-Pesticide degradation studies are essential to evaluate its impact in the environment and on non-target organisms. The effect of repeated soil applications of the herbicide glyphosate on its dissipation and on soil microorganisms was studied by radiometric and microbial techniques. Results indicated fast dissipation of the [ 14 C]-glyphosate or [ 14 C]metabolites extractable residues (half-life of 0.92±0.29 month), but increasing half-lives of total mineralization ranging from 2.2 to 3.4 months as the number of applications increased from 1 to 4. No significant correlation was found between 14 CO 2 production and dehydrogenase activity.Index terms: pesticide residues, mineralization, dehydrogenases, soil treatment. Influência de repetidas aplicações de glifosato na sua persistência e na bioatividade do soloResumo -Estudos sobre degradação de pesticidas são essenciais à avaliação do impacto que causam no ambiente e em organismos não-alvo. Avaliou-se o efeito de repetidas aplicações do herbicida glifosato em solo na sua dissipação e em microrganismos do solo, por técnicas radiométricas e microbianas. Os resultados indicaram dissipação rápida dos resíduos extraíveis de glifosato marcado com 14 C ou de metabólitos contendo 14 C (meia-vida de 0,92±0,29 mês), mas meia-vida crescente de 2,2 a 3,4 meses para mineralização total, de acordo com o aumento de 1 a 4 aplicações. A persistência de um pesticida no solo depende de processos de dissipação, como a degradação, que pode estar relacionada com o metabolismo microbiano. Com o objetivo de se avaliar os mecanismos de dissipação do herbicida atrazina no solo e a importância dos microrganismos neste processo, avaliou-se sua mineralização, degradação intermediária e formação de resíduos não-extraíveis ou ligados ao solo após aplicação de 14C-atrazina em solo Glei Húmico. Os processos foram quantificados através de técnicas radiométricas e cromatográficas em solo natural, solo esterilizado e solo esterilizado e infectado com Pseudomonas putida. Observou-se a importância dos microrganismos através dos estudos de biomineralização nos quais detectou-se mineralização de 14C-atrazina somente em solo natural (cerca de 15%). Entretanto, houve formação de metabólitos extraíveis de atrazina, tanto em solo esterilizado (67%) como em solo natural (75%), o que indica que este processo não dependeu apenas da presença de microrganismos. Já os resíduos não-extraíveis foram formados em maior quantidade (cerca de 56%) no solo esterilizado.
The soil oxidative and anaerobic processes, as well as, the microbial biomass were followed during three years in a cotton farm (Tatuí) where the recommended pesticides have been used for several years, and in an experimental field (São Paulo) treated first time with the same pesticides. The oxidative process was monitored by the dehydrogenase (DHA)-activity using triphenyltetrazolium chloride (TTC) as substrate. The anaerobic process was followed by the iron-oxide reduction, and the microbial biomass was estimated by the substrate (glucose)-indiced respiration. Increases in DHA-activity and in the microbial biomass occurred only in the farm soil, with concomitant decreases in iron-reduction. In the experimental field soil, the increases in DHA-activity were followed only by decreases in iron-reduction. Soil characteristics were the determining factor for different biological parameters after pesticide inputs. All the pesticides produced at least one clear but transient effect.
Microwave-assisted solvent extraction (MASE) was investigated as an alternative for extraction of parathion (O,O-diethyl O-4-nitrophenyl phosphorothioate), methyl parathion (O,O-dimethyl O-4-nitrophenyl phosphorothioate), p,p'-DDE [1,1'-dichloro-2,2-bis(4-chlorophenyl)ethane], hexachlorobenzene (HCB), simazine (6-chloro-N2,N4-diethyl- 1 ,3,5-triazine-2,4-diamine) and paraquat dichoride (1,1'-dimethyl-4,4'-bipyridinium) from two different soils and from an earthworm-growing substrate. The matrices were fortified with 14C-radiolabeled pesticides and extracted with various solvent systems under different microwave conditions. Recoveries of more than 80% could be obtained depending on the used microwave conditions and solvent, except for paraquat whose recovery was generally less efficient. Thus, MASE can be successfully used to extract pesticides from environmental and biological samples and could be a viable alternative to conventional extraction methods. The technique uses smaller amounts of organic solvents, thereby minimizing the costs of the analysis and the disposal of waste solvent.
This paper reports on the residues of methyl parathion (O,O-dimethyl O-4-nitrophenyl phosphorothioate), trifluralin (alpha, alpha, alpha-trifluoro-2,6-dinitro-N,N-dipropyl-p-toluidine), endosulfan [(1, 4, 5, 6, 7, 7-hexachloro-8, 9, 10-trinorborn-5-en-2, 3-ylenebismethylene) sulfite] and dimethoate (O, O-dimethyl S-methylcarbamoylmethyl phosphorodithioate) in a cotton crop soil. Soil samples (0-15 cm) were collected at different periods from the cotton crop farm and subjected to Soxhlet extraction. The extracted material was analysed after clean-up by a HP5890 II gas chromatograph equipped with a 63Ni electron-capture detector (ECD-63Ni) and fitted with a 25 m x 0.2 mm i.d. fused silica capillary column [Ultra-2 (5% phenylmethyl polysiloxane)]. The recoveries of the pesticide residues from the spiked control soil were determined after Soxhlet extraction and C18 cartridges clean-up by using radiotracer techniques with the corresponding 14C-pesticides. The results show that in the cotton crop soil the pesticide residues under study were present in the range of 0.1 to 0.4 mg.kg-1. Endosulfan was found to be rapidly degraded in the soil and formed a sulfate metabolite.
