Nattokinase là enzyme làm tan huyết khối, được phát hiện trong Natto vào năm 1980. Trong bài báo này, hai chủng Bacillus sp.7.2 và Bacillus sp.NP3 chọn lọc được, có khả năng sinh tổng hợp mạnh enzyme làm tan huyết khối (nattokinase). Trên môi trường hạt đậu nành, chiều dày môi trường 2,5 cm, nhiệt độ phòng, sau 40 giờ lên men, hai chủng Bacillus sp.7.2 và Bacillus sp.NP3 sinh ra nattokinase (470 FU/g), chiếm khoảng 70-85% so với hoạt tính protease tổng. Kết quả này cho thấy rằng, chúng ta có thể chủ động sản xuất sản phẩm nattokinase hoạt tính cao, giá thành thấp để vệ sức khỏe người dân, mà không phụ thuộc vào thực phẩm chức năng chứa enzyme nattokinase ngoại nhập. The supply of non-renewable energy sources from fossil fuel is depleting day by day and it has become one of the greatest challenges to find the alternative sources. Bioethanol as an alternative biofuels has stimulated the worldwide interest. The use of bioethanol as a fuel will minimize the amounts of fossil-derived carbon dioxide (CO2) to the Earth's atmosphere. Yeast is the most acceptable of all organisms for ethanol production because of its diverse substrate specificity and ease of production of ethanol. Temperature is one of the most important environmental factors affecting microbial activity and production cost. The main objective of this research work was to isolation, characterization and process optimization of ethanol producing thermotolerant yeast strains. In total 5 yeast isolates have been characterized on the basis of morphological and physico-chemical characters. Most of the strains were thermotolerant, ethanol tolerant, pH tolerant, as well as osmotolerant. All the strains were formed psedomycelium under stress condition. They were resistant to chloramphenicol but growth was inhibited in the presence of 1% acetic Acid. The strains C and E showed good Invertase activity and all the strains were capable of fermenting glucose, fructose, sucrose, maltose and amylose among 10 carbohydrates. Ethanol producing capability of the strains was studied using sugarcane molasses as substrate. Productivity was observed at different incubation temperature, pH and reducing sugar concentration. After fermentation at different condition, at temperature 35°C, pH 6.0, reducing sugar concentration 6.0% and shaking condition (115 rpm) found to be optimum for ethanol production by both C and E strains. The C and E strains produced maximum 11.25 % and 13.20% respectively at 48 hours under optimum condition. Pilot scale (5 L) production by both strains was found 9.55% and 11.50% under optimum condition. These strains could be potential for ethanol production from cane molasses in commercial scale.

Hồ Chí Minh TÓM TẮT: Nước thải sau chưng cất cồn (NTSCCC) của nhà máy rượu Bình Tây được sử dụng làm môi trường nuôi cấy giống hỗn hợp vi sinh vật (VSV) có ích tạo chế phẩm sinh học BIO-HR. Chế phẩm BIO-HR chứa mật độ tế bào Bacillus sp. ≥ 10 7 CFU/ml, Lactobacillus sp. ≥ 10 7 CFU/ml, Saccharomyces sp. ≥ 10 6 CFU/ml, pH 4-5, không chứa Coliforms, có mùi thơm và vị chua và bảo quản được trong thời gian trên một tháng. Hiệu quả sử dụng của chế phẩm BIO-HR (2,5 ml Bio-HR/lít nước) cho gà Lương Phượng 3 tuần tuổi uống, sau 8 tuần nuôi, tăng trọng bình quân (kg/con) cao hơn lô đối chứng 30%; hệ số tiêu tốn thức ăn (FCR) thấp hơn lô đối chứng 16%. Vì vậy, các nhà máy sản xuất cồn thay vì tốn chi phí để xứ lý toàn bộ nước thải, có thể tận dụng NTSCCC để sản xuất chế phẩm sinh học giàu VSV có ích, tạo thêm việc làm cho người lao động và tăng lợi nhuận cho nhà máy; giúp người chăn nuôi mua được chế phẩm sinh học giá rẻ và hiệu quả sử dụng cao.Từ khóa: Bacillus, Coliforms, Lactobacillus, Saccharomyces, chăn nuôi, vi sinh vật có ích. MỞ ĐẦUTính trung bình, lượng nước thải của các nhà máy sản xuất ethanol gấp 10 lần thể tích ethanol thành phẩm. Tùy thuộc vào loại nguyên liệu và qui mô sản xuất ethanol mà các nhà máy lựa chọn các phương pháp tái sử dụng và xử lý khác nhau.Nước thải chưng chất cồn từ nguyên liệu tinh bột (bắp, lúa miến, lúa mạch, lúa mì, khoai tây...) có giá trị dinh dưỡng cao, gồm tế bào nấm men tự phân giải, giàu các vitamin nhóm B, protein thô (1-4%, dựa trên chất khô), chất xơ, các axít amin và các chất khoáng [4]. Ở Hoa Kỳ, ethanol được sản xuất từ tinh bột ngô, trung bình 25,4 kg bột ngô sản xuất ra 10,6 lít ethanol, 8,2 kg bã rượu khô và 8,2 kg CO 2 [5]. Chất thải sau chứng cất cồn được ly tâm để tách nước, phần chất rắn khoảng 65% độ ẩm, gọi là bã rượu ướt, hoặc được sấy khô gọi là bã rượu khô, phần chất lỏng được cô đặc và phối trộn lại với bã rượu ướt. Tất cả các loại bã rượu được sử dụng làm thức ăn chăn nuôi [3].Nước thải chưng cất cồn từ tinh bột còn được sử dụng để sản xuất phân bón hữu cơ [6], nhân sinh khối Lactobacillus sakei CY1 [8] và Saccharomyces cerevisiae [5].Công ty cổ phần rượu Bình Tây có dây chuyền công nghệ sản xuất cồn thực phẩm tiên tiến, sử dụng nguyên liệu tinh bột gạo và một phần nhỏ tinh bột khoai mì qui mô 20.000 lít/ngày, NTSCCC (100-200 m 3 /ngày) được cho qua máy lọc khung bản để tách nước, phần chất rắn dạng bột nhão được bán giá rẻ làm thức ăn gia súc, phần nước được cho qua hệ thống xử lý nước thải theo phương pháp sinh học (vốn đầu tư 16 tỉ đồng) đạt tiêu chuẩn loại A trước khi thải ra môi trường. Tuy nhiên, chi phí để vận hành hệ thống xử lý nước thải rất cao (khoảng 35.000 đ/khối nước thải).Vì vậy, mục đích của nghiên cứu này là tận dụng NTSCCC của nhà máy rượu Bình Tây làm môi trường nuôi cấy hỗn hợp VSV có ích, tạo chế phẩm sinh học giá rẻ dùng trong chăn nuôi.
