PURPOSE: Development of an improved animal model for studying skin burns in rats. METHODS: Twenty-four male Wistar rats were randomly assigned to four groups (n=6): G1-Control, G2-T100°C, G3-T150°C and G4-T200°C. Two 10 x 10 mm squares were outlined with a sterile surgical marker on each side and along the vertebral column using a prepared template positioned between the anterior and posterior limbs. G2-G4 rats were subjected to 100°C, 150°C and 200ºC thermal burns, respectively. G1 rats served as controls. Burns were inflicted by applying a copper plate connected to an electronic temperature controlling device to the dorsal skin of anesthetized rats. Four burns were produced on each animal (total area: 4 cm 2 / animal) leaving about 1 cm of undamaged skin between burn areas. Analgesia was administered during 24 h after burn injury by adding 30 mg codeine phosphate hemihydrate to 500 ml tap water. RESULTS:The application of 100°C and 150ºC resulted in partial thickness skin burns with central reepithelialization of the burned area only at 100°C. In G4 group the whole thickness of the skin was injured without central reepithelialization. However, there was marginal reepithelialization in all groups. CONCLUSION: The model studied is inexpensive and easily reproducible, enabling the achievement of controlled burns with partial or total impairment of the skin in experimental animals. MÉTODOS: Vinte e quatro ratos Wistar, machos, foram distribuídos aleatoriamente em quatro grupos (n=6): G1-Controle, G2-T100°C, G3-T150°C e G4-T200°C. Dois quadrados medindo 10x10 mm foram delineados com um marcador cirúrgico estéril em cada lado e ao longo da coluna vertebral e posicionados entre os membros anteriores e posteriores, utilizando um molde previamente preparado. Os ratos dos grupos G2-G4 foram submetidos a queimaduras térmicas de 100°C, 150°C e 200°C, respectivamente. O grupo G1 foi utilizado como controle. As queimaduras foram infligidas pela aplicação de uma placa de cobre, ligada a um dispositivo de controle eletrônico de temperatura, na pele dorsal de ratos anestesiados. Quatro queimaduras foram produzidas em cada animal (área total: 4 cm2/animal), deixando cerca de 1 cm de pele intacta entre as áreas queimadas. Analgesia foi obtida durante 24 horas após a queimadura por adição de 30mg de fosfato hemi-hidratado de codeína a 500 ml de água potável. RESULTADOS: A aplicação 100°C e 150°C resultou na produção de queimaduras profundas comprometendo parte da espessura da pele, com reepitelização central da área queimada, somente a 100°C. No grupo G4 houve lesão de toda a espessura da pele sem reepitelização central. Entretanto, observou-se reepitelização marginal em todos os grupos estudados. Purpose:To evaluate the effects of the oxidative stress in an experimental model of torsion/detorsion of the spermatic cord and the legitimacy of this model for oxidative stress studies. Methods: Forty-eight male Wistar rats were randomized in two groups (n=24): G-1 (Sham) and G-2 (Ischemia/Reperfusion). All rats received intraperitoneal saline injections (2.0 ml), at 21, 9, and 1 h before right spermatic cord torsion or first sham operation. Detorsion or second sham operation was carried out 3 h later followed by testis and blood samples collection (T-0). Additional samples were collected at 1-3-6 h time-points for assessment of testis malonaldehyde, glutathione, and plasma total antioxidant power (TAP). Results: Spermatic cord torsion/detorsion induced a significant increase in testicular malonaldehyde contents and a significant decrease in glutathione concentrations in ischemic rats compared with sham animals. Additional increase in malonaldehyde levels occurred during reperfusion in G-2 rats. TAP was similar in both groups