Local riverine experts from the North Araguaia microregion use a wide variety of medicinal plants in self-care health, especially those species used to treat IPD. The therapeutic potential of some of these plants has been scientifically validated; however, there are other species whose pharmacological effects and safety remain to be properly investigated. Thus, the present study, aside from being a basis for future chemical, pharmacological and agronomic bioprospecting studies, may contribute to the development of the management, conservation and sustainable use of medicinal flora in the microregion studied.
RESUMO:O presente trabalho verificou a utilização de plantas medicinais encontradas no Cerrado mato-grossense para o tratamento de hiperlipidemias e obesidade. Entrevistas com 180 pessoas acima de 50 anos foram realizadas em Nova Xavantina-MT. O questionário abordou nome popular, parte utilizada, forma de preparo e uso das espécies citadas, além de informações gerais sobre o uso de plantas. -Hil.). A folha foi a parte mais utilizada (46%) e o preparo das plantas ocorre principalmente por meio de infusão citado por 36,5% dos entrevistados. This study checked the use of medicinal plants found in the Cerrado mato-grossense for the treatment of hyperlipidemia and obesity. Interviews with 180 people over 50 years were held in Nova Xavantina-MT. The questionnaire addressed popular name, part used, type of preparation and use of the species mentioned, beyond general information about the use of plants. The ten plants most cited were collected, identified and studied through a literature review. Most respondents (95.6%) said using medicinal plants regularly, while 71.5% of them inherited the knowledge of plants of parents and grandparents and 94.20% reported the use to advise youngsters. Furthermore, 93.6% attested that the plants are more effective than the drugs from pharmacy and 57% considered the strong effect of the same, i.e., they always solved health problems. Regarding adverse effects, 95.9% said they have never felt them after the use of plants. The SIECm is safe, contain useful phytochemicals and exhibited significant gastroprotective/anti-ulcer effects. The results justify its folkloric usage, and provided scientific evidence of its potential as a source of new phytodrug to treat gastric ulcers.
Macrosiphonia velame (Apocynaceae), popularly known as "velame-branco", is mainly used for treating inflammatory conditions. The antiinflammatory, antinociceptive and antipyretic effects of the hydroethanolic extract of the xylopodium from M. velame (HEMv) were evaluated using several animal models. HEMv showed low acute oral toxicity with LD 50 of 4.176 ± 218.5 mg/kg in mice. In tests of carrageenan and dextran-induced paw edema and carrageenan-induced pleurisy in rats, and croton oilinduced cutaneous dermatitis in mice, HEMv presented systemic and topical antiinflammatory activities. In experiments of nociception induced by acetic acid, formalin and capsaicin in mice, the HEMv evidenced an antinociceptive effect, being active against both inflammatory and neurogenic pain. Additionally, the HEMv prevented brewer's yeast-induced pyrexia in rats. It is likely that the pharmacologic mechanism of HEMv may involve the inhibition of different mediators of the inflammatory response, such as histamine, serotonin, prostaglandins and leukotrienes. A preliminary phytochemical study was also undertaken on HEMv, which revealed the presence of flavonoids, phenolic compounds, pentacyclic triterpenoids, saponins, coumarins, catechins, tannins, and alkaloids. Taken together, these results suggest that M. velame extract has antiinflammatory, antinociceptive and antipyretic properties and further validate the traditional use of this plant in inflammatory conditions. Avaliou-se nesse estudo, os efeitos antiinflamatório, antinociceptivo e antipirético do extrato hidroetanólico do xilopódio de M. velame (HEMv) em modelos animais. O HEMv apresentou baixa toxicidade aguda oral, com DL 50 = 4.176 ± 218,5 mg/kg nos camundongos. Nos testes de edema de pata por carragenina e dextrana e pleurisia por carragenina em ratos e dermatite cutânea por óleo de croton em camundongos, o HEMv apresentou atividade antiinflamatória sistêmica e tópica. Nos experimentos de nocicepção induzida por ácido acético, formalina e capsaicina em camundongos, o HEMv inibiu a resposta nociceptiva nos três modelos, mostrando-se ativo tanto na dor de origem inflamatória como neurogênica. Adicionalmente, o HEMv bloqueou a hipertermia induzida por levedura de cerveja nos ratos. É provável que os efeitos farmacológicos observados para o HEMv decorram da inibição de mediadores da resposta inflamatória produzidos pelas vias da COX e/ou LOX bem como da inibição da liberação de histamina e serotonina. A análise fitoquímica do HEMv revelou a presença de flavonóides, compostos fenólicos, triterpenóides pentacíclicos, saponinas, cumarinas, catequinas, taninos catéquicos e alcalóides. Terminalia argentea Mart. (Combretaceae), known mainly as "capitão", is a native tree, not endemic, that occurs in the Amazon, Caatinga, Cerrado and Atlantic Forest in Brazil. Leaf infusion is popularly mentioned by riverine communities that inhabit the microregion of Northern Araguaia (Mato Grosso, Brazil) for the treatment of gastric ulcer, bronchitis and haemorrhage. Considering the wide medicinal use, lack of studies that evaluate the safety of use and the scarcity