This study investigated the frequency and distribution of patellar luxation in the dogs presented to the Chonbuk National University Animal Medical Centre during January 2000 to September 2005. Patellar luxations were classified as medial or lateral, and unilateral or bilateral, were graded I to IV, and were subdivided according to age, sex and size of the dogs. The incidence of medial patellar luxation (MPL) was greater than the incidence of lateral patellar luxation (LPL) in both small and large dogs. Small-breed dogs were admitted almost exclusively with MPL. LPL was found uncommon; however it was observed more often in larger-breed dogs. Surgical correction was performed primarily in the dogs (165 stifles in 111 dogs) with grade II, III and IV patellar luxations following different surgical techniques. The combination of the surgical techniques was found to be more effective for the management of the disease. The prognosis was found to be favourable, because when the grade was low, the dog was younger, without cruciate ligament rupture, and as the surgical correction was performed with combination of more techniques.
The present study was described thin layer chromatography (TLC) and ultra-high performance liquid chromatography (UHPLC) method for the detection of antibacterial substances in poultry muscle (breast and thigh), kidney, and liver.Materials and Methods: TLC method was used for screening detection of tetracycline, amoxicillin, ciprofloxacin, and enrofloxacin residues in poultry tissues. The samples were extracted with trichloroacetic acid (30%), diethyl ether, followed by detection in pre-coated TLC paper on ultraviolet detector. The UHPLC method was used for the quantification of antimicrobial residues in poultry tissues. Results:The residues of tetracycline were 48% in livers, 24% in kidneys, 20% in thigh muscles, and 24% in breast muscles. Ciprofloxacin residues were found 44% in liver, 42% in kidneys, 34% in thigh muscles and 30% in breast muscles. Enrofloxacin residues were found 40% in liver, 34% in kidneys, 22% in thigh muscles, and 18% in breast muscles. Amoxicillin residues were found 42% in liver, 30% in kidneys, 26% in thigh muscles and 22% in breast muscles. Most of the cases highest residues were found in liver such as tetracycline (48%), ciprofloxacin (44%), enrofloxacin (40%) and amoxicillin (42%) and almost lowest in breast muscles. In addition, nine positive samples from broiler were selected for amoxicillin residue quantification by UHPLC. It was observed that the concentration of amoxicillin residue in liver was ranging from 16.92 μg/kg to 152.62 μg/kg and in breast muscle was 45.38 μg/kg to 60.55 μg/kg, respectively. The maximum and minimum peak time was 4.7-5.2 min. Among the poultry tissues, liver had the highest level of antibiotic residues in comparison to other samples but the variation was not significant (p>0.05). Conclusions:Evidence suggests that more judicious use of antimicrobials in food animals will reduce the selection of resistant bacteria and help to preserve these valuable drugs for both human and veterinary medicine. The method described in this study is a simple, easy inexpensive which can be readily adopted by any laboratory for the detection of antibiotic residues in tissues of food-producing animals.
Aim:The study was conducted to detection and determination of concentration or level of antibiotic residues in milk and egg of local and commercial farms at Chittagong during December 2011 to June 2012.Materials and Methods:A total of 400 (200 milk and 200 egg) samples were collected from local and commercial dairy cows and local scavenging and commercial poultry farms, respectively. Microbial inhibition test and thin layer chromatography were used for screening and ultra-high performance liquid chromatography was used to estimate the concentrations of antibiotic residues in samples.Results:Tetracycline, amoxicillin, and ciprofloxacin residues were significantly (p ≤ 0.05) higher in commercial farms than local. The boiling insignificantly (p>0.05) reduced residues level in milk and egg. The average concentrations of amoxicillin residue in local milk, commercial milk, local egg, and commercial egg were 9.84 µg/ml, 56.16 µg/ml, 10.46 µg/g and 48.82 µg/g, respectively, in raw samples and were reduced to 9.81 µg/ml, 55.54 µg/ml, 10.29 µg/g, and 48.38 µg/g, respectively, after boiling.Conclusions:Proper maintaining of the withdrawal period and development of active surveillance system are highly recommended for public health security.
Aim: Salmonellosis is one of the most common and widely distributed foodborne illnesses in human, and multidrugresistance of Salmonella spp. has increased in developing countries with the indiscriminate use of antibiotics in the poultry production system. A cross-sectional study was conducted on randomly selected retail markets of Chittagong City Corporation to determine the prevalence and antimicrobial resistance pattern of Salmonella isolated from commercial layer eggs, eggshell surface, and egg-storing trays. Materials and Methods:Chicken eggs, egg surface, and egg-storing trays samples from the retail markets were collected for isolating Salmonella spp. (bacteriological culture methods) followed by antimicrobial susceptibility testing (disc diffusion method) against Salmonella isolates during the period from July to December 2013.Results: Out of the 310 layer eggs, egg surface, and egg-storing trays samples, the highest prevalence of Salmonella spp. was found in eggs trays (57.15%) and the lowest (13.33%) in eggs and the prevalence differed significantly (p<0.01). On the other hand, the prevalence was higher (45%) in samples of Pahartali bazar and lower (31.43%) in samples of Bohderhat bazar but the variation among the sites was not varied significantly (p>0.05). Isolated Salmonella was tested for resistance to eight different antimicrobial agents, using disc diffusion method. Among eight antimicrobial tested (n=111), 100% resistance were found to ampicillin and amoxicillin followed by erythromycin (60-100%), tetracycline (72-93%), ciprofloxacin (22-66%), colistin (27-66%), enrofloxacin (42-54%), and pefloxacin 23.07% across the study sites. Ciprofloxacin remained sensitive in 40.9% cases and, pefloxacin and colistin appeared to be almost sensitive (61-72%) against Salmonella isolates at studied areas. Salmonella isolates showed multidrug-resistance pattern up to five of the eight antimicrobials tested. Conclusion:It can be said that the rational use of antibiotics needs to be adopted in commercial poultry farming system of Bangladesh to prevent the emergence of drug-resistance Salmonella to protect the public health consequences.
