Methods for double staining plant materials using astra blue and basic fuchsin are described here. These methods can be applied to free hand and microtome sections embedded in paraffin, paraplast or historesin. Also, they can be used to study isolated epidermal peels and pollen preparations. Temporary, semipermanent and permanent preparations were studied. Astra blue stained polysaccharides of the cell wall such as cellulose and pectins. Basic fuchsin showed an affinity for lignified, suberized or cutinized walls. The easy preparation of the reagents, excellent color contrast of the histological preparations, and brief staining times of some methods makes them useful for both routine research and didactic purposes. Also, excellent color or black and white photomicrography can be obtained after the double staining described here.
Eugenia uniflora L. é utilizada popularmente como anti-hipertensivo, diurético, adstringente, entre outros. Objetivou-se estudar a anatomia das folhas e caule jovem de E. uniflora e fazer o estudo farmacognóstico para a identificação e controle de qualidade da matéria prima vegetal. O material botânico foi coletado em Goiânia, Goiás, dessecado e pulverizado. Determinaram-se os teores de umidade, cinzas totais e insolúveis em ácido. Nas análises microscópicas utilizaram-se as colorações: azul de Alcian/safranina 9:1, Steinmetz, Sudan III, cloreto férrico, lugol, safrablau. O material pulverizado foi utilizado para a triagem fitoquímica, obtenção do extrato etanólico bruto e posteriormente as frações. Por cromatografia em camada delgada (CCD) avaliou-se o perfil cromatográfico das frações. Observaram-se na epiderme da face adaxial da folha, células com paredes anticlinais espessadas, sinuosas e na face abaxial, a presença de estômatos predominantemente anomocíticos. O pecíolo apresentou forma côncava-convexa com parênquima cortical contendo cristais em forma de drusa. Verificou-se no caule jovem em secção transversal forma oval, colênquima com uma a duas camadas de células. Na triagem fitoquímica evidenciou-se a presença de taninos, esteróides, triterpenos, heterosídeos antraquinônicos, saponínicos e flavonóides e em CCD, a presença de flavonóides, taninos e terpenos. Os resultados obtidos podem ajudar na identificação morfo-anatômica de E. uniflora, assim como nos parâmetros de controle de qualidade para a identificação da matéria-prima.
-(Leaf anatomy in young plants of Solanum lycocarpum A.St.-Hil. (Solanaceae)). Solanum lycocarpum A.St.-Hil. is a widely distributed and well adapted shrub of cerrado areas in Brazil. Anatomic studies of the leaves of 60 days old plants obtained from seeds germinated at the greenhouse were carried out. Transversal and paradermal sections were prepared using the usual microthecniques. Both leaf surfaces are covered with simple glandular and non-glandular trichomes, and stellate trichomes. At front sight, the walls of the epidermal cells are sinuous with stomata occurring on both upper and lower surfaces. At cross-section it is observed that the epidermis consists of a single layer of cells and the mesophyll is dorsiventral. The palisade parenchyma consists of a single layer of cells and the parenchyma which is faced to abaxial surface is shaped by three to five layers of cells with compact arrangement. The results constitute a basic reference for further studies about this species, on this stage of development.Key words -early growth, histology, "lobeira", savanna RESUMO -(Anatomia foliar em plantas jovens de Solanum lycocarpum A.St.-Hil. (Solanaceae)). Solanum lycocarpum A.St.-Hil. é um arbusto com ampla distribuição no cerrado brasileiro e se mostra bem adaptado às condições deste ambiente. Foram estudadas anatomicamente folhas provenientes de plantas jovens de S. lycocarpum com 60 dias obtidas por meio da germinação de sementes em casa de vegetação. Cortes transversais e paradérmicos foram preparados de acordo com as técnicas usuais de microtécnica vegetal. Ambas as superfícies foliares são recobertas por tricomas tectores simples bicelulares, estrelados e tricomas glandulares. Em vista frontal, as paredes das células epidérmicas são sinuosas e os estômatos estão distribuídos nas faces adaxial e abaxial. A partir das secções transversais, observa-se que a epiderme é unisseriada e o mesofilo é dorsiventral. O parênquima paliçádico é uniestratificado e o parênquima voltado para a face abaxial é formado por três a cinco camadas de células com disposição compacta. Os resultados constituem uma referência básica para outros estudos envolvendo indivíduos desta espécie, neste estádio de desenvolvimento.Palavras-chave -crescimento inicial, histologia, lobeira, savana 1.

Foi realizado o estudo morfológico e anatômico da folha de 8aulúnia curvula Benth., espécie de cerrado. As observações foram feitas em folhas adultas , tratadas segundo a metodologia usual utilizada em anatomia vegetal. Foi observado que a espécie apresenta as seguintes características: folhas bilobadas cuja incisão foliar chega próximo à região motora; um par de estípulas membranáceas; um par de nectários extraflorais, localizadaos na base do pulvino proximal; a venação possui padrão acródomo; o pecíolo, que possui dois pulvinos, um proximal e um distai, apresenta epiderme contendo grande quantidade de tricomas tectores simples e glândulas; a lâmina foliar é anfiestomática com predomínio de estômatos dos tipos anomocítico e paracítico; a epiderme adaxial é monoestratificada, subpapilosa, desprovida de tricomas tectores simples e glândulas; a epiderme abaxial é subpapilosa, apresentando numerosos tricomas tectores simples e glândulas; e o mesof1ll0 é formado predominantemente por parênquima paliçádico.Palavras-chave: 8auhinia curvula, morfologia foliar, anatomia foliar.ABSTRACT -(Morphology and anatomy of 8auhinia curvula Benth. The observations, in grown up leaves, prepared according to the costumary methodology used in plant anatomy, showed the following characteristics ofthe species: bilobed leaves with foliar incision close to the motor region; one pai r of membranaceous stipules; one pair of extrafloral nectaries located on the basis of the proximal pulvinus; acrodromous patterns of leaf venetion; the petiole, which has two pulvinus, one proximal and one distai, has an epidermis containing large quantities of simple tector trichomes and glands; the leaf blade, amphiestomatic, with predominance of anomocytic and paracytic stomata; the adaxial epidermis uni serial, sub papillose, without tector trichomes and glands; the abaxial epiderms, sub-papillose, presents a high number of simple trichomes and glands; and the mesophyll with a basic composition of palisadic parenchyma.