of phytochemical studies of T. argentea leaves, this work was carried out with the objective of evaluating the toxicity of the hydroethanolic extract of the leaves of T. argentea Mart. (HETa) in experimental models in vivo and in vitro, as well as to advance the phytochemical analysis of HETa. Materials and methods: HETa was prepared by macerating the leaf powder in hydroethanolic solution. Phytochemical characterisation was carried out by thin-layer chromatography (TLC), high-performance liquid chromatography (HPLC) and mass spectrometry through direct flow infusion coupled with electrospray ionization and ion-trap analyzer (DFI-ESI-IT-MS analyses) The contents of phenols, flavonoids and phytosterols were analysed by colorimetric methods. Cytotoxicity was assessed by the Alamar blue assay on Chinese hamster ovary epithelial cells (CHO-K1) and human gastric adenocarcinoma cells (AGS). In vitro genotoxicity of HETa (10, 30 or 100 μg/mL) was assessed by micronucleus (MN) and comet tests using CHO-K1 cells. The acute toxicity assessment was performed by oral administration of HETa in single dose Swiss mice (males and females) up to 2000 mg/kg and sub-chronic toxicity by daily oral administration of HETa (50, 200 and 800 mg/kg) in Wistar rats for 30 days. The parameters related to the clinical and toxicological observations were determined every 6 days and at the end of the treatment the blood was collected for biochemical and haematological analysis, and some organs were removed for macroscopic and histopathological analysis. Results: Preliminary phytochemistry and TLC analysis of HETa revealed the presence of phenolic compounds (18.8%), flavonoids (10.8%), saponins, tannins and phytosterols (19%). The HPLC data revealed the presence of gallic acid, rutin, ellagic acid, catechin, quercetin and kaempferol. In the analysis by DFI-ESI-IT-MS, the presence of gallic acid, rutin, ellagic acid and quercetin was confirmed and identified caffeic acid, quinic acid, galloylmucic acid, quercetin xyloside, quercetin rhamnoside, quercetin glucoside, caffeoyl ellagic acid, quercetin galloyl xyloside, terminalin, quercetin galloyl glucose, corilagin, quercetin digalloyl xyloside, quercetin digalloyl glucoside, punicalin and punicalagin. HETa showed no cytotoxic effect on CHO-K1 and AGS cells. In the MN assay, HETa increased the number of MNs and nuclear buds (NBUDs) in binucleate cells at the three concentrations tested and the nucleoplasmic bridges (NPBs) number at 30 μg/mL. In the comet test, HETa (10 and 100 μg/mL) alone showed a genotoxic effect on CHO-K1 cells. ...
Objetivando-se avaliar nuggets elaborados a partir de aparas de jacaré do pantanal com adição de carne mecanicamente separada (CMS), cinco tratamentos (0, 15, 30, 45 e 60%) com adição de CMS de jacaré do pantanal foram elaborados e avaliados quanto à composição centesimal, pH, Aw, força de cisalhamento, índice de peróxido, bases voláteis totais, cor objetiva CIE L*, a* e b*. Observou-se diferença significativa para as análises de composição centesimal apresentando teores de umidade entre 39,54% a 48,03%. A proteína e cinzas aumentaram de acordo com o índice de CMS adicionada. Os nugetts com adição de CMS de jacaré do pantanal apresentaram valores superiores de luminosidade, croma a* e b* em relação ao sem CMS. A força de cisalhamento variou entre 0,27 a 0,35. Houve diferença significativa para Aw entre os tratamentos. O resultado microbiológico dos tratamentos de nuggets foi evidenciado em acordo com o estabelecido por legislação. As formulações com adição de 45 e 60% CMS de jacaré do pantanal em nuggets de jacaré demonstraram melhores características físico-quimicas e todos os níveis de adição de CMS não interferiram na percepção sensorial de cor, textura, sabor e aparência global.
As amilases fazem parte do grupo de enzimas mais utilizadas nas indústrias de alimento, farmácia, bioetanol, têxtil e outros. Esses biocatalisadores são produzidos por vegetais, animais e microrganismos, especialmente por fungos. O objetivo deste trabalho foi bioprospectar linhagens fúngicas produtoras de amilase e caracterizar suas atividades enzimáticas. Para isso, 10 linhagens isoladas de amostras de solo foram disponibilizadas pela micoteca do IFMT Campus Avançado Lucas do Rio Verde foram selecionadas e reativadas. Dessas, 09 apresentaram viabilidade e foram submetidas ao teste atividade amilolítica (meio BDA a 2% de amido), onde foi determinado o Índice Enzimático (IE), obtido pela relação entre o diâmetro do halo de hidrólise e o diâmetro da colônia. As linhagens com maior IE (A25, A12, S01 e D12) foram selecionadas para a avaliação das condições ideais de temperatura e pH dos extratos enzimáticos. As linhagens selecionadas pertencem ao gênero Aspergillus e os melhores resultados obtidos foram observados em temperatura de 50º C e com pH 5,0. Nessas condições, os extratos enzimáticos amilolíticos apresentaram maior desempenho de hidrólise do amido, sendo capazes de degradar o substrato em 42,3% (D12), 44,7% (A12), 47,8% (A25) e 60,1% (S01), enquanto amostras de alfa-amilase comercial promoverams atividade relativas de até 86%. Considerando os resultados observados, pode-se concluir que entre as linhagens estudadas, quatro apresentaram considerável potencial para produção de amilases (S01, A25, A12 e D12), com destaque para a linhagem S01, que apresentou maior capacidade em produzir enzimas amilolíticas com potencial para exploração biotecnológica.
