Interleukin (IL)-33 is involved in T helper (Th)2-biased immune responses in mice infected with Schistosoma, but the precise mechanism remains to be elucidated. Herein, we investigated the role of IL-33 and its receptor ST2L in hepatic granuloma pathology induced by Schistosoma japonicum infection. We found that IL-33 induced the increased production of IL-5 and IL-13 from splenocytes and liver mononuclear cells (MNCs) of infected mice. The infected mice developed significantly higher number of ST2L-expressing cells in spleen and liver. Most of the ST2L-expressing cells in liver were F4/80+ macrophages, indicating the key role of macrophages in the response to IL-33. However, the liver MNCs in male-only worm infection had a poor response to IL-33, though elevated serum IL-33 was observed. ST2L+F4/80+ cells were lower in male-only worm infection than that of mixed infection. IL-33 and soluble egg antigen (SEA) upregulated ST2L expression on macrophages in vitro and ST2L-expressing macrophage displayed MHCII-CD11b+M2 phenotype. Macrophage deletion significantly attenuated IL-33-induced type 2 immunity and egg granuloma formation during S. japonicum infection. These data demonstrate that IL-33 contributes to hepatic granuloma pathology through induction of M2 macrophages during S. japonicum infection.
Abstract:Irrigation is crucial to agriculture in arid and semi-arid areas and significantly contributes to crop development, food diversity and the sustainability of agro-ecosystems. For a specific crop, the separation of its irrigated and rainfed areas is difficult, because their phenology is similar and therefore less distinguishable, especially when there are phenology shifts due to various factors, such as elevation and latitude. In this study, we present a simple, but robust method to map irrigated and rainfed wheat areas in a semi-arid region of China. We used the Normalized Difference Vegetation Index (NDVI) at a 30ˆ30 m spatial resolution derived from the Chinese HJ-1A/B (HuanJing(HJ) means environment in Chinese) satellite to create a time series spanning the whole growth period of wheat from September 2010 to July 2011. The maximum NDVI and time-integrated NDVI (TIN) that usually exhibit significant differences between irrigated and rainfed wheat were selected to establish a classification model using a support vector machine (SVM) algorithm. The overall accuracy of the Google-Earth testing samples was 96.0%, indicating that the classification results are accurate. The estimated irrigated-to-rainfed ratio was 4.4:5.6, close to the estimates provided by the agricultural sector in Shanxi Province. Our results illustrate that the SVM classification model can effectively avoid empirical thresholds in supervised classification and realistically capture the magnitude and spatial patterns of rainfed and irrigated wheat areas. The approach in this study can be applied to map irrigated/rainfed areas in other regions when field observational data are available.
Shotgun proteomics has grown rapidly in recent decades, but a large fraction of tandem mass spectrometry (MS/MS) data in shotgun proteomics are not successfully identified. We have developed a novel database search algorithm, Open-pFind, to efficiently identify peptides even in an ultra-large search space which takes into account unexpected modifications, amino acid mutations, semi-or non-specific digestion and co-eluting peptides. Tested on two metabolically labeled MS/MS datasets, Open-pFind reported 50.5-117.0% more peptide-spectrum matches (PSMs) than the seven other advanced algorithms. More importantly, the Open-pFind results were more credible judged by the verification experiments using stable isotopic labeling. Tested on four additional large-scale datasets, 70-85% of the spectra were confidently identified, and high-quality spectra were nearly completely interpreted by Open-pFind. Further, Open-pFind was over 40 times faster than the other three open search algorithms and 2-3 times faster than three restricted search algorithms. Re-analysis of an entire human proteome dataset consisting of ~25 million spectra using Open-pFind identified a total of 14,064 proteins encoded by 12,723 genes by requiring at least two uniquely identified peptides. In this search results, Open-pFind also excelled in an independent test for false positives based on the presence or absence of olfactory receptors. Thus, a practical use of the open search strategy has been realized by Open-pFind for the truly global-scale proteomics experiments of today and in the future.
Angiostrongylus cantonensis can induce central nervous system (CNS) injury and cause human eosinophilic meningitis. The CNS has been found to have high expression of interleukin 33 (IL-33), which promotes the pathogenesis of T-helper 2 (Th2)-related disease. Given the predominantly type 2 response induced by A. cantonensis-infected mice and human, it is likely that IL-33 may play a role in aiding this process. We report here that IL-33 protein and ST2L messenger RNA (mRNA) transcripts in the brains were upregulated during A. cantonensis infection and that both splenocytes and brain mononuclear cells became IL-33 responsive and produced interleukin 5 and interleukin 13. Furthermore, administration of IL-33 to A. cantonensis-infected mice enhanced ST2L expression and cytokine production. Interestingly, brain IL-33 protein and ST2L mRNA levels were elevated 14-21 days after infection in BALB/c mice, compared with C57BL/6 mice. Thus, our data indicate that IL-33 produced in the brain may function as an inflammatory mediator in eosinophilic meningitis induced by A. cantonensis.