The pyrethroid insecticide cypermethrin is used for agricultural and public health campaigns. Its residues may contaminate soils and the beneficial soil organisms, like the earthworms, that may ingest the contaminated soil particles. Due to its ecological relevance, earthworms Eisenia andrei/fetida have been used in different ecotoxicological tests. The avoidance of soils treated with cypermethrin by compost worms Eisenia andrei was studied here as a bioindicator of the influence of treatment dosage and the pesticide formulation in three different agricultural soils indicated by the Brazilian environmental authorities for ecotoxicological tests. This earthworms’ behavior was studied here as a first attempt to propose the test for regulation purposes. The two-compartment test systems, where the earthworms were placed for a two-day exposure period, contained samples of untreated soil alone or together with soil treated with technical grade or wettable powder formulation of cypermethrin. After 48 h, there was no mortality, but the avoidance was clear because all earthworms were found in the untreated section of each type of soil (p < 0.05). No differences were found by the Fisher’s exact test (p ≤ 1.000) for each soil and treatment, demonstrating that the different soil characteristics, the cypermethrin concentrations and formulation, as well as the smaller amounts of soil and earthworms did not influence the avoidance behavior of the earthworms to cypermethrin. The number and range of treatments used in this study do not allow a detailed recommendation of the conditions applied here, but to the best of our knowledge, this is the first reported attempt to identify the avoidance of pesticide treated tropical soils by earthworms.
RESUMOGeralmente, os herbicidas são aplicados diretamente no solo, razão pela qual entram em contato direto com organismos deste ambiente, dentre eles as minhocas, as quais pelo metabolismo podem agir sobre resíduos desses compostos. Tomando minhocas da espécie Eisenia foetida como exemplo, determinaram-se a dissipação dos 14 C-herbicidas (simazina e paraquat) e a bioacumulação destes compostos em seus tecidos, a partir de solo tratado com as concentrações recomendadas e, no caso do paraquat, também com concentrações superiores. Solo e minhocas foram analisados por extração com solventes e técnicas radiométricas, após 30 ou 90 dias de contato. As minhocas alteraram a dissipação do simazina, visto que houve 100 % de recuperação do radiocarbono na ausência de minhocas e 90 % na presença dos animais. Além disso, elas acumularam resíduos e, ou, metabólitos de simazina em seus tecidos, tendo-se detectado Fator de Bioacumulação (FB) de 1,45 e 1,17 após exposição ao solo tratado durante 30 e 90 dias, respectivamente. Por outro lado, a presença de minhocas não alterou o comportamento do herbicida paraquat aplicado ao solo, mas houve bioacumulação crescente de seus resíduos e, ou, metabólitos com o aumento da dose de tratamento (FB de 0,5; 3,2 e 5,5, respectivamente, nas concentrações recomendadas, 10 e 100 vezes superior).Termos de indexação: 14 C-simazina, 14 C-paraquat, persistência, bioacumulação.(1) Recebido para publicação em fevereiro de 2003 e aprovado em novembro de 2003. RESUMO -Com a difusão do uso de agrotóxicos, torna-se necessário aprofundar os conhecimentos sobre seu comportamento no ambiente, visando diminuir os riscos à biota e a possível contaminação de água, solo e alimentos. A influência da presença de minhocas da espécie Eisenia foetida sobre a dissipação do herbicida glyphosate, a bioacumulação do herbicida nestes vermes e a influência do herbicida sobre a bioatividade da microbiota endógena foram avaliadas em amostra de terra agrícola tratada com solução aquosa de 14 C-glyphosate. Os estudos foram conduzidos em sistemas mantidos por dois ou quatro meses em amostras de terra tratadas com três diferentes concentrações de 14 C-glyphosate, contendo ou não minhocas. Após esses períodos, amostras de terra e de minhocas foram submetidas à extração e combustão, para quantificação da radioatividade por espectrometria de cintilação em líquido (ECL). Paralelamente, a atividade da microbiota foi avaliada em relação à atividade da enzima desidrogenase. Os resultados mostraram que a presença de minhocas não alterou a dissipação de glyphosate na terra independentemente do período de contato, embora elas tenham bioacumulado resíduos do herbicida, numa relação diretamente proporcional ao tempo de contato com a terra tratada. O período de quatro meses favoreceu a formação de 14 C-resíduos não-extraíveis ou ligados. Já a bioatividade não sofreu qualquer alteração, nem pela presença de minhocas nem dos tratamentos ou do tempo de exposição. Palavras-chave: bioacumulação, desidrogenase, Eisenia foetida.