ABSTRACT:This study was performed to make a surgical model of osteoarthritis (OA) in the dog. Experimental medial patellar luxation (MPL) was surgically produced in the left stifle (index) of 24 skeletally mature mixed small breed dogs (age two to six years and weight 2.8 to 9 kg). The animals were randomly allocated in 2 groups; sham group (n = 12), where the right stifle was sham operated and control group (n = 12) with intact right stifle. Physical and radiographic examinations of both stifles were performed at 1.5 months intervals over a one-year experimental period. One dog was euthanatized every three months, and both stifles were explored, gross examination was performed and tissue samples from the articular cartilage, cranial cruciate ligament (CCL) and synovium were collected for histomorphology. The clinical signs of OA were obvious in the experimental dogs by 12 weeks of surgical induction of MPL, which was also evidenced in the histopathology of the joint tissues and electron microscopy of the articular cartilage. The radiographic changes of OA were not obvious until remarkable degenerative changes became abvious six months postoperatively. Surgically induced MPL can be a successful tool for experimental induction of OA in dogs.
The levels of tartrate resistant acid phosphatase (TRAP), matrix metalloproteinase-2 (MMP-2), and tissue inhibitor of matrix metalloproteinase-2 (TIMP-2) in synovial fluid (SF) and serum in cases of canine osteoarthritis (OA) were measured. OA was induced by a surgically-created medial patellar luxation in the left stifle of 24 dogs. SF and blood samples were collected at 1.5- and 3-month intervals, respectively. Every 3 months, one dog was euthanatized to collect tissue samples from both stifles. TRAP levels in SF and serum were measured using a spectrophotometer, and TRAP-positive cells in joint tissues were identified by enzyme histochemistry. MMP-2 and TIMP-2 in SF and serum were detected by Western blotting and ELISA, respectively. TRAP in SF from the stifles and serum was significantly increased (p < 0.05) after 3 months. TIMP-2 in SF and serum was significantly decreased (p < 0.05), whereas MMP-2 in SF was significantly increased (p < 0.05) during the progression of OA. Histochemistry revealed an increased number of TRAP-positive cells in tissues from OA-affected joints. Assays measuring TRAP, MMP-2, and TIMP-2 in SF and serum, and methods that detect increased numbers of TRAP-positive cells in the joint tissues can play an important role in identifying the early phases of degenerative changes in canine joint components.
The effects of Korean Safflower (Carthamus tinctorius L) seed oil (SSO) on osteoporosis induced-ovariectomized rats were investigated. A total of 90 female Sprague-Dawley rats, 4-month-old, weighing 200-230 g were randomly assigned into 3 groups (30 animals/group). The animals in group I were sham operated and those of group II and III were ovariectomized (Ovx). After eight weeks, the animals of group I and II received solvent vehicle daily, whereas those of group III were administered SSO orally (1 ml/kg) daily for 30 days. The changes in the serum levels of insulin-like growth factor-I (IGF-I), IGF-II, insulin-like growth factor binding protein-3 (IGBP-3), estrogen, total alkaline phosphatase (TALP), bone-specific alkaline phosphatase (BALP), calcium and phosphorous in serum, and also the histomorphology of the proximal tibia metaphysis and femur/body weight (F/B) ratio were examined in all the groups at every 10 days interval. Thirty days later, IGF-I, IGF-II, IGBP-3 and BALP levels were significantly increased (p < 0.05) in group III as compared to groups I and II. There were no significant differences in serum levels of estrogen, TALP and F/B ratio between groups II and III, but estrogen levels were higher in group I. These results suggest that the safflower seeds have possible roles in the improvement of osteoporosis induced-ovariectomized rats.
ABSTRACT. Two Holstein Friesian calves were referred to the Animal Medical Center of Chonbuk National University with severe lameness on the hind limb(s), flexed stifle joint and an inability to walk since birth. Based on the clinical history, clinical findings and radiography of the stifle joint, the cases were diagnosed as grade III lateral patellar luxation (LPL). Trochleoplasty along with medial retinacular reinforcement and lateral release were performed in both calves. The calves recovered 3 to 4 weeks after surgery. Trochleoplasty along with medial retinacular reinforcement and lateral release can be an effective technique for the treatment of LPL in calves.
