The influence of glutathione on the prooxidant-antioxidant balance in rats' kidneys and blood under the conditions of rhabdomyolytic model of acute kidney injury was studied in the experiment. It is observed that intraperitoneal administration of glutathioneіn dose 30 mg/kg normalizes the prooxidant-antioxidant balance, confirmed by the reduction of lipid and protein peroxidation intensity, normalization of catalase and glutathione peroxidase activity, SH-groups аndceruloplasmin content.
Вплив статинів на розвиток ниркової недостатності у білих щурів В експерименті на щурах досліджена дія трьох статинів (аторвастатин, ловастатин, симвастатин) на функцію нирок за умов гострої ниркової недостатності (ГНН) і встановлено ефективні дози, у яких вони найбільш виражено виявляють нефропротекторні властивості. Виявлено, що всі статини у різних дозах викликали відновлення функціональної активності нирок на тлі змодельованої рабдоміолітичної ГНН, проте у дозі 20 мг/кг відзначали найбільш істотне покращення основних показників функцій нирок: посилення діурезу на 32 %, швидкості клубочкової фільтрації на 90 %, зменшення протеїнурії більше ніж удвічі. При цьому активність креатинфосфокінази плазми крові тварин була всього на 14 % вища за значення інтактного контролю. Водночас у тварин із модельною патологією без застосування препаратів цей показник підвищувався на 141 %, що вказує на незначну міотропну активність статинів у обраному режимі введення. Отже, застосування дози 20 мг/кг є найбільш обґрунтованим з точки зору нефропротекторної ефективності та безпеки. Мета роботи. Вивчення процесу ознайомлення студентів І курсу фармацевтичних факультетів з майбутньою спеціальністю у вищому навчальному закладі (ВНЗ) порівняно з аналогічним курсом Боннського університету (Німеччина).Матеріали і методи. Використано нормативно-правову базу, що регулює освітній процес в Україні та Німеччині: проведено аналіз організації підготовки студентів фармацевтичних факультетів. Для порівняльного аналізу обрано фармацевтичний факультет Боннського університету (Німеччина).Результати й обговорення. Встановлено, що відмінності у дисциплінах, які дають можливість ознайомити студентів-магістрів із майбутньою спеціальністю, є незначними, а саме більше теоретичного матеріалу, що можна пояснити відносно молодим періодом становлення фармації в Україні. Перевагою у підготовці вітчизняних студентів є проходження практичної підготовки на базі навчально-виробничої аптеки, що сприяє адаптації майбутніх випускників на робочому місці.Висновки. Дисципліна «Вступ у фармацію» забезпечує ознайомлення першокурсників фармацевтичних факультетів з особливостями фармацевтичної справи аналогічно як і в європейському університеті м. Бонн (Німеччина) під час вивчення курсу «Arzneimittelformenlehre».
The effect of derivatives of sulfur-containing amino acids (ademetionine, taurine and glutathione) on survival of animals with acute kidney injury of various genesis Aim. To study the nephroprotective effect of sulfur-containing amino acids (ademetionine, taurine and glutathione) on different models of acute kidney injury (AKI) by the integral index of the survival rate of animals. Materials and methods. The experiments were performed on 105 non-linear mature white laboratory rats and 35 mice. The drugs studied-ademetionine (Geptral, Abbott SpA, Italy) in the dose of 20 mg/kg, glutathione (TAD 600, Biomedica Foscama, Italy) in the dose of 30 mg/kg and taurine (Sigma-Aldrich, USA) in the dose of dose 100 mg/kgwere injected intraperitoneally 3-7 days before AKI modeling. Rhabdomyolysis-induced AKI was modeled by the intramuscular injection of 50 % glycerol solution in the dose of 10 ml/kg, toxic AKI-by the injection of gentamicin solution in the dose of 80 mg/kg and the subcutaneous injection of ethylene glycol in the dose of 10 ml/kg. Simulation of renal ischemia was performed under general anesthesia (ethaminal sodium, 40 mg/kg) by clamping of both renal pedicles for a period of 60 min, followed by reperfusion for 24 h. Results and discusion. The experimental results show a significant improvement in the survival of animals under the effect of the drugs of derivatives of sulfur-containing amino acids studied in conditions of various models of AKI. Ademetionine, taurine and glutathione reduced the lethality of animals on models of rhabdomyolytic, gentamicin, ischemia-reperfusion AKI. In conditions of ethylene glycol intoxication the highest nephroprotective activity by the survival criterion was shown by glutathione (57.1 % survival at 48 h after administration of ethylene glycol against 100 % of mortality in the control group (p < 0.05). Conclusions. All derivatives of sulfur-containing amino acids studied (ademetionine, taurine and glutathione) exhibit the nephroprotective activity by the criterion of survival of animals on various models of AKI. Glutathione demonstrates the best survival of animals with acute kidney injury of various genesis, while ademetionine has the lowest survival.
Renal effects of the sulfur-containing aminoacid derivatives (ademetionine, taurin and glutathion) in conditionally healthy animals Derivatives of sulfur-containing aminoacids (ademetionin, taurine and glutathione) selected for the experimental studies are used in various fields of practical medicine due to their multifaceted pleiotropic effects. The study of their renal effects in the conditions of the physiological norm makes it possible to expand and supplement their pharmacological characteristics, and forms the basis for further study of the nephroprotective potential in renal pathology. Aim. To study the effect of sulfur-containing amino acid derivatives (ademetionine, taurine and glutathione) on the morphofunctional state of the rat kidneys in the daily administration for 7 days. Materials and methods. The experiments were performed on mature non-linear white rats weighing 130-180 g. Animals were divided into 4 groups (n = 7): group I-intact control, group II-animals which were given ademetionine ("Geptral", "Abbott SpA", Italy) in the dose of 20 mg/kg, group III-animals received taurine ("Sigma-Aldrich", USA) in the dose of 100 mg/kg, group IV-animals received glutathione ("TAD 600", "Biomedica Foscama", Italy) in the dose of 30 mg/kg. All drugs were injected intramuscularly for 7 days. Results. The study of the renal effects of the sulfur-containing amino acid derivatives (SAD) under research in the daily administration for 7 days to conditionally healthy animals showed that these drugs had a weak diuretic effect, which was probably due to a decreased tubular reabsorption of water without significant changes in the glomerular filtration rate. Besides, the use of ademetionine resulted in a slight reduction of azotemia. There was not any significant change in the acid regulatory kidney function under the effect of SAD. Administration of ademetionine, taurine and glutathione resulted in a significant decrease in proteinuria, which was probably caused by the effect on the processes of protein reabsorption. The effect on the ion regulatory kidney function upon the course of SAD administration was characterized by an increase in urinary sodium excretion against the background of decreased relative sodium reabsorption, which was accompanied by intensification of distal transport of sodium ions due to the activation of the tubular-tubular balance. The morphological examination did not reveal any histopathological changes in the renal tissue, confirming the absence of nephrotoxicity of the SAD studied. Conclusions. According to the data obtained the 7-day administration of ademetionine, taurine and glutathione to conditionally healthy animals moderately affects the processes of glomerular filtration and tubular transport in nephrons, which results in a slight increase in diuresis along with preservation of the renal mechanisms of autoregulation and the absence of histopathological changes in the kidneys.
