Abscisic acid (ABA) plays a key role in plant growth and development. The effect of ABA in plants mainly depends on its concentration, which is determined by a balance between biosynthesis and catabolism of ABA. In this study, we characterize a unique UDP-glucosyltransferase (UGT), UGT71C5, which plays an important role in ABA homeostasis by glucosylating ABA to abscisic acid-glucose ester (GE) in Arabidopsis (Arabidopsis thaliana). Biochemical analyses show that UGT71C5 glucosylates ABA in vitro and in vivo. Mutation of UGT71C5 and down-expression of UGT71C5 in Arabidopsis cause delay in seed germination and enhanced drought tolerance. In contrast, overexpression of UGT71C5 accelerates seed germination and reduces drought tolerance. Determination of the content of ABA and ABA-GE in Arabidopsis revealed that mutation in UGT71C5 and down-expression of UGT71C5 resulted in increased level of ABA and reduced level of ABA-GE, whereas overexpression of UGT71C5 resulted in reduced level of ABA and increased level of ABA-GE. Furthermore, altered levels of ABA in plants lead to changes in transcript abundance of ABA-responsive genes, correlating with the concentration of ABA regulated by UGT71C5 in Arabidopsis. Our work shows that UGT71C5 plays a major role in ABA glucosylation for ABA homeostasis.
Climate change is now affecting global agriculture and food production worldwide. Nonetheless the direct link between climate change and food security at the national scale is poorly understood. Here we simulated the effect of climate change on food security in China using the CERES crop models and the IPCC SRES A2 and B2 scenarios including CO2 fertilization effect. Models took into account population size, urbanization rate, cropland area, cropping intensity and technology development. Our results predict that food crop yield will increase +3-11 % under A2 scenario and +4 % under B2 scenario during 2030-2050, despite disparities among individual crops. As a consequence China will be able to achieve a production of 572 and 615 MT in 2030, then 635 and 646 MT in 2050 under A2 and B2 scenarios, respectively. In 2030 the food security index (FSI) will drop from +24 % in 2009 to −4.5 % and +10.2 % under A2 and B2 scenarios, respectively. In 2050, however, the FSI is predicted to increase to +7.1 % and +20.0 % under A2 and B2 scenarios, respectively, but this increase will be achieved only with the projected decrease of Chinese population. We conclude that 1) the proposed food security index is a simple yet powerful tool for food security analysis; (2) yield growth rate is a much better indicator of food security than yield per se; and (3) climate change only has a moderate positive effect on food security as compared to other factors such as cropland area, population growth, socio-economic pathway and technology development. Relevant policy options and research topics are suggested accordingly.
Protection is not conferred by preexposure to Norwalk virus (NV). By use of an ELISA with baculovirus-expressed recombinant NV (rNV) capsid protein, the pattern of NV fecal shedding and the protective effect of rNV-specific fecal IgA (flgA) were investigated in volunteers who were repeatedly challenged with NV. After the first challenge, ill volunteers were significantly more likely than well volunteers to have NV antigen in their stool (P < .05). After challenge, antigen shedding was detected on days 1-13; ill volunteers shed the antigen longer (P = .02). A higher prechallenge rNV-specific flgA geometric mean titer was found in ill compared with well volunteers (P < .05) and in infected versus noninfected volunteers (P < .05). NV shedding was common after infection and was present up to 2 weeks after challenge. Preexisting rNV-specific flgA, like serum IgG, is not protective and may be a marker for symptomatic disease.
Dynamics driving the El Niño–Southern Oscillation (ENSO) over longer-than-interannual time scales are poorly understood. Here, we compile thermocline temperature records of the Indo-Pacific warm pool over the past 25,000 years, which reveal a major warming in the Early Holocene and a secondary warming in the Middle Holocene. We suggest that the first thermocline warming corresponds to heat transport of southern Pacific shallow overturning circulation driven by June (austral winter) insolation maximum. The second thermocline warming follows equatorial September insolation maximum, which may have caused a steeper west-east upper-ocean thermal gradient and an intensified Walker circulation in the equatorial Pacific. We propose that the warm pool thermocline warming ultimately reduced the interannual ENSO activity in the Early to Middle Holocene. Thus, a substantially increased oceanic heat content of the warm pool, acting as a negative feedback for ENSO in the past, may play its role in the ongoing global warming.
International audienceThe depth of equatorial Pacific thermocline is diagnostic of the main modes of tropical climates. Past estimates of Pacific thermocline dynamics have been reconstructed either for the Last Glacial Maximum or on longer timescales at low resolution. Here we document a new high-resolution set of reconstructed past sea surface and subsurface waters temperatures from the southwestern subequatorial Pacific, core MD05-2930, in the Gulf of Papua, over the last 800ka. We used two morphotypes of Globigerinoides ruber known to live at different water depths to reconstruct past stratification. We estimated calcification temperature of each morphotypes by Mg/Ca paleothermometry. Our subequatorial Pacific thermocline paleotemperature record indicates a response of the thermocline to both direct orbital forcing and glacial-interglacial changes. Our stratification record shows a systematic shallower glacial thermocline, whereas sea surface temperatures are characterized by precessional forcing. The record is indicative of a progressive long-term shoaling of the thermocline during the glacial stages during the late Pleistocene. The shoaling of the subequatorial Pacific thermocline is consistent with regional estimates. An enhanced South Pacific shallow overturning wind-driven circulation could have driven this progressive shoaling. We speculate that this late Pleistocene glacial shoaling of the thermocline could be related to an increase in the amplitude of the obliquity
