Sea cucumber (H.Scabra) is a species of marine biota providing high economical and medical values. It has been hundreds years consumed for drug to cure various diseases in China. The aim of this research was to determine the level of the toxicity against brine shrimp larvae A.salina Leach in each dried sea cucumber (H.scabra) extract collected from Kenjeran Coast, Surabaya. The information would be beneficial for anticancer drug discovery .The research was conducted by extracting samples using both ethanol and n-hexane solvents. The concentrated extract obtained then were used for the toxicity test against brine shrimp larvae of A.salina Leach (BSLT). The mortality data of A.salina Leach was analysed using probit analysis to determine the value of LC 50 on each extract. The phytochemical assay and thin layer chromatography (TLC) was also apllied for both extracts. The result showed that both ethanol and n-hexane exctracts performed toxicity level against brine shrimp larvae of A.salina Leach with LC 50 less than 1000 ppm.The n-hexane extract indicated LC 50 of 189.093 ppm, higher than the ethanol extract, 286.031 ppm. The presence of steroids were detected in both extract whilst the alkaloids were only detected in the ethanol extract. Further separation of the n -hexane exctrat using TLC in n-hexane;ethyl acetate;amonia (66:33:0.8) yielded 5 spots with the Rf of 0.3;0.44;0.54;0.72 and 0.78, respectively. Kaemferia galanga L. rhizome is a traditional crop which is widely used for herbal medicine. This research is started from the extraction of maceration using ethanol 80% and partition with chloroform:water. 80 % ethanol extract and chloroform fraction tested antioxidant activity with DPPH (1,1-diphenyl-2-picrylhydrazyl), phytochemicals with reagents, separation of the active compound by TLC (thin layer chromatography) analytical and preparative TLC, the identification of compounds with sliding reagent using UV-Vis. The test results on the antioxidant activity of 80% ethanol extract of 64,93% (100 ppm) and chloroform fraction of 54,9 % (100 ppm) with indigo IC50 of 13,07 mg/mL and 81,9 mg mL. 80 % ethanol extract and chloroform fraction has potential as a source of natural antioxidants. Phytochemical test showed 80% ethanol extract contains flavonoids, alkaloids and tannins. At the chloroform fraction extract containing flavonoids. The separation of the active compounds with analytical TLC using eluent PE: ethyl acetate (5: 1) resulted in 7 stain (Rf 0,06 to 0,96) and preparative TLC produce 8 spots (Rf from 0,19 to 0,83). Results from UV-Vis spectrum can be presumed that such a class of flavonoid compounds flavanones or dihidroflavonol. It can be seen from the wavelength range 210-285 nm in the bands I and 300-550 nm in band II. The addition of the reagent slide showed the presence of hydroxyl groups in ring A, B and C and O-hydroxy on ring A. AbstrakKencur merupakan tanaman tradisional yang banyak dimanfaatkan sebagai obat alami.

Extraction, Toxicity Assay and Identification ofActive Compounds of Red Algae Extract (Eucheuma cottonii) from Tanjung Sumenep Madura was analysed. Extraction of active compound was conducted by maceration method using methanol as a solvent. The methanol extract was hydrolyzed with HCl 2 N. The result was partitioned using five solvents (1-butanol, ethyl acetate, chloroform, petroleum ether and n-hexane). Toxicity level was determined by Brine Shrimp Lethality Test (BSLT). The identification of the active compound was analyzed by phytochemical test, qualitatively. The result showed that the 1-butanol is the best solvent to extract active compounds of red algae E. Cottonii in partition process. 1-Butanol fraction gives the highest activity of toxicity with LC5070,32 ppm. LC50 value for ethyl acetate, chloroform, petroleum ether and n-hexane was 143,43; 194,40; 195,28; 303,66; and 634,97 ppm, respectively. Identification of active compounds indicated that 1-butanol fraction contains steroids and alkaloids.

Keywords: Eucheuma cottonii, extraction, Brine Shrimp Lethality Test, phythochemical