denoting absence of systemic effects in this study. Conclusion: Torsion/detorsion of the spermatic cord for 3 h induces significant lipid peroxidation and reduction in glutathione content of the testis and is, therefore, a valid model for studying the oxidative stress effects of the ischemia/ reperfusion injury in young rat testis. RESUMOObjetivo: Investigar os efeitos do estresse oxidativo utilizando um modelo experimental de torção/destorção do cordão espermático e a aplicabilidade do modelo para estudo do estresse oxidativo. Métodos: Foram utilizados 48 ratos distribuídos aleatoriamente em dois grupos (n=24): G-1 (Simulado) e G-2 (Isquemia/Reperfusão). Todos os animais receberam injeções intraperitoniais de solução salina (2,0 ml) 21-9-1 horas antes da torção ou primeira operação simulada. A destorção/ segunda operação simulada (T-0) foi realizada após 3 horas, com coleta das amostras (testículo e sangue arterial) nesta ocasião e após 1-3-6 horas para determinação das concentrações de malonaldeído, glutationa e capacidade antioxidante total do plasma (CAT). Resultados: a torção do cordão espermático induziu aumento significante das concentrações de malonaldeído e redução simultânea das concentrações de glutationa nos ratos do grupo G-2, comparados ao grupo simulado. Ocorreu aumento adicional das concentrações de malonaldeído durante a reperfusão (G-2). A capacidade antioxidante total do plasma foi semelhante nos dois grupos estudados, denotando ausência de efeitos sistêmicos. Conclusão: A torção durante 3 horas seguida de destorção do cordão espermático induz um nível significante de peroxidação lipídica e redução da concentração de glutationa constituindo-se um modelo válido para o estudo dos efeitos do estresse oxidativo decorrente da lesão de isquemia/reperfusão no testículo de ratos jovens.
PURPOSE:To address the effects of fructooligosaccharides (FOS) intake on serum cholesterol levels. METHODS:We performed a search for scientific articles in MEDLINE database from 1987 to 2014, using the following English keywords: fructooligosaccharides; fructooligosaccharides and cholesterol. A total of 493 articles were found. After careful selection and exclusion of duplicate articles 34 references were selected. Revised texts were divided into two topics: "FOS Metabolism" and "FOS effects on plasma cholesterol." RESULTS:The use of a FOS diet prevented some lipid disorders and lowered fatty acid synthase activity in the liver in insulin-resistant rats. There was also reduction in weight and total cholesterol in beagle dogs on a calorie-restricted diet enriched with short-chain FOS. Another study found that 2g FOS daily consumption increased significantly serum HDL cholesterol levels but did not ensure a significant reduction in levels of total cholesterol and triglycerides.. Patients with mild hypercholesterolemia receiving short-chain FOS 10.6g daily presented no statistically significant reduction in serum cholesterol levels. However, when FOS was offered to patients that changed their lifestyle, the reduction of LDL cholesterol and steatosis was higher. CONCLUSIONS:Fructooligosaccharides intake may have a beneficial effect on lipid metabolism and regulation of serum cholesterol levels in individuals that change their lifestyle. FOS supplementation use in diets may therefore be a strategy for lowering cholesterol.
The oil-resin of copaíba presents discrete antilipoperoxidation action, intense antioxidant action, and antiinflammatory activity during the ischemia and reperfusion of randomized cutaneous flaps. The effects of ischemia-reperfusion are complex and substances capable of increasing the tolerance of tissue to those effects by reducing the production or neutralizing the action of free radicals are needed.
