Engineered nanoscale materials (ENMs) are considered emerging contaminants since they are perceived as a potential threat to the environment and the human health. The reactions of living organisms when exposed to metal nanoparticles (NPs) or NPs of different size are not well known. Very few studies on NPs–plant interactions have been published, so far. For this reason there is also great concern regarding the potential NPs impact to food safety. Early genotoxic and phytotoxic effects of cerium oxide NPs (nCeO2) and titanium dioxide NPs (nTiO2) were investigated in seedlings of Hordeum vulgare L. Caryopses were exposed to an aqueous dispersion of nCeO2 and nTiO2 at, respectively 0, 500, 1000, and 2000 mg l-1 for 7 days. Genotoxicity was studied by Randomly Amplified Polymorphism DNA (RAPDs) and mitotic index on root tip cells. Differences between treated and control plants were observed in RAPD banding patterns as well as at the chromosomal level with a reduction of cell divisions. At cellular level we monitored the oxidative stress of treated plants in terms of reactive oxygen species (ROS) generation and ATP content. Again nCeO2 influenced clearly these two physiological parameters, while nTiO2 were ineffective. In particular, the dose 500 mg l-1 showed the highest increase regarding both ROS generation and ATP content; the phenomenon were detectable, at different extent, both at root and shoot level. Total Ce and Ti concentration in seedlings was detected by ICP-OES. TEM EDSX microanalysis demonstrated the presence of aggregates of nCeO2 and nTiO2 within root cells of barley. nCeO2 induced modifications in the chromatin aggregation mode in the nuclei of both root and shoot cells.

First developed for biomedical and industrial applications, nanovectors have recently been extended to agriculture. Therefore, innovative plant growing procedures making use of nanoparticles should be adapted to sustainable processes and materials. This work aims at proposing newly synthetized polymeric nanocapsules (NCs) to be used as biocompatible vectors for delivery of bioactive compounds to plants. Nanoparticles were fabricated from lignin, which is the main byproduct of wood processing and is currently a waste material. Lignin can thus find a virtuous fate and be reused in the context of circular economy. Specifically, we loaded lignin NCs with Gibberellic Acid (GA), assessing that stable and reproducible nanoparticles could be obtained in a range of GA content that is relevant for delivery purposes, i.e. 0.5-1.5 mg ml-1. Plain and GA-loaded NCs were characterized by Dynamic Light Scattering (DLS) and Scanning Electron Microscopy (SEM). Assays of cytotoxicity and cargo release were carried out in two model plants, Eruca vesicaria and Solanum lycopersicum. These experiments, conducted both in vitro and in vivo, included the investigations of the percentage of germination, the stem and primary root lengths, as well as the fresh and dry weight of treated plants vs non-treated ones. Furthermore, NCs were loaded with Fluorol Yellow 088 to track their entrance and accumulation in seeds and seedlings.

A major environmental pollution problem is the release into the atmosphere of particulate matter, including nanoparticles (NPs), which causes serious hazards to human and ecosystem health, particularly in urban areas. However, knowledge about the uptake, translocation and accumulation of NPs in plant tissues is almost completely lacking. The uptake of silver nanoparticles (Ag-NPs) and their transport and accumulation in the leaves, stems and roots of three different tree species, downy oak (Quercus pubescens Willd.), Scots pine (Pinus sylvestris L.) and black poplar (Populus nigra L.), were assessed. In the experiment, Ag-NPs were supplied separately to the leaves (via spraying, the foliar treatment) and roots (via watering, the root treatment) of the three species. Uptake, transport and accumulation of Ag were investigated through spectroscopy. The concentration of Ag in the stem was higher in the foliar than in the root treatment, and in poplar more than in oak and pine. Foliar treatment with Ag-NPs reduced aboveground biomass and stem length in poplars, but not in oaks or pines. Species-specific signals of oxidative stress were observed; foliar treatment of oak caused the accumulation of H2O2 in leaves, and both foliar and root treatments of poplar led to increased O2− in leaves. Ag-NPs affected leaf and root bacteria and fungi; in the case of leaves, foliar treatment reduced bacterial populations in oak and poplar and fungi populations in pine, and in the case of roots, root treatment reduced bacteria and increased fungi in poplar. Species-specific mechanisms of interaction, transport, allocation and storage of NPs in trees were found. We demonstrated definitively that NPs enter into the tree stem through leaves faster than through roots in all of the investigated tree species.

Posidonia oceanica (L.) Delile is a marine plant endemic of Mediterranean Sea endowed with interesting bioactivities. The hydroalcholic extract of P. oceanica leaves (POE), rich in polyphenols and carbohydrates, has been shown to inhibit human cancer cell migration. Neuroblastoma is a common childhood extracranial solid tumor with high rate of invasiveness. Novel therapeutics loaded into nanocarriers may be used to target the migratory and metastatic ability of neuroblastoma. Our goal was to improve both the aqueous solubility of POE and its inhibitory effect on cancer cell migration. Methods: Chitosan nanoparticles (NP) and Soluplus polymeric micelles (PM) loaded with POE have been developed. Nanoformulations were chemically and physically defined and characterized. In vitro release studies were also performed. Finally, the inhibitory effect of both nanoformulations was tested on SH-SY5Y cell migration by wound healing assay and compared to that of unformulated POE. Results: Both nanoformulations showed excellent physical and chemical stability during storage, and enhanced the solubility of POE. PM-POE improved the inhibitory effect of POE on cell migration probably due to the high encapsulation efficiency and the prolonged release of the extract. Conclusions: For the first time, a phytocomplex of marine origin, i.e., P. oceanica extract, has enhanced in terms of acqueous solubility and bioactivity once encapsulated inside nanomicelles.
