2‐(N‐Formyl‐N‐prop‐2′‐inyl)‐aminopyridin cyclisiert in siedender Ameisensäure zum 3‐Methylimidazo‐[1,2‐a]pyridin, wobei 3‐Methylen‐2H‐imidazo[1,2‐a]pyridin als Intermediat auftritt. Unter den gleichen Bedingungen bildet sich aus dem 1,3‐Diprop‐2‐inylpyrimido[4,5‐b]chinolin‐2,4‐dion das 1‐Methylimidazo‐[1,2‐a]chinolin‐4‐carbonsäure‐N‐prop‐2′‐inylamid und aus dem 1‐Prop‐2′‐inylbenzo[b][1,8]naphthyridin‐2‐on als neues Ringsystem das 1‐Methyl‐benzo[b]imidazo[1,2,3‐ij][1,8]naphthyridin‐4,7‐dion.
Die Friedländer-Kondensation von o-Aminobenzaldehyd (1) mit 2,6-Dihydroxypyridinen führte zu den BenzofbH 1,81naphthyridinen 6a und 6b. Die Umsetzung von 1 mit 3-Cyano-2,4,6-trihydroxypyridin (4) lieferte außer dem erwarteten 1,3(2H,5H)-Dioxobenzof b 111,6Inaphthyridin-4-carbonitril (7) auch 4-Hydroxy-2( I H)-oxo-benzo! bl! I ,8Inaphthyridin-3-carbonitril (8a). Neben der N -Methylierung wurden diese Ringsysteme mit zur Cyclisation befähigten Alkylresten substituiert.
Friedländer Condensations of o-Aminobenzaldehyde with HydroxypyridinesCondensations of o-aminobenzaldehyd (1) with 2,6-dihydroxypyridines under Friedländer conditions result in the formation ofthe benzolblll,8lnaphthyridines 6a and 6b. The reaction of 1 with 2,4,6-trihydroxypyridine-3-carbonitrile (4) yields the expected 1.3(2H.5H)-dioxobenzol bill ,6Inaphthyridin-4-carbonitrile (7). The 4-hydroxy-2(lH)-oxobenzofblf 1.8Inaphthyridin-3-carbonitrile (8a) was also isolated. N-Methylation and substitution ofthe ring systems with alkyl groups which allow cyclisation reactions to be carried out, are also reported. 1901 setzten Conrad und Reinbaeh 2 ) o-Aminobenzaldehyd (1) mit Barbitursäure (2) nach Art einer Friedländer-Kondensation zum PyrimidoI4.5-blchinolin-2A-dion (5a) um. Bei der Suche nach synthetischen Analoga vom Acronycin bzw. Flavin sollten aus 1 und Hydroxypyridinen (3) (neuartige) Tricyclen hergestellt werden. Um im gewünschten Sinne zu reagieren. muß 3 in einer Tautomerform vorliegen. die eine für die Kondensation notwendige a-Methylencarbonyl-Struktur aufweist. Diese Voraussetzung wird besonders vom 2,6-Dihydroxy-und 2,4,6-Trihydroxypyridin erfüllt. Die vorliegende Studie befaßt sich mit der Reaktion von 1 mit 2,6-Dihydroxypyridinen (3a u. 3b) und 3-Cyano-2,4,6-trihydroxypyridin (4).

Similar ring closure processes take place with the benzonaphthyridine (III) or with the aminopyridine (V), yielding the fused imidazoles (IV) and (VI) or (VII).
Condensation of 2.4.5‐trimethoxybenzaldehyde with hydantoin followed by catalytic hydrogenation gave 5‐(2.4.5‐trimethoxybenzyl)‐hydantoin, which was converted to 3‐amino‐3.4‐dihydro‐6.7‐dihydroxycoumarin by treatment with hydrobromic acid. Hydrolysis of the lactone led to 3‐(2.4.5‐trihydroxyphenyl)‐DL‐alanine.

The latter is prepared independently from the aminoquinolinecarboxylate (XI) and the cyanoacetate (XII). The pyridones (III), (X), and the pyrimidone (VI) are alkylated to form the N-substituted derivatives (V), (VII), and (XIII).
