Actividad antioxidante in vitro del extracto etanólico de Phoradendron bathyoryctum Eichler por el método de captura del radical libre 1,1-difenil-2-picril-hidrazilo (DPPH•). Este trabajo describe el análisis in vitro del potencial antioxidante que presenta el extracto etanólico de la especie Phoradendron bathyoryctum Eichler, evaluado a través del método de captura del radical orgánico 1,1difenil-2-picril-hidrazilo (DPPH•). Esta especie es utilizada como medicinal por la población paraguaya. La identificación botánica, así como el análisis morfoanatómico de la especie vegetal en estudio, fueron determinados en un trabajo anterior a éste por el equipo de investigación del autor principal del presente trabajo. Los ensayos in vitro demostraron que el extracto de Phoradendron bathyoryctum Eichler posee la capacidad de captar el radical libre DPPH• de forma análoga al ácido ascórbico, utilizado como patrón de referencia. Aunque el poder neutralizante del extracto frente al radical DPPH• haya sido 36 veces menor comparado con el compuesto de referencia, demostró su poder antioxidante, lo que otorga al vegetal un valor agregado en el momento de ser consumido como medicinal por la población paraguaya. La IC50 del extracto fue de 169 ± 1,0 µg frente a la IC50 del ácido ascórbico cuyo valor calculado fue de 4,69 ± 0,03 µg. La capacidad de 50 µg de extracto vegetal de neutralizar el radical libre DPPH• fue del 39,7 ± 1,99% y finalmente se determinó que cada gramo de extracto de Phoradendron bathyoryctum Eichler contiene un equivalente a 75.400 ± 3764 µg de ácido ascórbico. This species is used as a medicinal plant by the Paraguayan population. The botanical identification and the morphoanatomical analysis of the plant were determined in a previous work by the research team of the main author of this work. Thein vitro assays demonstrated the Phoradendron bathyoryctum Eichler extract's ability to capture the DPPH• free radical in a way analogous to ascorbic acid (AA), used as a standard for this study. Although the neutralizing power of the extract was 36 times lower than that of the standard used, its antioxidant powerwas still demonstrated, which adds value to it as a medicinal plant. The IC50 of the extract was 169 ± 1.0 µg vs. the IC50 of AA whose estimated value was 4.69 ± 0.03 µg. The capacity of 50 µg of plant extract to inhibit the free radical DPPH• was 39.7 ± 1.99%, and it was determined that each gram of extract of Phoradendron bathyoryctum Eichler contains an equivalent to 75,400 ± 3764 µg of ascorbic acid.
Extracción y caracterización físico-química del aceite esencial de Origanum syriacum L. extraído en diferentes tiempos. Se estudió la composición química del aceite esencial de Origanum syriacum L. -nueva variedad introducida en Paraguay-extraído a las 2, 4 y 6 horas del proceso respectivamente, por el método de destilación por arrastre de vapor de agua, a partir de hojas previamente desecadas. Se llevaron a cabo la identificación botánica y el análisis morfoanatómico de la especie vegetal en estudio con el propósito de comprobar su autenticidad, paso fundamental para el inicio de la investigación. La caracterización química del aceite esencial se realizó por cromatografía gaseosa acoplada a espectrometría de masas (GC-MS); la cual reveló que el aceite está constituido mayoritariamente por carvacrol en porcentajes de 19,2; 11,9 y 19,5% correspondientes a las 2, 4 y 6 horas de extracción, además de la variedad de otros compuestos en menor proporción, apareciendo algunos particulares en los diferentes periodos de extracción. Se determinaron además la densidad, el índice de refracción, el pH, la solubilidad y el espectro UV como parámetros adicionales de caracterización. El rendimiento del aceite esencial fue del 1,71% al final del proceso de extracción, expresado en base seca. Palabras Claves: Origanum syriacum, Aceite esencial, Carvacrol, destilación por arrastre de vapor de agua Extraction and physicochemical characterization of Origanum syriacum L. essential oil extracted at different times through steam distillation.This research studied the chemical composition of the essential oil of Origanum syriacum L. -a new variety introduced in Paraguay-extracted at 2, 4 and 6 hours of processing respectively, by the method of steam distillation from dried leaves. Botanical identification and morpho-anatomical analysis of the plant species under study were conducted in order to check its authenticity, an essential step in the initiation of the investigation. Gas chromatography mass spectrometry (GC-MS) revealed that the essential oil is mainly constituted by carvacrol in percentages of 19.2, 11.9 and 19.5% corresponding to the 2, 4 and 6 hours of extraction respectively; as well as a variety of other compounds in lower amounts, a few particular ones appearing in the different extraction periods. The density, refractive index, pH, solubility, and UV spectra were also determined as additional characterization parameters. The essential oil yield was 1.71% on a dry base.
Estudio y utilización de dos especies vegetales para la obtención de un medicamento piloto rico en minerales y omega-3. Se realizaron los estudios morfoanatómicos y micrográficos, de las especies vegetales Talinum paniculatum (Jacq) Gaertn y Medicago sativa L., con el fin de comprobar la autenticidad de las mismas, para ser utilizadas en la formulación de un medicamento piloto, que a la vez podría servir perfectamente como un suplemento nutricional, gracias al valioso contenido químico de ambas. En cada una de ellas se detecta la presencia de minerales como el sodio, potasio, manganeso, calcio, magnesio, hierro y zinc, indispensables para la homeostasis. Ensayos de identificación por cromatografía gaseosa acoplada a masas, delata la presencia de Omega-3, importante en la prevención de problemas cardiovasculares, contribuyendo eficazmente con la disminución de los niveles de triglicéridos y colesterol (Kris P., 2003). Los minerales mencionados con anterioridad, así como el Omega-3, contenidos de forma natural dentro de las células de las especies vegetales involucradas en esta investigación, son potenciados, al ser mezclados con resveratrol, que combate radicales libres agresivos para las células humanas, contribuyendo de esa manera a aumentar la longevidad de las mismas, retardando la vejez. Ensayos preliminares toxicológicos como el test de hemólisis y el bioensayo de letalidad con Artemia salina, fueron realizados en los extractos de cada especie vegetal, así como en el medicamento piloto propuesto, generando resultados interesentes que han demostrado la baja o nula toxicidad de las mismas. Morphoanatomic and micrographic studies on Talinum paniculatum (Jacq) Gaertn y Medicago sativa L. were performed in order to validate their characterization and uses. A tentative medicine using these two vegetable species is evaluated based on their nutritional and chemical composition. Both species contains minerals like sodium, potassium, manganese, calcium, iron and zinc, important for homeostasis. Also, Gas chromatographic-Mass spectroscopic (GC-MS) results show the presence of Omega-3, which prevent cardiovascular problems and contribute to minimize triglycerides and cholesterol values (Kris P., 2003). Minerals and Omega-3 elements naturally found in these vegetable species are enhanced by mixing with resveratrol. Resveratrol fights free radicals that are harmful to human cells improving their life time. Furthermore, toxicological studies like the hemolysis test and lethal bioassays with Artemia salina, were performed at the vegetal extract and the tentative medicine, resulting in low or none toxicity.
