Both dilutions were able to potentiate the decrease of most of cytokines and chemokines induced by the parasite infection in vitro, which explains the clinical improvement seen previously in vivo, however, the mechanisms involved and the epidemiological significance of these findings are still under discussion.
ABSTRACT. - Bronchopneumonia is important for world livestock production and one of the major causes of calf mortality during the first months of life. The preventive and therapeutic measures adopted for the disease in calves in Brazil are usually based on the results of international studies; however there is not enough known which bacteria are implicated. In the first month of life calves show immaturity of their immune system, what has been little studied in regard to pneumonia. The present investigation sought to study neonate bronchopneumonia in calves, to identify which bacteria are present in the respiratory tract of healthy and, with naturally acquired pneumonia calves, to analyze the pulmonary cytological response against pathogens. For this purpose, samples of the respiratory tract were collected by tracheocenthesis. It was noted that the microflora of the tracheobronchial region of healthy and diseased calves was the same, but they were different from the one reported by international papers. The flora was constituted mainly by Staphylococcus sp., Bacillus sp., Streptococcus sp., Pseudomonas aeruginosa and enterobacteriae, allowing to infer that the prophylactic and therapeutic measures adopted internationally may not be as effective for the Brazilian condition. It was also found that newborn calves have an approximate ratio of 1:1 of macrophages and neutrophils in the tracheobronchial region when they were healthy, reaching a ratio of approximately 1:3 of macrophages and neutrophils when they were with bronchopneumonia. Probably, these profiles are characteristic of the age, a period when exists immaturity of the immune system and influenced by management factors which lead to greater inhalation of bacterial agents. Infectious bovine mastitis is responsible for serious economic losses in dairy cattle breeding. Information about the damages caused by micro-organisms is of great interest and importance. The purpose of the present study was to report on the microbiological and histopathological aspects of the mammary parenchymas of slaughtered dairy cows. A total of 184 mammary glands were examined and samples of mammary parenchyma were collected for microbiological and histopathological examinations. Micro-organisms were isolated from 69.6% samples; 23 (12.5%) of the 184 samples did not show histological changes; inflammatory response was observed in 56 (30.4%) samples; inflammatory response and repair were present in 82 (44.6%) samples; repair process was verified in 23 (12.5%) of the mammary glands. Coagulase-negative Staphylococci were observed in 53.8% of the samples followed by coagulase-positive Staphylococci (7.6%), Prototheca sp. (2.2%) and Streptococcus dysgalactiae (1.6%). Coagulase-negative Staphylococci and coagulase-positive Staphylococci were associated mainly to chronic inflammatory response and chronic inflammatory response and repair. Samples from which no micro-organisms were isolated (n = 56) had no histological changes in 82.6% of the cases. These results were higher (P < 0.05) when compared to the samples with micro-organisms and without histological changes (17.4%).
The aim of this study was to develop and evaluate a real-time quantitative PCR (qPCR)-based method to detect and quantify Staphylococcus aureus in bronopol-preserved milk samples from subclinical intramammary infections (IMI). Serial dilutions of milk artificially inoculated with Staph. aureus ATCC 29213 were used to establish a standard curve (cfu/mL) of the qPCR assay targeting the Staph. aureus thermonuclease-encoding gene nuc according to the strain plate count. The analytical sensitivity, specificity, and repeatability of the qPCR assay were determined. A total of 60 milk samples, collected from mammary quarters without abnormal appearance and with positive isolation of Staph. aureus, were submitted to both the qPCR protocol and Staph. aureus plate counting and results from both methods were compared. Staphylococcus aureus from bronopol-preserved, subclinical IMI milk samples were not accurately enumerated by qPCR compared with plate counting of the nonpreserved, raw milk sample. The detection limit of the qPCR protocol of inoculated Staph. aureus ATCC 29213 in bronopol-preserved milk samples was 1.04 × 10(1) cfu/mL. The qPCR protocol can be a high-throughput and rapid diagnostic assay to accurately detect Staph. aureus IMI from bronopol-preserved milk samples compared with a traditional culturing method. However, the proposed qPCR protocol is not accurate for counting of Staph. aureus in bronopol-preserved milk samples from naturally infected mammary glands.
The presence of yeasts in milk may cause physical and chemical changes limiting the durability and compromising the quality of the product. Moreover, milk and dairy products contaminated by yeasts may be a potential means of transmission of these microorganisms to man and animals causing several kinds of infections. This study aimed to determine whether different species of yeasts isolated from bovine raw milk had the ability to develop at 37°C and/or under refrigeration temperature. Proteinase and phospholipase activities resulting from these yeasts were also monitored at different temperatures. Five genera of yeasts (Aureobasidium sp., Candida spp., Geotrichum spp., Trichosporon spp. and Rhodotorula spp.) isolated from bovine raw milk samples were evaluated. All strains showed one or a combination of characteristics: growth at 37°C (99·09% of the strains), psychrotrophic behaviour (50·9%), proteinase production (16·81% of the strains at 37°C and 4·09% under refrigeration) and phospholipase production (36·36% of the isolates at 37°C and 10·9% under refrigeration), and all these factors may compromise the quality of the product. Proteinase production was similar for strains incubated at 37°C (16·81% of the isolates) and room temperature (17·27%) but there was less amount of phospholipase-producing strains at room temperature (15·45% of the isolates were positive) when compared with incubation at 37°C (36·36%). Enzymes production at 37°C by yeasts isolated from milk confirmed their pathogenic potential. The refrigeration temperature was found to be most efficient to inhibit enzymes production and consequently ensure better quality of milk. The viability of yeasts and the activity of their enzymes at different temperatures are worrying because this can compromise the quality of dairy products at all stages of production and/or storage, and represent a risk to the consumer.
Mastitis is an inflammation of the mammary glands and in most cases it is caused by the presence of microorganisms. High mastitis rates in dairy cattle herds can cause an increase in total microorganism counts of bulk tank milk. The present paper was aimed at verifying whether the occurrence of mastitis in dairy cattle herds is reflected in raw-milk indicators of hygienic-sanitary quality. To observe the correlation among the analysed variables, we performed a logarithmical transformation (log10) of different indicator counts of raw milk and compared them with the occurrence of mastitis in dairy cattle herds. Few correlations were observed among mastitis cases in dairy cattle herds and the raw-milk indicators of hygienic-sanitary quality. We observed a negative correlation between the log10 of mesophilic aerobic plate counts and psychotropic aerobic plate counts when compared with the occurrence of no bacterial growth. The log10 of thermophilic aerobic plate counts and yeasts and mould aerobic plate counts presented a positive correlation with the cases of infectious mastitis and mastitis caused by Staphylococcus spp.
