In a randomized comparison of nevirapine or abacavir with zidovudine plus lamivudine, routine viral load monitoring was not performed, yet 27% of individuals with viral failure at week 48 experienced resuppression by week 96 without switching. This supports World Health Organization recommendations that suspected viral failure should trigger adherence counseling and repeat measurement before a treatment switch is considered.
Zidovudine/lamivudine/tenofovir has good virological efficacy in advanced HIV disease. In this population, who were infected with HIV-1 subtypes A, C or D, M184V with or without NAMs was the most common route to resistance, whereas K65R was identified less often.
SUMMARYThis study aimed to determine the occurrence of the etiologic agents of subclinical mastitis in Santa Inês and Morada Nova ewes and their susceptibility to developing the disease when submitted to the same management conditions. We analyzed 250 mammary glands of 130 Santa Inês ewes and 143 mammary glands of 77 Morada Nova ewes. The California Mastitis Test, somatic cell counts and microbiological analysis were performed at the moment of drying off. The occurrences of subclinical mastitis in different breeds were analyzed using the chi-square test by adjusting the values according to the Yates continuity correction. RESUMOFoi avaliada a resposta sorológica induzida pela vacinação com a dose padrão de 60-120x10 9 organismos da cepa B19 de Brucella abortus em bezerras Tabapuãs, entre 3 e 8 meses de idade, pelos testes sorológicos preconizados pelo Programa Nacional de Controle e Erradicação da Brucelose e Tuberculose (PNCEBT) para diagnóstico da brucelose bovina. Foram examinadas amostras de soro sanguíneo de 72 animais em quatro provas sorológicas: antígeno acidificado tamponado (AAT), teste de soroaglutinação lenta juntamente com o teste de 2-mercaptoetanol (2-ME), reação de fixação de complemento (RFC) e teste de polarização fluorescente (TPF), o qual foi interpretado de duas formas: ponto de corte variável e ponto de corte fixo. As amostras de soro sanguíneo foram colhidas imediatamente antes da vacinação, e após 30, 60, 90, 120, 150, 180, 210, 240, 270, 300, 330 e 360 dias. Foi calculada a especificidade dos testes e os resultados foram comparados pelo indicador kappa. O TPF apresentou especificidade mais elevada do que os outros testes nas amostras colhidas entre 30 e 90 dias após a vacinação. Aos 270 dias de vacinação, observou-se especificidade de 93,74% no AAT, no 2-ME e na RFC. No TPF com ponto de corte variável a especificidade foi 87,5% e com ponto de corte fixo foi 100,0%. O TPF apresentou maior capacidade de discriminação dos títulos de anticorpos vacinais logo após a vacinação, quando comparado com os outros testes preconizados pelo PNCEBT, mostrando-se um teste útil para o diagnóstico da brucelose bovina. SUMMARYThe study aimed to analyze the antibody response induced by vaccination with the standard dose of 60-120x10 9 organisms of Brucella abortus strain 19 in Tabapuã heifers, aged between 3 and 8 months, by serological tests recommended by the National Program for Brucellosis and Tuberculosis Control and Eradication (PNCEBT) for the diagnosis of bovine brucellosis. Serum samples of 72 animals were examined by four serological tests: rose Bengal test (RBT), standard agglutination test with the 2-mercaptoetanol (2-ME) test, complement fixation test (CFT) and fluorescence polarization assay (FPA), which was interpreted in two ways: variable cutoff and fixed cutoff. Heifers were bled immediately prior to vaccination, and after 30, 60, 90, 120, 150, 180, 210, 240, 270, 300, 330 and 360 days. Specificity of the tests was calculated, and the results were compared using the kappa statistic. FPA had a higher specificity than the other tests on the samples collected from 30 to 90 days after vaccination, but over time this difference decreased. At 270 days of vaccination, the specificity of RBT, 2-ME and RFC was 93.74%. The specificity of the FPA with variable cutoff was 87.5%, and with fixed cutoff was 100.0%. Abstract:The objective of this study was to confirm the occurrence of swine brucellosis by isolating Brucella suis from an outbreak that occurred in December 2012 in a Certified Pig Breeding Farm (Granja de Reprodutores Suínos Certificada, GRSC), in Guariba, São Paulo, Brazil. To this end, we collected the livers and spleens of seven stillborn piglets born to six different sows that had aborted. Furthermore, the stomach content from one piglet was also collected. The organs were prepared using sterile materials and seeded in Petri dishes containing Brucella agar with 5% rabbit serum