This research was undertaken to study the influence of different concentrations of the MT medium, sucrose, vitamins, activated charcoal and gibberellic acid (GA3) on the culture of immature embryos from the crossing between 'Pêra Rio' sweet orange and 'PONCÃ' mandarin. The embryos were excised under aseptic conditions and inoculated in 15 mL of the MT medium according to the following experiments: 1) MT concentrations (0%, 50%, 100%, 150% and 200%) supplemented with 0, 30, 60 and 90 g.L-1 of sucrose; 2) vitamins concentrations of the MT (0%, 50%, 100%, 150% and 200%) supplemented with 0, 30, 60 and 90 g.L-1 of sucrose; 3) activated charcoal concentrations (0, 0.5, 1, 1.5 and 2 g.L-1) supplemented with GA3 (0, 0.01, 0.1; 1 and 10 mg.L-1). After the inoculation, the embryos were kept in a growth room for 90 days at 27 ± 1ºC, in a 16-hour photoperiod with 32 µmol.m-2.s-1 of irradiance. The best development of embryos at the globular stage was achieved using 50% and 100% of the MT medium plus 60 g.L-1 and 90 g.L-1 of sucrose, respectively, supplemented with 0.01 mg.L-1 of GA3. The addition of activated charcoal or vitamins in the MT medium has shown to be unnecessary to the development of globular embryos.
O objetivo do trabalho é analisar diálogos permitidos pelo filme Wall-E para compreensão da Educação Ambiental Crítica (EAC) a partir de uma prática educativa realizada com alunos de licenciatura em Ciências Biológicas da UFLA. Após exibição do filme foi realizada uma discussão, em que os discentes compartilharam suas respostas referentes à compreensão da EAC presente no filme. As falas foram categorizadas via Método da Análise de Conteúdo. Concluímos pelas categorias formuladas que a obra ajudou para construção de saberes e reflexões sobre temáticas contemporâneas as quais o filme fala abertamente ou por suas lacunas. Consideramos, ainda, a importância das animações, pois atuam como frestas para compreensão da realidade de modo lúdico e instigante, podendo ser aplicados em aula para problematização de questões ambientais.Palavras Chave: Cinema. RESUMO -Objetivou-se verificar qual o melhor estádio embrionário para o cultivo de embriões imaturos oriundos de frutos provenientes de hibridação entre 'Pêra Rio' x 'Poncã', bem como o efeito de diferentes concentrações do meio de cultura MT. Os embriões em diferentes estádios de desenvolvimento (globulares, torpedo e cordiforme) foram excisados e inoculados em tubos de ensaio contendo 15 mL do meio MT com diferentes concentrações (0; 50; 100 e 150% da composição original e acrescido de 50 g.L -1 de sacarose). Após a inoculação, os embriões foram incubados à 27±1ºC, fotoperíodo de 16 horas e irradiância de 32 µmol.m -2 .s -1 . Após 90 dias, avaliou-se o comprimento da parte aérea e do sistema radicular, massa fresca e número de folhas das plântulas. Melhor desenvolvimento dos embriões imaturos foi obtido em estádio cotiledonar e com a concentração de 150% do meio MT. Embryos in globular, torped and cotiledonary stage were excised and inoculated individually in test tubes with 15 mL in distinct MT medium concentrations (0; 50; 100 and 150% of original composition and added sucrose (50 g.L -1 ). After inoculation, the embryos were maintained in growth room at 27±1ºC, 16 hour photoperiod and 32 umol.m -2 .s -1 irradiance. After 90 days, the length of the aerial part and roots system, fresh mass and number of leaves, were evaluated. The best results were obtained by incubating embryos in the cotiledonary stage, in MT medium with concentrations of 150%.
A formação inicial de professores é um processo de muita reflexão e diálogo, pois, além de terem que se ater às questões burocráticas com relação ao currículo, é preciso que estes façam uma relação entre os conhecimentos curriculares e as questões que envolvem a realidade dos alunos de maneira transversal. Nessa perspectiva, tendo em vista que vivemos em uma sociedade marcada por uma grande multiculturalidade, os professores precisam realizar um resgate histórico-cultural. Além disso, é possível problematizar estas questões tomando o meio ambiente como ponto de partida, caracterizando-o e percebendo suas relações. Nesse sentido, o objetivo deste trabalho é relatar e analisar uma prática educativa para o ensino de bioma utilizando o poema "O Uraguai" e suas contribuições para a formação inicial de professores. Como metodologia de análise dos dados foi utilizada a análise de categoria. A partir da categorização dos dados foi possível elencar duas categorias, sendo a primeira "Curiosidade e Imaginação instigadas por meio dos recursos utilizados", e a segunda "Aula dinâmica". Os resultados mostraram que a utilização dos recursos metodológicos, poema, desenhos e fotografias, foram essenciais para instigar a curiosidade e imaginação dos mesmos para o desenvolvimento do conteúdo. Além disso, foi possível perceber que essas ferramentas utilizadas proporcionaram uma aprendizagem mais lúdica, desta forma os alunos consideraram que a aula teve um caráter dinâmico, pois não ficou centrada na fala do professor e instigou a todo o momento que os estudantes contribuíssem com seus conhecimentos prévios e suas reflexões sobre o que estava sendo construído.
