The experiment on white rats was targeted at the examination of influence of statins (atorvastatin, lovastatin, simvastatin) under the conditions of acute renal failure, caused by rhabdomyolysis. Renoprotec tive effects of statins were demonstrated by reduction of hyperazotemia and proteinuria and improvement of renal excretory function, which correlated with antioxidant properties of drugs.
Вплив статинів на розвиток ниркової недостатності у білих щурів В експерименті на щурах досліджена дія трьох статинів (аторвастатин, ловастатин, симвастатин) на функцію нирок за умов гострої ниркової недостатності (ГНН) і встановлено ефективні дози, у яких вони найбільш виражено виявляють нефропротекторні властивості. Виявлено, що всі статини у різних дозах викликали відновлення функціональної активності нирок на тлі змодельованої рабдоміолітичної ГНН, проте у дозі 20 мг/кг відзначали найбільш істотне покращення основних показників функцій нирок: посилення діурезу на 32 %, швидкості клубочкової фільтрації на 90 %, зменшення протеїнурії більше ніж удвічі. При цьому активність креатинфосфокінази плазми крові тварин була всього на 14 % вища за значення інтактного контролю. Водночас у тварин із модельною патологією без застосування препаратів цей показник підвищувався на 141 %, що вказує на незначну міотропну активність статинів у обраному режимі введення. Отже, застосування дози 20 мг/кг є найбільш обґрунтованим з точки зору нефропротекторної ефективності та безпеки. Вищий державний навчальний заклад України «Буковинський державний медичний університет»Вплив нанохрому цитрату на осмотичну резистентність мембран еритроцитів крові щурів з експериментальним цукровим діабетом Цукровий діабет (ЦД) -серйозна медико-соціальна проблема, оскільки характеризується значним поширен-ням у світі та сталою тенденцією до збільшення кількості хворих. Мета роботи -дослідити вплив нанохрому цитрату на осмотичну резистентність мембран еритроцитів крові тварин при цукровому діабеті. Матеріали та методи. Робота виконана на 18-міс. лабораторних щурах-самцях. Цукровий діабет викликали дексаметазоном (0,125 мг/кг, п/шк,14 діб). Нанохрому цитрат (НХЦ) вводили в шлунок (0,01 мг/кг, 14 діб). Проникність еритроцитарних мембран (осмотична стійкість еритроцитів) визначали за стійкістю еритро-цитів до суміші ізотонічних розчинів сечовини і натрію хлориду з їх різним об'ємним вмістом. Ступінь осмо-тичного гемолізу (%) вимірювали фотоколориметрично по відношенню до оптичної щільності еталонного розчину сечовини. Осмотичну резистентність мембран еритроцитів (ОРМЕ) -(% гемолізованих клітин) по-рівнювали з аналогічним показником у контрольних інтактних тварин. Рівень глюкози крові визначали за допомогою глюкометра. Результати. У тварин з експериментальним ЦД поряд зі збільшенням рівня глюкози в крові до 14,2 ± 0,4 ммоль/л відсоток гемолізованих еритроцитів порівняно з контролем зріс у 8,2-2,3 рази відповідно до збільшення кон-центрації сечовини в розчині. Підвищення проникності еритроцитарних мембран, що призвело до зниження осмотичної стійкості еритроцитів та їх гемолізу, свідчить про активацію ПОЛ і зниження активності систем анти-оксидантного захисту у тварин з ЦД. Уведення НХЦ при ЦД зменшило вміст глюкози в крові до 6,6±0,4 ммоль/л і значно підвищило ОРМЕ, що свідчить про наявність у НХЦ мембранопротекторних властивостей. Висновки. При експериментальному (дексаметазоновому) ЦД знижується осмотична резистентність мембран еритроцитів, активуються деструктивні процеси, що призводить до їх прискореного руйнування. НХЦ змен-шує рівень глюкози в крові тварин з ЦД і підвищує осмотичну резистентність мембран еритроцитів. Ключові слова: мембрани еритроцитів; нанохрому цитрат; експериментальний цукровий діабет