medium. The routine tests confirmed that the B. suis biovar 1 strain was the causative agent. Therefore, the results showed that despite the reduced incidence of swine brucellosis caused by Brucella suis in the country, the causative agent of the disease is still present, able to reach herds with higher biosafety levels, threatening human and animal health.Keywords: Isolation, Brucellosis, Swine.Resumo: O objetivo deste trabalho foi confirmar a ocorrência de brucelose suína por meio do isolamento de Brucella suis de um foco ocorrido em dezembro de 2012 em uma Granja de Reprodutores Suínos Certificada (GRSC), localizada no município de Guariba, São Paulo, Brasil. Para isso, foram colhidos fígados e baços de sete leitões natimortos, filhos de seis matrizes diferentes, que apresentaram aborto. Além disso, foi colhido o conteúdo estomacal de um dos leitões. Os órgãos foram dilacerados, usando materiais estéreis, e semeados em placas de Petri com meio Brucella ágar com 5% de soro de coelho. A identificação foi confirmada por provas de rotina, e que levaram à conclusão de que se tratava de uma cepa de B.suis da biovariedade 1. Nesse sentido, observa-se que, apesar da redução da incidência de brucelose suína, por Brucella suis, no país, o agente etiológico da doença ainda está presente, atingindo até mesmo rebanhos com nível de biossegurança mais elevado e demonstrando ser um risco tanto à saúde humana quanto à saúde animal.Palavras-chave: Isolamento, Brucelose, Suinocultura. _____________________________ Autor para correspondência. Diversos problemas sanitários são relevantes na criação de ovinos. Existe uma carência de estudos que abordem o diagnóstico da mastite ovina, especialmente das raças de aptidão para a produção de carne. Este trabalho objetivou determinar a ocorrência dos agentes etiológicos da mastite subclínica em ovelhas das raças Santa Inês e Morada Nova no momento da secagem e sua susceptibilidade em desenvolver a doença quando submetidas às mesmas condições de manejo.
SUMMARYThe aim of this work is to determine the endemic level of reproductive losses and positivity for four infectious agents related to reproductive problems in dairy cattle of a farm in São Carlos, SP, Brazil. Blood samples were collected from 142 animals at reproductive age, older than two years, 21.1% of which had a history of abortions or stillbirths in at least one pregnancy. Immunofluorescent assay test (IFAT), buffered acidified antigen test (BAAT), serum neutralization technique (SNT) and microscopic agglutination test (MAT) were used for the detection of anti-Neospora caninum, antiBrucella abortus, anti-Bovine Herpesvirus Type 1 (anti-BHV-1) and anti-Leptospira spp., respectively. The serological tests conducted showed that 28.9% of the animals had titers greater than or equal to 100 for anti-Neospora caninum. Viral neutralization tests demonstrated that 26.8% of the animals had titers greater or equal to 256 for anti-BHV-1. Only 7.7% of the studied animals had titers equal to or greater than 100 for Leptospira spp. in the microscopic agglutination test. Furthermore, no positive reactions were observed for the buffered acidified antigen test for the diagnosis of bovine brucellosis. The results suggest the presence of BHV-1, N. caninum and Leptospira spp. in contact with dairy cattle in the experimental farm, which could be responsible for the occurrence of abortions and stillbirths in the herd. Reproductive losses are endemic, thus implying the possible presence of chronic infections caused by these microorganisms. KEY-WORDS:Cattle. Etiological agents. Reproduction. RESUMOEste trabalho objetivou traçar o nível endêmico de perdas reprodutivas e determinar a positividade para quatro agentes infecciosos relacionados com problemas reprodutivos em um rebanho bovino leiteiro no município de São Carlos-SP. Foram coletadas amostras de soro sanguíneo de 142 animais em reprodução com idades acima de dois anos, dos quais 21,1% apresentaram casos de abortamentos ou natimortos em pelo menos uma gestação. Utilizaram-se os testes sorológicos de imunofluorescência indireta, teste do antígeno acidificado tamponado, virusneutralização e soroaglutinação microscópica, virusneutralizaçãopara detecção de anticorpos anti-Neospora caninum, anti-Brucella abortus, anti-Herpesvírus Bovino Tipo-1(BoHV-1) e anti-Leptospira spp., respectivamente. Os exames sorológicos realizados demonstraram que 28,9% dos animais apresentaram títulos superiores ou iguais a 100 de anticorpos antiNeospora caninum. Os exames de virusneutralização demonstraram que 26,8% dos animais apresentaram títulos maiores ou iguais a 256 para anticorpos anti-BoHV-1.
