Bovine cysticercosis is a zoonotic infection widely spread throughout Brazil, creating a burden on hygiene maintenance and the economy. Diagnosis of cysticercosis usually relies on post mortem inspection of carcasses in slaughterhouses. This detection method provides only low sensitivity. Recent advancements have improved the performance of serologic tests, such as ELISA, providing greater sensitivity and specificity. The objective of the current study was to identify and evaluate a synthetic peptide derived from the Taenia saginata 18kDa oncospheric surface protein for the diagnosis of bovine cysticercosis in ELISA. Test performance of the identified peptide was compared to an ELISA based on a heterologous crude Taenia crassiceps antigen (Tcra), widely used for the sero-diagnosis of bovine cysticercosis. Based on the primary sequence of an in silico structural model of the 18kDa protein, an epitope region designated EP1 was selected (46-WDTKDMAGYGVKKIEV-61). The peptide derived from this region yielded 91.6% (CI=80-96%) sensitivity and 90% (CI=82-95%) specificity when used in an ELISA, whereas the crude antigen yielded 70% (CI=56-8%) sensitivity and 82% (CI=73-89%) specificity. Thus, we conclude that EP1 has higher diagnostic potential for detecting bovine cysticercosis than the crude antigen Tcra.
Cysticercosis affects cattle, which develop the larva of Taenia saginata after ingesting their eggs, which are released by humans. It is a parasite that is distributed worldwide. The use of geoprocessing allows a better definition of endemic areas and populations that face different levels of risk and thus creates a basis for specific control programs for each farming system. The objective of this study was to evaluate the epidemiological situation of bovine cysticercosis in four municipalities in the Triângulo Mineiro region (Romaria, Iraí de Minas, Grupiara and Douradoquara), with special attention given to the influence of the hydrographic network on the spatial distribution of the disease. The properties sampled (68) for laboratory and epidemiological analysis (questionnaire) were also georeferenced. The risk factors were analyzed using the ArcGIS 10.1 program. The animals were submitted to immunodiagnostic tests (enzyme-linked immunosorbent assay and immunoblot) for the diagnosis of bovine cysticercosis. After confirming the disease focus properties, the spatial distribution of the cases was analyzed in relation to the watershed of the region. The positive results for bovine cysticercosis on the properties of the municipalities of Romaria and Iraí de Minas were grouped together, which was a significant result (p < 0.01). Although the positive results for bovine cysticercosis in the municipalities of Grupiara and Douradoquara were randomly distributed (p = 0.27), the risk factor that proved to be statistically significant in the regression analysis for cysticercosis transmission was the water source from the river/ stream. From the analysis of the map based on the hydrography of the region, it can be observed that for the distribution of positive cases for bovine cysticercosis in the properties, the main risk factor was the interference of the rivers and their tributaries that fed the municipalities sampled. This study allowed the identification of the prevalence of the taeniasis-cysticercosis complex in the properties of the sampled municipalities (53.84% in Romaria, 45.45% in Grupiara, 37.03% in Iraí de Minas and 35.29% in Douradoquara). ResumoO complexo teníase-cisticercose é uma zoonose de grande importância para a saúde pública e animal. O ser humano desempenha importante papel sendo o único hospedeiro definitivo do parasita. Albergando no intestino as formas adultas da Taenia saginata ou da Taenia solium, cabendo à forma larvar (cisticerco) o desenvolvimento na musculatura de bovinos e suínos ou do próprio homem, quando este também atua como hospedeiro intermediário. Visando obter maior conhecimento sobre esta zoonose, o objetivo do presente estudo foi determinar o perfil epidemiológico do complexo teníase-cisticercose na zona rural do município de Matias Barbosa, localizado na Zona da Mata de Minas Gerais. Para tanto, foram realizadas coletas de sangue de 350 bovinos e 81 suínos e amostras de fezes de 124 pessoas da população presente em 74 propriedades rurais. Além disso, foi