PURPOSE:To identify the current status of scientific knowledge in fructo-oligosaccharides (FOS), non-conventional sugars that play an important role in glycemia control. METHODS:We performed a search for scientific articles in MEDLINE and LILACS databases, from January 1962 to December 2011, using English/Portuguese key words: "blood glucose/glicemia", "prebiotics/prebióticos" and "dietary fiber/fibras na dieta". From an initial number of 434 references, some repeated, 43 references published from 1962 to 2011 were included in this study. The selected texts were distributed in three topics: (1) metabolism of FOS, (2) FOS and experimental studies involving glucose and (3) human studies involving glucose and FOS. RESULTS:Five studies have shown that the use of FOS reduces the fecal content and increases intestinal transit time. Experimental studies have shown that dietary supplementation with high doses (60 g/Kg) of propionate, a short-chain fatty acid decreased glycemia.The use of lower doses (3 g/kg) did not produce the same results. Study in subjects with diabetes type II showed that the addition of 8 grams of FOS in the diet for 14 days, caused a reduction in serum glucose. In another study with healthy subjects, there were no changes in glycemic control. CONCLUSIONS:This review demonstrates that consumption of FOS has a beneficial influence on glucose metabolism. The controversies appear to be due to inadequate methodological designs and/or the small number of individuals included in some studies.Key words: Review. Blood Glucose. Oligosaccharides. Prebiotics. MÉTODOS: Realizamos uma busca de artigos científicos nas bases de dados MEDLINE e Literatura Latino-Americana e do Caribe emCiências da Saúde (LILACS), a partir de janeiro 1962 a dezembro de 2011, usando como descritores termos Português/Inglês: "glicemia/ blood glucose, "prebióticos/prebiotics" e "fibras na dieta/dietary fiber. A partir de um número inicial de 434 referências, algumas repetidas, 43 referências foram consideradas adequadas para a finalidade deste estudo e foram, portanto, selecionadas para análise. Os textos selecionados foram distribuídos em três tópicos: (1) metabolismo de FOS (2), FOS e estudos experimentais envolvendo glicose e (3) estudos em humanos envolvendo glicose e FOS. RESULTADOS: Cinco trabalhos mostraram que o uso de FOS diminui o conteúdo fecal e aumenta o tempo do transito intestinal.Estudos experimentais demonstraram que a suplementação dietética com altas doses (60 g/Kg) de propionato, um ácido graxo de cadeia curta, diminuiu a glicemia. A utilização de doses menores (3 g/Kg) não produziu os mesmos resultados. Em indivíduos diabéticos tipo II a adição de 8 g de FOS na dieta, durante 14 dias, induziu uma redução da glicemia. Purpose:To evaluate the protective role of dimethylsulfoxide (DMSO) in a rat model of testis ischemia/reperfusion (I/R). Methods: Twenty-four male Wistar rats were randomized in two equal groups. Control rats (G-1) received saline 2.0 ml intraperitoneally (ip) 21, 9 and 1 h before torsion. Experimental rats (G-2) received ip injections of 3% aqueous solution of DMSO, 0.1ml/10g body weight. Saline was added to complete 2.0ml when necessary. I/R injury was induced in anesthetized rats by torsion of the right testis lasting 3 hours. Testis and blood samples were collected at the end of ischemia (T-0) and 3 hours later (T-3) for assessment of testis malonaldehyde (MDA), reduced glutathione (GSH), and plasma total antioxidant power (TAP). Results: MDA levels decreased significantly in G-2 rats compared with G-1 animals in all time-points. GSH levels increased significantly in T-0 and T-3 time-points in DMSO pretreated rats compared with G-1 rats. GSH levels increased significantly during reperfusion in G-2 rats. TAP was similar in both groups denoting absence of systemic effects in this study. Métodos: Vinte e quatro ratos machos Wistar, randomizados em dois grupos iguais e tratados com solução salina 2,0 ml (grupo Controle-G-1) ou solução de DMSO (3%, 0,1ml/10g peso corporal), acrescida de solução salina até completar 2,0 ml (grupo Experimento-G-2) por via intraperitoneal, as 21, 9 e 1 h antes da torção. .A lesão de I/R foi induzida por torção do testículo direito e mantida por 3 horas, em ratos previamente anestesiados. Amostras (sangue e testículo direito) foram coletadas ao término da isquemia (T-0) e após 3 horas de reperfusão (T-3) para determinação das concentrações de malonaldeído (MDA), glutationa reduzida (GSH) e capacidade antioxidante total do plasma (CAT). Resultados: As concentrações de MDA diminuíram significativamente nos ratos do G-2. comparados ao G-1, nos tempos estudados. As concentrações de GSH aumentaram significativamente nos testículos dos ratos do G-2 comparado aos controles no T-0 e no T-3 e durante a reperfusão no G-2. Os valores obtidos na análise do CAT foram semelhantes. Conclusão: O pré-tratamento com DMSO reduz a peroxidação lipídica e o estresse oxidativo decorrentes da torção/destorção do testículo, no rato.
