The objective of this study was to determine the requirement of digestible methionine + cystine of brown-eggs laying hens from 50 to 66 weeks age at the end of the first production cycle. The design was completely randomized, with 150 Brown Shaver hens, which were distributed in five treatments with six replications of five birds each. Birds received a basal diet with 2857 kcal/kg metabolizable energy and 15.97% crude protein, supplemented with 0.132; 0.174, 0.215, 0.256 and 0.298% DL-methionine (98%), in order to provide 0.572, 0.613, 0.653, 0.693 and 0.734% digestible methionine + cystine. The levels of digestible methionine + digestible cystine followed, respectively, the relations of 67, 72, 77, 81 and 86% with lysine fixed at 0.851%. Feed intake, methionine + cystine intake, feed conversion per dozen eggs, egg weigth and mass, percentage of egg components, internal egg quality and weight gain were evaluated. Methionine + cystine levels showed a quadratic effect on feed conversion per dozen eggs and egg weight, a linear effect on feed conversion per kilogram of eggs and percentage of albumen. An experiment was conducted in the Department of Experimental Stations of the Universidade Tecnológica Federal do Parana -UTFPR campus Dois Vizinhos -PR, with the objective of determining the nutritional levels of threonine for laying hens aged 75 to 90 weeks. One hundred fifty laying Shaver Brown pullets were used in a completely randomized design (CRD) submitted to a basal diet containing 2.85 kcal ME/g and 148.7 g/kg CP, supplemented with 4.6; 4.9; 5.2; 5.5 or 5.8 g/kg of digestible Lthreonine (98 %), which provided 0.000, 0.027, 0.058, 0.089 or 0.120% threonine, respectively. The feed conversion, egg production, egg weight, egg mass and internal egg quality were not affected (P> 0.05) by the threonine in the diet. The Haugh unit showed a linear effect with threonine consumption in that it increased levels of digestible amino acids in the diet. The variables percent albumin and shell showed quadratic effects, so the recommended dietary digestible threonine level was 5.20 g/kg, which corresponds to 0.648 g threonine/hen/day, and 5.25 g/kg, which is equivalent to 0.654 g threonine/hen/ day, respectively. The specific gravity was affected by the level of threonine; the indicated level is 5.22 g/kg, equivalent to 0.651 g of threonine/hen/day obtained by the quadratic effect. Foram utilizadas 150 poedeiras Shaver Brown semipesadas num delineamento inteiramente casualizado (DIC), submetidas à uma ração basal contendo 2,85 Kcal EM/g, 148,7g/kg de PB, suplementada com 4,6; 4,9; 5,2; 5,5 e 5,8g/kg de L-treonina digestível (98%), que forneceu 0,000; 0,027; 0,058; 0,089 e 0,120% de treonina digestível, respectivamente. A conversão alimentar, a produção de ovos, o peso do ovo, massa de ovos e a qualidade interna dos ovos não foram influenciados (P>0,05) pelos níveis de treonina na dieta. A unidade Haugh e o consumo de treonina apresentaram efeito linear à medida em que aumentavam os níveis do aminoácido digestível na dieta. As variáveis porcentagem de albúmen e casca apresentaram efeito quadrático e os níveis nutricionais de treonina digestível recomendados foram de 5,20g/kg, o que corresponde a 0,648 g de treonina/ave/dia e, 5,25g/kg que equivale a 0,654 g de treonina/ave/dia, respectivamente. A gravidade específica foi afetada pelos níveis de treonina digestível, e o nível indicado é de 5,22g/kg, que equivale a 0,651 g de treonina/ave/dia obtido por meio de efeito quadrático. The dietary requirement of threonine was determined for brown laying hens in the period 50-66 weeks of age, based on performance parameters and quality of eggs. For this we used 150 laying Shaver Brown hens distributed in a completely randomized design with five treatments (0.460; 0.490; 0.520; 0.550 and 0.580%) of digestible threonine, six replications and five birds each. The digestible threonine levels did not affect (p>0.05) feed intake, egg production, egg weight, egg mass or feed conversion kg kg -1 , but presented a quadratic effect (p<0.05) on feed conversion dozen kg, where the lower conversion was obtained with a supply of 0.521% threonine in the diet. There was no effect of dietary digestible threonine levels (p>0.05) on the yolk and albumen index, specific gravity, yolk percentage, thickness or shell weight per surface area, since the variable Haugh unit displayed linear behavior (p<0.05) and increased with increasing levels of dietary threonine. The percentage of albumen and shell presented a quadratic effect (p<0.05) according to the dietary levels of threonine, and the best levels of these variables were obtained with a supply of 0.520% and 0.521% digestible threonine in the diet. Para isso foram utilizadas 150 poedeiras semipesadas da linhagem Shaver Brown distribuídas em um delineamento inteiramente casualizado com 5 tratamentos (0,460; 0,490; 0,520; 0,550; e 0,580%) de treonina digestível), seis repetições e cinco aves por unidade experimental. Os níveis de treonina digestível não influenciaram (p>0,05) o consumo de ração, produção de ovos, peso de ovos, massa de ovos e conversão alimentar kg kg -1 , mas apresentaram efeito quadrático (p<0,05) sobre a conversão alimentar kg dúzia -1 , onde o menor valor de conversão foi obtido com o fornecimento de 0,521% de treonina digestível na dieta. Não foi observado efeito dos níveis dietéticos de treonina digestível (p>0,05) sobre o índice de gema e de albúmen, gravidade específica, porcentagem de gema, espessura e peso da casca por superfície de área, já a variável unidade haugh apresentou comportamento linear (p<0,05), aumentando com o aumento dos níveis de treonina dietéticos. ResumoOs coeficientes de digestibilidade e o conteúdo de aminoácidos digestíveis