AimThe aim of this study was to evaluate the effect of telmisartan (TELM) on inflammation, oxidation and the expression of matrix metalloproteinases (MMPs) and the expression RANKL/RANK/OPG in the periodontal tissue of a rat model for ligature-induced periodontitis.Materials and methodsMale Wistar albino rats were randomly divided into five groups of 10 rats each: (i) non-ligated, given water; (ii) ligated, given water; (iii) ligated, given 1 mg/kg TELM; (iv) ligated, given 5 mg/kg TELM; and (v) ligated, given 10 mg/kg TELM. All groups were treated with saline or TELM for 10 days. Periodontal tissue was analysed by histopathology; by the immunohistochemical examination of COX-2, MMP-2, MMP-9 and the RANKL/RANK/OPG pathway; and by ELISA analysis of the levels of IL-1β, IL-10, TNF-α, myeloperoxidase (MPO), malonaldehyde (MDA) and glutathione (GSH).ResultsTreatment with 10 mg/kg TELM resulted in reduced concentrations of MPO, MDA (p < 0.05) and the pro-inflammatory cytokine IL-1β (p < 0.05); reduced expression of MMP-2, MMP-9, RANK, RANKL and COX-2; and an increase in OPG. The levels of TNF-α were significantly reduced in all TELM-treated groups.ConclusionsThese findings confirm the involvement of TELM in reducing the inflammatory response, oxidative stress and bone loss in ligature-induced periodontitis in rats.
Oral mucositis (OM) is a common complication of treatments for head and neck cancer, particularly radiotherapy with or without chemotherapy. OM is characterised by oral erythema, ulceration, and pain. The aim of this study was to evaluate the effect of azilsartan (AZT), an angiotensin II receptor antagonist, on 5-fluorouracil (5-FU)-induced oral mucositis (OM) in Syrian hamsters. OM was induced by the intraperitoneal administration of 5-FU on experimental days 1 (60mg/Kg) and 2 (40mg/Kg). Animals were pretreated with oral AZT (1, 5, or 10 mg/kg) or vehicle 30 min before 5-FU injection and daily until day 10. Experimental treatment protocols were approved by the Animal Ethics Committee Use/CEUA (Number 28/2012) of the UFRN. Macroscopic analysis and cheek pouch samples were removed for histopathologic analysis. Myeloperoxidase (MPO), Malonyldialdehyde (MDA), interleukin-1 beta (IL-1β), interleukin-10 (IL-10), and tumour necrosis factor-alpha (TNF-α) were analysed by Enzyme Linked Immuno Sorbent Assay (ELISA). Vascular endothelial growth factor (VEGF), fibroblast growth factor (FGF), keratinocyte growth factor (KGF), and transforming growth factor (TGF)-α were measured by immunohistochemistry. Analysis of variance followed by Bonferroni’s test was used to calculate the means of intergroup differences (p ≤ 0.05). Treatment with 1 mg/kg AZT reduced levels MPO (p<0.01), MDA (p<0.5) and histological inflammatory cell infiltration, and increased the presence of granulation tissue. AZT treatment at 1 mg/kg reduced the TNF-α (p<0.05) and IL-1β (p<0.05) levels, increased the cheek pouch levels of IL-10 (p<0.01), and upregulated VEGF, FGF, KGF, and TGF-α. Administration of AZT at higher doses (5 and 10 mg/kg) did not significantly reverse the OM. AZT at a dose of 1 mg/kg prevented the mucosal damage and inflammation associated with 5-FU-induced OM, increasing granulation and tissue repair.
