Heme oxygenase-1 (HO-1) is an enzyme that catabolizes free heme, which induces an intense inflammatory response. The expression of HO-1 is induced by different stimuli, triggering an anti-inflammatory response during biological stress. It was previously verified that HO-1 is able to induce indoleamine 2,3-dioxygenase (IDO), an enzyme that is induced by IFN-γ in Toxoplasma gondii infection. To verify the role of HO-1 during in vivo T. gondii infection, BALB/c and C57BL/6 mice were infected with the ME49 strain and treated with zinc protoporphyrin IX (ZnPPIX) or hemin, which inhibit or induce HO-1 activity, respectively. The results show that T. gondii infection induced high levels of HO-1 expression in the lung of BALB/c and C57BL6 mice. The animals treated with ZnPPIX presented higher parasitism in the lungs of both lineages of mice, whereas hemin treatment decreased the parasite replication in this organ and in the small intestine of infected C57BL/6 mice. Furthermore, C57BL/6 mice infected with T. gondii and treated with hemin showed higher levels of IDO expression in the lungs and small intestine than uninfected mice. In conclusion, our data suggest that HO-1 activity is involved in the control of T. gondii in the lungs of both mouse lineages, whereas the hemin, a HO-1 inducer, seems to be involved in the control of parasitism in the small intestine of C57BL/6 mice.
Lettuce is the most-consumed leafy vegetable in Brazil due to its ease of preparation, nutritional quality, and affordability. Despite these benefits, there are few cultivars of crisphead lettuce (Lactuca sativa) that possess agronomic potential, high carotenoid content and resistance to nematodes. Therefore, we evaluated the agronomic potential of crisphead lettuce genotypes that are resistant to the root-knot nematode. The experiment was conducted at the Vegetable Experiment Station of the Federal University of Uberlandia (UFU), Monte Carmelo Campus. The trial was set up in a randomized block design with 49 treatments (43 genotypes obtained from crosses between Uberlandia 10000 and Belissima and six commercial cultivars) and three replications. After preparing the beds and before transplanting the seedlings, 11 simple samples were taken from each of the three experimental blocks to confirm the presence of nematodes within the experimental area. The following variables were determined 48 days after transplanting: chlorophyll content, leaf temperature, plant diameter, number of leaves per plant, fresh mass and incidence of root gall. The data were submitted to analysis of variance (ANOVA) and an F-test and the means were compared by the Scott-Knott test. Genetic diversity was represented by a dendrogram that was obtained using the hierarchical method of UPGMA and the optimization method of Tocher. We found genetic divergence and agronomic potential among the evaluated genotypes. Specifically, 199#1#1-E, 199#2#1-E, 199#2#2-E, 199#3#1-E, 199#6#1-E and 75#2#2-E presented good agronomic potential, were rich in carotenoids and showed resistance to the root-knot nematode.
The frequent tank mixing of phytosanitary products, adjuvants, and foliar fertilizers highlights the lack of information which sustains decisions about what products can be mixed for spray application. Thus, the aim of this study was to evaluate the physical and chemical characteristics of fungicides, herbicides, and insecticides in combination with some adjuvants on the Brazilian market. The experimental design was completely randomized and spray mixes of the phytosanitary products: fungicide (azoxystrobin+benzovindiflupir), herbicide (diamônio salt of N-(phosponomethyl)) or insecticide (fenpropathrin) were evaluated in combination with adjuvants (mineral oil base, foliar fertilizer or lecithin + propionic acid), and in two application rates (95 and 52 L ha-1); all with four replications. Surface tension, electrical conductivity, pH, dynamic viscosity and density of spray mixes were evaluated. The adjuvants presented characteristics capable of significantly altering the physicochemical properties of the phytosanitary spray mixes, and thus, alter the biological effectiveness and efficiency of the spray applications. However, its effects are also dependent on the phytosanitary product added to the spray mix, which makes general recommendations a difficult task. The greatest pH reduction, as well as the greatest increase in electrical conductivity, were caused by the adjuvant lecithin + propionic acid. All phytosanitary products and adjuvants studied, associated or not, resulted in a reduction in the surface tension of the spray mix in relation to water. The magnitude of change of density and dynamic viscosity promoted by adjuvants was lower than the other characteristics evaluated.
RESUMO A ferrugem asiática da soja (FAS), causada pelo fungo Phakopsora pachyrhizi, é uma das doenças mais severas na cultura. Dentre as estratégias para seu manejo propõe-se a utilização de genótipos com resistência horizontal ou parcial, ainda restritos no mercado, mas promissores pela durabilidade da resistência. O objetivo deste trabalho foi avaliar a resistência de diferentes genótipos de soja à ferrugem asiática para futura utilização em programas de melhoramento. Na safra 2015/2016, foram avaliados 11 genótipos de soja provenientes do LAGER-UFU e a cultivar Desafio 8473 (padrão de suscetibilidade) frente à infecção artificial e natural de P. pachyrhizi no campo. Com os dados de severidade da FAS, calculou-se a área abaixo da curva de progresso da doença (AACPD). O percentual de área foliar afetada e os valores da AACPD dos genótipos avaliados oscilaram de 1,56 a 80,75 e de 380,62 a 975,62, respectivamente. Os genótipos L-210, L-166, L-268, L-203, L-279, L-224, L-216 e L-218 apresentam menores valores de AACPD, indicando resistência parcial, enquanto os genótipos L-144, L-104, L-266 e a cv. Desafio 8473 apresentaram maiores valores de AACPD, portanto, com menor resistência parcial ou suscetíveis. Ainda assim, os genótipos L-144, L-104 e L-266 apresentaram coeficientes de progresso da severidade da doença de 1,8 a 2,0 vezes menores comparativamente à cv. Desafio 8473. Conclui-se que os genótipos do Programa de Melhoramento Genético da UFU (LAGER-UFU), apresentam potencial de utilização no desenvolvimento de cultivares de soja resistentes à P. pachyrhizi.
