This research was conducted to evaluate the motility of Bali cattle spermatozoa stored at 5°C. Bali cattle was given three different types of ration, those are PI: 40% field grass (RL) + 60% concentrate, P2: 40% RL + 60% concentrate supplemented by 5% soybean calcium oil soap, and P3: 40% RL + 60% of the concentrate which was supplemented by 5% COS + 10% cashew fruit flour (CFF). The variables measured were the motility of spermatozoa stored with Egg Yolk Tris diluent at 5°C and observed for 3 (three) days. This study used a completely randomized design (CRD) with 3 treatments and 3 replications. The data of the percentage of motility analyzed using one-way analysis of variance (ANOVA). If there are significant differences (P <0.05) or very significant (P <0.01), then the Tukey-W-Procedure test with SPSS is continued. Bali cattle consuming P3 rations had higher motility (P <0.05) than P1 and P2. Feeding of 5% soybean oil calcium soap (SOCS) and 10% cashew fruit flour CFF had a significant influence on the motility of Bali cattle.
(RPH) Kota Ternate Menggunakan Kafein", bertujuan untuk mengkaji pengaruh penambahan kafein terhadap motilitas spermatozoa kauda epididimis sapi Bali dan mengukur berapa jumlah penambahan kafein yang dapat meningkatkan motilitas spermatozoa kauda epididimis sapi Bali. Koleksi testes diambil dari Rumah Potong Hewan (RPH) Kota Ternate, proses analisis sperma dilakukan di laboratorium Bioteknologi Universitas Khairun. Metode penelitian ini adalah sperma kauda epididimis dimasukkan dalam pengencer tris kuning telur kemudian diberi perlakuan penambahan kafein T0 (0 mg/ml), T2 (2 mg/ml), T4 (4 mg/ml), dan T6 (6 mg/ml) dengan 4 kali ulangan. Parameter dianalisis adalah kualitas sperma secara makroskopis dan mikroskopis. Data yang diperoleh dianalisis dengan analisis variance (ANOVA) satu arah, apabila terdapat perbedaan yang nyata (P<0,05) atau sangat nyata (P<0,01), dilanjutkan uji Tukey-W-Procedure dengan SPSS 18. Hasil penelitian menunjukkan bahwa penambahan kafein 2-4 mg/ml pada pengencer tris kuning telur sperma kauda epididymis sapi Bali mampu meningkatkan motilitas sperma.
This study aims to determine the effect of different concentrations of Yellow Tris Egg Duck on the quality of sheep spermatozoa. The method used for cement shelter with an artificial vagina is then diluted with Tris Yellow Duck Egg at different concentrations from 10%, 20%, 30%, 40%, 50% and stored at 5 °C. The quality parameters of spermatozoa observed were motility, morphology, and viability of spermatozoa up to 2 days. The design used was a one-way complete randomized design (RAL) and continued with the Tukey-W-Procedure test with SPSS 16. The results showed concentrations of 50% Tris duck egg yolks have hight quality spermatozoa with motility 76.
The research aimed to examine the effect of caffeine's addition on diluents on Bali cattle epididymis' survival or sperm viability to get the right amount of caffeine. The epididymis collection was taken from the Slaughterhouse of Ternate City. Analysis of sperm vitality or viability was carried out in the Biotechnology Laboratory of Universitas Khairun. The methods in this research were by preparing egg yolk Tris solution treated with caffeine P0 (0 mg/ml), P2 (2 mg/ml), P4 (4 mg/ml), and P6 (6 mg/ml) with four replications, and then the epididymal sperm was inserted into each treatment. The epididymal sperm viability was calculated by evaluating the percentage of the number of living sperm. It compared to the dead sperm, which were observed microscopically by eosin-nigrosin staining. The data obtained were analyzed by one-way analysis of variance (ANOVA) to see if there were significant differences (P<0.05) or very real (P<0.01), the analysis were then continued with the Tukey-W-Procedure test with SPSS 18. The results show that administration of caffeine 2 mg/ml and 4 mg/ml (86.95% and 86.95%) have a significant effect on the sperm viability of epididymis compared with 0 mg/ml and 6 mg/ml (69.38% and 69.91%) treatments.
