278HELVETICA CHIMICA ACTA.Sintesi deb prodotto II: 10 g di p-nitrobenzamide dell'estere etilico della fenilserina (I) vengono sciolti in 250 em3 di etanolo puro in un pallone di riduzione. Si niescola la soluzione con la quantitk si catalizzatore (niche1 di Raney) ottenuta scomponendo 12 g di lega. La curva di assorbimento raggiunge il massimo dopo circa due ore. La quantitk di idrogeno assorbita 15 di 1850 em3 pari a1 98,5% della quantith teorica. Verso la fine della reazione si nota nel recipiente la formazione di una sostanza bianca sotto forma di scagliette lucenti. Si filtra a caldo. Dal filtrato per raffreddamento si separa di nuovo la sostanza che raccolta sul filtro ed asciugata presenta un P. -2-tricloroacetamido-1,3-propandiolo. 2,12 g di amina vengono inescolati in un piccolo matraccio con 5 cm3 di estcre etilico dell'acido tricloroacetico. Si scalda per un'ora leggermente su b.m. e si lascia riposare per una settimana. Si aggiungono quindi alla soluzione alcune goccie di acqua. Per aggiunta di etere di petrolio si ottiene una separazione della soluzione in due strati. Per sfregamento si ottiene il cristallo che, raffreddando la soluzione, aumenta sensibilmente. Si raccoglie su filtro e si pesa. Resa 2,l g pari a1 60% della resa teorica. La sostanza si presenta sotto forma di grossi cristalli giallognoli con P. F. 145O. Dopo alcune ricristallizzazioni da etanolo-acqua si ottiene tin prodotto purissimo a P.F. 147-148O. (Letteratiira: 6) 148-150°, 7) 148-149O.j Analisi vcdi tabella. RIASSUNTO.Sono stati sintetizzati alcuni derivati dell'estere etilico della fenilserina, del fenilserinolo e del p-nitrofenilserinolo mediante condensazione delle rispettive amine con cloruri di acidi aromatici ed eterociclici e con esteri di acidi alifatici.

E i n l e i t u n g .I n einer fruheren Mitteilungl) 2, haben wir iiber einige Abwandlungsprodukte des Percains (I) berichtet, bei denen wir die SBureamid-Brucke des Percainmodells umgekehrt hatten (11). Dabei gelang uns aber vorerst nur die Herstellung der 2-Alkoxy-4-(diathylaminoa c e tylamino )-chinolin-Derivate (111). Die Synthese der entsprechenden Diathylamino-p r o p io n y 1 -Verbindungen, speziell des mit dem Percain isomeren 2-n-Butoxy-4-( diathylamino-propiony1amino)-chinolins (11) bereitete infolge der Unbestandigkeit der freien Basen grosse Schwierigkeiten. Die 4-Amino-chinolin-Verbindungen zeigen im allgemeinen die Tendenz, in die ,,Chinoh-imino-Formen (IV) uberzugehen 2, 3). Die leichte Zersetzlichkeit dieser, infolge Beschwerung der Iminogruppe durch einen hoheren Acylrest entstandenen Produkte ist daher erklarlich. Andererseits konnten wir bei den N-( /?-Diathylamino-propiony1)-p-alkoxyaniliden (V) beweisen4), dass hei der Hochvakuumdestillation der freien Basen die DiathylaminoGruppe abgespalten wird unter Rildung des ungesattigten N-( /?-Acryloyl-p-alkovyanilids (VI). Bei der Hochvakuumdestillation der freien Base der Forrnel I1 erhielten wir scharf riechende Zersetzungsprodukte, deren Mikroanalysen weder auf die gewunschte Base noch auf ein durch Zersetzung entstandenes Acryloyl-Derivat hinwiesen. Es ist uns nun gelungen, die mit dem Percain isomere Verbindung I1 ohne Destillation zu reinigen und in das kristalline Hydrochlorid iiberzuf iihren.I m Surf a c a i n (VII), dem y-Pipecolino-propylester der p-Cyclohexyloxy-benzoesaure, ist vor kurzem ein neues, starkes Lokalanastheticum mit relativ geringer Toxizitat in den Arzneischatz eingefuhrt worden5). Es interessierte uns nun, welchen Einfluss der Ersatz der Diathylamino-Gruppe durch den Pipecolino-Rest in unseren Verbindungen der Formel I11 auf die pharmakologischen Eigen-