Objetivou-se estudar o potencial antagônico de isolados de Bacillus subtilis a Colletotrichum gloeosporioides, agente causal da antracnose em frutos de pimenta. Foram estudados 21 isolados de Bacillus subtilis quanto a capacidade de inibir o desenvolvimento do fitopatógeno nas seguintes metodologias in vitro: cultivo pareado, metabólitos produzidos pelos isolados, influência da inoculação conjunta e controle da antracnose in vivo (frutos destacados de pimenta). Os isolados BSV-11, BSV-05, BSV-12, BSV-17, BSV-07, BSV-16, BSV-09, BSV-20, BSV-13 e BSV-18 foram promissores como potenciais antagonistas a C. gloeosporioides, com evidente formação de halo de inibição, para a metodologia do cultivo pareado. Estes isolados produziram metabólitos termoestáveis contra C. gloeosporioides evidenciando ser antibiose o mecanismo antagônico envolvido na supressão do patógeno. O contato direto dos isolados bacterianos com o patógeno inibiu totalmente o crescimento micelial de C. gloeosporioides, exceto os isolados BSV-20 e BSV-09. Os isolados BSV-20, BSV-18, BSV-17, BSV-07 e BSV-12 inibiram significativamente infecções de antracnose em frutos de pimenta em pós-colheita.

<p>Durante uma seleção de bactérias antagonistas realizada no Laboratório de Microbiologia e Fitopatologia da UFU, o isolado de <em>Bacillus subtilis</em>, BSV – 05 se destacou. Diante disso, objetivou-se no presente trabalho avaliar o potencial antagonístico <em>in vitro</em> deste isolado bacteriano contra quatro patógenos radiculares do feijoeiro: <em>Fusarium solani </em>f. sp. <em>phaseoli, Fusarium oxysporum</em> f. sp. <em>phaseoli, Macrophomina phaseolina </em>e <em>Rhizoctonia solani</em>. Foram utilizadas as seguintes metodologias <em>in vitro </em>versus os patógenos citados:<em> </em>cultivo pareado, influência da inoculação conjunta, produção de metabolitos voláteis e não voláteis. Os percentuais de inibição do isolado BSV – 05 sobre <em>M. phaseolina, R. solani, F. solani,</em> <em>F. oxysporum</em>, foram de 61,43%; 50,36%; 13,74% e 7,7%, respectivamente, para a metodologia do cultivo pareado. O contato direto da bactéria com os patógenos inibiu em 100% o crescimento micelial de <em>M. phaseolina, R. solani</em>; e para <em>F. solani </em>e <em>F. oxysporum</em>, foram observadas inibições de 90 e 92 %, respectivamente. Possíveis metabólitos secretados pelo isolado BSV 05 apresentaram níveis de inibição de 100% para <em>R. solani</em>. Para <em>M. phaseolina</em>, observou-se uma porcentagem de inibição de 80,26%, e 45,31% e 47,80% para <em>F. solani </em>e <em>F. oxysporum</em>, respectivamente. A porcentagem de inibição da germinação de conídios de <em>F. solani </em>e <em>F. oxysporum </em>e<em> M. phaseolina</em> num meio de cultura contendo substâncias metabolizadas por BSV – 05 foi de 87,70; 91,28% e 100%, respectivamente. O isolado bacteriano BSV-05 não apresentou produzir nenhum metabólito volátil.</p>
Lettuce is the main leafy vegetable, presenting the highest consumption and economic importance in the world. In the recent decades, concerns about vitamin A deficiency have led Brazilian researchers to develop cultivars with higher levels of carotenoids provitamin A. Therefore the experiment aimed to verify the genetic dissimilarity of biofortified lettuce genotypes, investigating the correlation between agronomic characteristics and the potential for use to increase the carotenoid content in future breeding programs. Ninety one genotypes were evaluated, with 86 lettuce strains from hybridization between cultivars Pira 72 versus Uberlândia 10000 (rich in carotenoids) followed by three successive self-fertilizations and 5 commercial cultivars (Grand Rapids; UFU MC BIOFORT1; Pira 72; Uberlândia 10000 and Robusta).During the course of the trial, were evaluated characteristics as chlorophyll content of the leaves, plant diameter, stem diameter and the number of leaves per plant were counted. Genetic diversity was represented by a dendrogram that was obtained using the hierarchical method of UPGMA and the optimization method of Tocher. There is genetic divergence between the lettuce genotypes analyzed, and the carotenoid content character contributed most significantly to the divergence between the evaluated genotypes. The UFU215#12 genotypes; UFU215#14; UFU215#2; UFU 215#1; UFU215#4; UFU199#3 and UFU199#2 have good agronomic characteristics and high levels of carotenoids, being promising to continue the biofortified lettuce breeding program, aiming to obtain new cultivars rich in carotenoids pro vitamin A.