The aim of this study is to determine the effects of Atorvastatin treatment, an inhibitor of 3-hydroxy-3-methylglutaryl-coenzyme A (HMG-CoA) reductase, in periodontal disease. Male Wistar albino rats were randomly divided into five groups of ten rats each: (1) non-ligated treatment (NL), (2) ligature only (L), (3) ligature plus 1 mg/kg Atorvastatin daily for 10 days, (4) ligature plus 5 mg/kg Atorvastatin daily for 10 days, and (5) ligature plus 10 mg/kg Atorvastatin daily for 10 days. Following the treatment course, the periodontal tissue of the animals was analyzed by Measurement of alveolar bone loss, Histopathology and immunohistochemistry to determine of the expression of COX-2, MMP-2, MMP9, and RANKL/RANK/OPG. ELISA assay was used to quantitate the levels of IL-1β, IL-10, TNF-α, myeloperoxidase, malondialdehyde, and glutathione. The periodontal group treated with 10 mg/kg of Atorvastatin (3.9±0.9 mm; p<0.05) showed reverse the alveolar bone loss caused Experimental Periodontal Disease compared to (L) (7.02±0.17 mm). The periodontal group treated with 10 mg/kg of Atorvastatin showed a significant reduction in MPO and MDA (p<0.05) compared to ligature only group (L). Similarly in this group, the levels of the proinflammatory cytokines IL-1β and TNF-α were significantly decreased (p<0.05). Furthermore, MMP-2, MMP-9, RANKL/RANK, and COX-2 were all downregulated by Atorvastatin treatment, while OPG expression was increased. The findings support a role of Atorvastatin for reducing the bone loss, inflammatory response, oxidative stress, and expression of extracellular matrix proteins, while reducing RANK/RANKL and increase OPG in periodontal disease.
O objetivo do presente trabalho foi analisar os conhecimentos e atitudes de gestantes da cidade de Natal/RN, sobre sua saúde bucal durante a gestação e sobre os cuidados à saúde bucal que se deve ter com o bebê. No período de outubro de 2013 até março de 2014, foi aplicado um questionário adaptado para gestantes, com idade entre 18 e 39 anos, de 04 Unidades de Saúde da família, informações sobre a escolaridade materna, aspectos básicos dos cuidados do pré-natal, bem como dos cuidados com a saúde oral e, por último, aspectos acerca dos cuidados odontológicos para com os bebês. Os dados foram analisados pelo teste X2 com nível de significância de 5%, dos testes Mann-Whitney e Kruskal-Wallis. Foi utilizada ainda a frequência absoluta e percentual para descrição das variáveis socioeconômicas e demográficas. Nesse estudo, prevaleceram mães com doze ou mais anos de estudo (40%). Foi verificado que a maioria (93,8 %) apresentou uma renda familiar menor ou igual a três salários mínimos e que quando foi realizada a pesquisa 84,6% já tinham realizado 06 ou mais consultas pré-natais. As gestantes não direcionaram o conhecimento da saúde bucal, à importância dada à visita ao dentista durante a gravidez, considerando que dentre as que têm 12 anos ou mais de estudo, em sua maioria (15,4%), não sabiam desta capacidade (p˂0,5). As mulheres que passaram por orientações sobre a amamentação, sobre a importância no aleitamento materno, e sobre hábitos bucais como o uso de chupeta ou sucção digital são prejudiciais (p˂0,5). Considerando os cuidados que relataram adotar com a saúde bucal e a do bebê, conclui-se que as gestantes não demostraram preocupação com os cuidados à saúde bucal, evidenciado pela falta de conhecimento.
O Transtorno do Espectro Autista (TEA) é caracterizado pela dificuldade na linguagem falada e interação social. A condição bucal do paciente com TEA é de altos índices de placa dental, explicados pelas dificuldades na realização de higiene bucal, aumentando também os índices de cárie e doença periodontal. O que mostra a importância de um tratamento odontológico eficaz, porém se trata de um dos maiores desafios para os cirurgiões dentistas, devido às suas manifestações clinicas complexas e variadas. O objetivo desse estudo é ressaltar a importância de uma abordagem eficaz e segura por parte do cirurgião dentista, para obter a colaboração do paciente com TEA durante o atendimento odontológico não farmacológico. O estudo consiste em uma revisão de literatura narrativa, e foram realizadas buscas nas bases de dados bibliográficos; Pubmed, BVS (Biblioteca virtual em saúde) e Lilacs (Literatura Latino-Americana e do Caribe em Ciências da saúde) nos anos de 2012 a 2021. Com os resultados observou-se que, para a realização de um bom atendimento, inicialmente é necessário conseguir a colaboração do paciente, com métodos e técnicas educativas e lúdicas, ensinando e estimulando a fazer sua higiene oral sempre prezando a prevenção. Desse modo é possível concluir-se que, é importante realizar o atendimento odontológico precocemente, desde a primeira infância para uma melhor adaptação por parte do paciente autista e assim obter-se-á maior cooperação na manutenção da sua saúde bucal dentro de suas limitações.