Volunien XXS, Fasciculus IT (1947). 507Der Analyse nach handelt es sich um ein Anhydrid des 2-Desoxy-d-gulose-3-methylathers (XX), das wahrscheinlich erst beim Trocknen unter Verlust von 1 Mol H,O entstanden ist. Vgl. die entsprechende Ueobachtung beim 2-Desoxy-d-allose-3-methylatherl).I ( -) -M e t h o x y -h e r n s t e i n s a u r e (XXI) a u s (XX). Die Hauptmenge des oben erhaltenen (XX) (ca. 380 mg) in 3 em3 Wasser wurde unter Durchleiten von CO, t,ropfenweise innert 2 Std. mit der Losung von 1,55 g KMnO, in 30 om3 Wasser versetzt und 16 St'd. in1 Dunkeln stehen gelassen. Die Aufarbeitung erfolgte genau nach der Vorschrift von Shoppee und Beicksteinh). Die Fraktionierung des entstandeuen Estergemisches bei 12 mm lieferte44 mg vorn Sdp. 30-70" 11 mg vom Sdp. 70--90° und 10 mg Ruckstand Die beiden Destillate wurden gotrennt mit je der 10-fachen Menge einer bei 0" gesiittigten Losung von trockenem NH3 in Methanol versetzt und 48 Std. gut verschlossen stehen gelassen. Hierauf wurde eingedampft und hei 0,005 mm und 10&160° Badtemperatur im Molekularkolben sublimiert. Aus der hohersiedenden Fraktion konnte keine kryst. Substanz gewonnen werden. Die tiefersiedend: Fraktion gab 14 mg Sublimat, das nach zweimaligem Unikrystallisieren aus Methanol-Ather 6 mg Diamid von (XXI) vom Smp. 183-185O lieferte. Die spez. Drehung betrug [~]2 = -51,7" & 6 O (c = 0,483 in Methanol). 4,858 ing Subst. zu 1,0053 mi3; 1 : ; = 1 dm; a : =-0,25" 5 0,03" h'hoppee nnd Reichsteinh) fanden fur I( -)-Methoxy-bernsteinsiure-diamid [a]1,8 = -57,2O & 2 O (c = 0,979inMethanol), Jeonloz, Prinsund Reichsteinl) [a]: = -55,S0 rt: 2" (c = 0,986 in Nethanol). Kach erneuter Sublimation im Hochvakuurn und Umkrystallisieren aus Met'hanol-Ather wurden feine Prismen vom Smp. 185-186O erhalten. Die bfischprobe mit aus Cymarose bereitetem Diamid vom Smp. 183-185Oh) schmolz bci 183--186O, diejenige mit d( + )-Methoxy-bernsteinsiure-diamid (Smp. 184-185O)") bei 180-182O.Die Mikroanalysen wurden teils im mikroanalytiscllen Laboratorium der Eidgen.

A. E i n l e i t u n g . Unter den Chinolin-Verbindungen befinden sich die starksten bis heute entdeckten Lokalanasthetica. Hinsichtlich Wirkungs-gonzentration und Anasthesie-Dauer iibertreffen sie die Lokalanasthetica der andern Stoffklassen. Andererseits haben aber ihre starken toxischen Nebenwirkungen dazu gefuhrt, dass his heute das P e r c a i n (III),) als einziges Chinolin-Derivat in den Arzneischatz eingefuhrt worden ist. Es stellt sich daher die Aufgabe, neue und weniger giftige Chinolin-Derivate mit gleicher Wirkung wie das Percain zu finden. Nach der Entdeekung des synthetischen Antimalariamittels P 1 a s m o chin (I) 3), das nebenbei auch lokalanasthetische Wirkung besitzt, stellte P O U~Y W~U~) eine Reihe lokalanasthetisch wirksamer Slkoxy-amino-chinolin-Derivate her. Nach mehrfacher Veranderung der Athergruppe und der basischen Blkylamin-Kette in der Plasmochin-Molekel, gelangte dieser Porseher zum ,,Produkt 665" (II), tlem 6-Athoxy-8-( 3'-diathylamino-2', 2'-dirnethgl-propyl-amino)-chinolin. Es zeigte bei der pharmakologischen Prufung5) von allen Homologen dieser Gruppe die stkkste Wirkung, die, bei geringerer Toxizitat, sogar diejenige des Percains ubertraf. iViescher6) beobachtete pine starke Abhangigkeit der lokalanasthetisehen Wirksamkeit von der Grosse des Ather-Restes bei der homologen Reihe der Percain-Gruppe. Die Wirksamkeit stieg dabei vom Methoxy-bis zum Butoxy-Ilerivat stark an, urn bei weiterer Vergrosserung des Ather-Restes wieder allmahlich abzunehmen. Ganz ahnliche Zusammenhange konnten in der Folge auch von andern Forschern bei einer grossen Zahl verschiedenartiger Stoffgruppen beobachtet werden. Wojahn') wollte die Wirkung des Percains ditrch Erhohung der Basizitat steigern und ersetzte die CO-Gruppe in der Saurearnidbrucke durch die CH,-Gruppe. Dieses Derivat IV war aber unwirksam; es hatte die optimale Basizitat bereits uberschritten. Ein Zwischenprodukt dieser
