavonoid total content of Nv-HA were determined. The photoprotective activity of Nv-HA was subsequently assessed using a spectrophotometric method. Nv-HA was incorporated in O/W emulsions in the presence or absence of synthetic filters and their photoprotective efficacy was evaluated by spectrophotometric SPF determination. Finally, the stability study of the formulations was performed through the freezing/defrosting method. Nv-HA showed significant phenolic and flavonoids content (61.66 ± 5.14 mg GAE/g and 90.27 ± 5.03 mg CE/g, resp.). Nv-HA showed SPF values of 5.43 ± 0.07 and 11.73 ± 0.04 for the concentrations of 0.5 and 1.0% (v/v), respectively. It was verified that Nv-HA potentiated the photoprotective effect of formulations containing only synthetic filters. Furthermore, the formulations have remained stable at the end of the preliminary stability study. According to the results, it was concluded that Nv-HA can be used as a chemical filter in cosmetic formulations.
The aim of this study was to investigate the effect of gels containing the monoterpene borneol in induced oral mucositis using an animal model. Gels were prepared with borneol at 1.2% and 2.4% (w/w). Oral mucositis was induced by administration of three doses of 5-fluorouracil (30 mg/kg, i.p.) and injury with acetic acid (50%, v/v) soaked in filter paper applied to right cheek mucosa for 60s. Four subgroups comprising 12 animals each were formed. Six animals from each group were sacrificed at days seven and fourteen after oral mucositis induction. Mucous samples were processed and stained with hematoxylineosin and Masson's Trichrome. The semiquantitative evaluation involved observation of inflammatory parameters. ImageJ® software was used in the quantitative evaluation. For statistical analyses, Two-way ANOVA, followed by Tukey's post-test (p <0.05), were employed. Borneol 2.4% gel proved effective in the treatment of oral mucositis with statistically significant differences between groups for angiogenesis control, inflammatory cell count reduction and percentage neoformed collagen increase. The confirmation of anti-inflammatory and healing action of borneol in oral mucositis in rats renders it a good marker for predicting this activity for plant extracts rich in this substance.
Morus nigra L. is a species popularly known in the Northeast of Brazil as “amora miúra”. This species is a source of flavonoids with antioxidant activity. Antioxidants play an important role in the preservation of cosmetic formulations, and they neutralize free radicals. The objective of this study was to develop a topical emulsion containing leaf extract of Morus nigra L., as well as to evaluate the stability, antioxidant and antibacterial activities of the formulations. A crude hydroalcoholic (70%) extract of M. nigra leaves (MnCE) was submitted to high-performance liquid chromatography with diode-array detection (HPLC–DAD) analysis and incorporated into an anionic base emulsion. Antioxidant activity was evaluated according to the DPPH (2,2-diphenyl-1-picrylhydrazyl) method, and the stability of the formulation was assessed for 90 days, submitting the emulsion to storage at 4, 20, and 37 °C. Microdilution techniques evaluated the antibacterial activity and a challenge test assessed the microbiological stability. Analysis by HPLC–DAD identified the flavonoids rutin and isoquercetin in the M. nigra extract. The emulsion and plant extract presented antioxidant activity, and the stability of the emulsion was preserved in terms of pH value and viscosity—which did not show significant changes, except for the spreadability, which was affected by the temperature. The antioxidant activity did not change significantly, except for the sample under 4 °C, which showed a considerable decrease in activity. The crude hydroalcoholic extract and formulation showed antimicrobial activity and the emulsion was considered stable in terms of organoleptic, physicochemical, and microbiological properties.