verdadeiros de oito cultivares de milho foram determinados utilizando-se o método de alimentação forçada com galos adultos cecectomizados. O delineamento experimental utilizado foi o inteiramente casualizado, representados por oito cultivares, cinco repetições e um galo por unidade experimental. Foram mantidos cinco galos em jejum para as correções correspondentes às perdas metabólicas e endógenas de aminoácidos. Foi observada diferença significativa (P<0,05) entre os cultivares de milho para os coeficientes de digestibilidade verdadeira dos aminoácidos metionina, treonina, arginina, cistina e serina. Os coeficientes de digestibilidade verdadeira dos aminoácidos essenciais metionina, treonina e arginina variaram de 85,61 a 93,57%, 66,27 a 84,97% e 83,39 a 93,10%, respectivamente. Para os demais aminoácidos essenciais, os valores médios foram: 72,84% para lisina, 85,64% para metionina+cistina, 87,29% para histidina, 85,33% para valina, 87,11% para isoleucina, 94,05% para leucina e 90,65% para fenilalanina. Os coeficientes de digestibilidade verdadeira dos aminoácidos não essenciais cistina e serina, que apresentaram diferença significativa, variaram de 73,97 a 89,21% e 77,31 a 92,68%, respectivamente. Para os demais aminoácidos não essenciais, os valores médios foram: 88,29% para alanina, 82,68% para o ácido aspártico, 92,20% para o ácido glutâmico e 87,84% para tirosina. Em função das variações observadas, análises periódicas dos cultivares de milho utilizados para a formulação das rações são recomendadas, a fim de que a qualidade das rações seja mantida garantindo o desempenho dos animais. In order to investigate the effect of different doses of essential oil of Moringa oleifera (EOM) on physiological potential and the expression of isoenzymes in bean seeds, an experiment was conducted with two cultivars (Colibri and Campos Gerais) and seven treatments (bactericidal, fungicide and the doses 0, 0.1, 0.2, 0.4 and 0.8% EOM). Germination, first count of germination, germination speed index (GSI), emergence and emergence rate index (ERI) were evaluated in addition to the enzymes esterase (EST), alcohol dehydrogenase (ADH), superoxide dismutase (SOD) and isocitrate lyase (ICL) by electrophoresis. In an experiment conducted in the laboratory, the dose containing 0.4% EOM promoted germination and higher averages for first germination in seeds and GSI in the Colibri cultivar. However, the highest dose used (0.8%) was responsible for the worst physiological performance by the cultivar. For Campos Gerais, increasing doses of essential oil stimulated germination, first count of germination and GSI. In the field, higher doses of essential oil gave higher germination percentages for the Colibri cultivar, not influencing the seeds of Campos Gerais. The ERI was not altered by the treatments with OEM and changes in the molecular forms of the isoenzymes were observed, with low expression values of bands for EST, ICL and ADH-related vigor, the germination of both cultivars, as well as increased expression of SOD, which is associated with reductions in germination.
O trabalho objetivou avaliar a qualidade microbiológica do ar de uma cooperativa de reciclagem situada em Santa Helena/PR. Foram realizadas cinco coletas de dados em 2021 seguindo-se a metodologia da sedimentação espontânea e utilizando placas de Petri com Ágar DRBC para cultivo de fungos e Ágar EMB para bactérias Gram negativas. As placas foram dispostas abertas em dez diferentes pontos de coleta no interior e exterior da cooperativa por 30 minutos. Após incubação, quantificaram-se as unidades formadoras de colônia em cada placa e procedeu-se o isolamento dos fungos e bactérias. Para identificação dos bolores houve a comparação das suas estruturas com chaves taxonômicas. Para as bactérias, utilizaram-se os Kits Bactray. De modo a conscientizar a população sobre a separação correta dos resíduos sólidos, elaborou-se uma cartilha educacional em formato digital aos estudantes do município. Na quantificação de bactérias Gram negativas no ambiente interno, as coletas 1, 2 e 3 excederam o limite de 500 UFC m-³ de ar, enquanto no ambiente externo, apenas a coleta 5 apresentou média que não atendeu à legislação portuguesa NT-SCE-02 (utilizada como parâmetro, pois o Brasil não possui legislação específica para tal). Para avaliação frente ao conceito de normalidade, a Relação I/E (onde I é a quantidade de microrganismos no ambiente interior e E é a quantidade de microrganismos no ambiente exterior) de bactérias foi superior a 1,5, indicando fontes internas de contaminação. Foram identificadas nove espécies de bactérias, pertencentes à cinco gêneros (Acinetobacter sp., Klebsiella sp., Serratia sp., Shigella sp. e Stenotrophomonas sp.) no ambiente interno e, cinco espécies no ambiente externo, com predominância do gênero Klebsiella sp. A contagem de fungos excedeu os limites da Resolução ANVISA 09/2003 (750 UFC m-³) na primeira, quarta e quinta coletas com médias de até 956,21 UFC m-³ de ar. O ambiente está de acordo com a Relação I/E. Foram identificados 13 gêneros de bolores no ambiente interno, com predominância de Aspergillus sp., Rhizopus sp., e Eupenicillium sp. e no ambiente externo, contabilizou-se 19 gêneros com prevalência de Aspergillus sp., Rhizopus sp., Eupenicillium sp., Trychophyton sp. e Mucor sp. Os resultados indicam uma qualidade microbiológica do ar ambiente significativamente ruim na cooperativa analisada, com três amostragens com valores superiores ao recomendado pelas legislações, sendo possível associar a grande quantidade de microrganismos encontrados, com a grande quantidade de materiais que chegam à cooperativa, contaminados, sujos ou contendo matéria orgânica em seu meio, facilitando a proliferação dos microrganismos.