aplicado um questionário epidemiológico sobre as condições sócio-econômicas e sanitárias de cada propriedade. Observou-se uma prevalência de 0,9% de bovinos positivos ao teste de Immunoblot. Não houve suínos positivos, assim como também não foi diagnosticada pessoa portadora de teníase. Alguns fatores de risco de transmissão do complexo teníase-cisticercose no município foram identificados como o consumo de carne bovina e suína mal aquecida, propriedades sem saneamento básico e criação de bovinos com acesso a fontes de contaminação. Apesar da baixa prevalência da cisticercose bovina encontrada no município de Matias Barbosa, é necessária a tomada de medidas de controle do complexo teníase-cisticercose para impedir que haja aumento de casos da doença no município, tendo em vista que foram registrados fatores favoráveis à sua manutenção. Esta pesquisa teve como objetivo avaliar as características epidemiológicas do complexo teníase-cisticercose na zona rural do município de Divinésia-MG. Para tanto, realizou-se coleta de 243 amostras de fezes humanas, 435 amostras de soro bovino e 56 de soro suíno. Além disso, foi aplicado questionário para coleta de informações sobre as características socioeconômicas e higiênico-sanitárias da população amostrada. Não foram encontrados animais positivos ao teste ELISA indireto e Immunoblot para cisticercose suína e bovina, nem ovos de Taenia sp. em amostras de fezes humanas. Verificou-se que, apesar da população estudada possuir renda mensal baixa, as famílias residiam em casas bem estruturadas, com banheiro e rede de esgoto, realizavam tratamento antiparasitário de rotina, não consumiam carne malpassada, e vermifugavam os bezerros. Essas características contribuem para o controle do complexo teníase-cisticercose, justificando os resultados obtidos neste estudo.
ResumoA cisticercose bovina é uma infecção zoonótica causada pelo estágio larval da Taenia saginata. A infecção é favorecida pela contaminação ambiental provocada pelo próprio homem, hospedeiro da forma adulta do parasito e por condições inadequadas de manejo. O conhecimento da prevalência da cisticercose bovina é importante para a implantação de programas de controle, para isto é fundamental determinar a fonte de infecção dos animais. Neste trabalho, foi realizado estudo a partir dos registros da cisticercose de três frigoríficos localizados no município de Colatina-ES. Por meio da guia de trânsito animal (GTA) foram obtidos os endereços das propriedades que destinaram animais ao abate. Tais propriedades foram georreferenciadas para o mapeamento da região estudada e estudo da distribuição dos casos e controles. Os fatores de risco foram estudados utilizando as informações obtidas em inquérito epidemiológico baseado na aplicação de questionários nas propriedades visitadas, utilizando como possíveis fatores de risco associados à infecção: o tamanho das propriedades, a origem dos animais, a idade dos animais adquiridos e o destino do esgoto. O diagnóstico da cisticercose no frigorífico, somado a informação de origem do animal, possibilitou definir as áreas de ocorrência da doença, demonstrando ser um importante indicador para o estabelecimento de programas eficientes para combate ao parasita. Knowing the prevalence of bovine cysticercosis is important for the implementation of control programs, however it is important to determine the source of the infection of the animals. In this paper, a study was conducted based on the records of three slaughterhouses located in the town of Colatina-ES, the addresses of properties, that sent animals to slaughtering, were obtained through the Animal Transit Guide (GTA). The properties were georeferenced for the mapping of the studied region and analysis of the distribution of cases and controls. Risk factors were studied using the information obtained in an epidemiological survey based on questionnaires application at the properties visited, being the size of the properties, the origin of animals, age of animals acquired and sewer destination, the main factors associated to the infection. The diagnosis of cysticercosis in the slaughterhouse, plus the information of the animal source, allowed defining the areas of disease occurrence and its quantification. Proving to be an important indicator for the establishment of effective programs to control the parasite.
