Biodegradation is a method of effectively removing petroleum hydrocarbons from the natural environment. This research focuses on the biodegradation of aliphatic hydrocarbons, monoaromatic hydrocarbons such as benzene, toluene, ethylbenzene, and all three xylene isomers (BTEX) and polycyclic aromatic hydrocarbons (PAHs) as a result of soil inoculation with a biopreparation A1 based on autochthonous microorganisms and a biopreparation A1 with the addition of γ-PGA. The research used biopreparation A1 made of the following strains: Dietzia sp. IN133, Gordonia sp. IN138 Mycolicibacterium frederiksbergense IN53, Rhodococcus erythropolis IN119, Rhodococcus sp. IN136 and Pseudomonas sp. IN132. The experiments were carried out in laboratory conditions (microbiological tests, respirometric tests, and in semi-technical conditions (ex-situ prism method). The biodegradation efficiency was assessed on the basis of respirometric tests, chromatographic analyses and toxicological tests. As a result of inoculation of AB soil with the biopreparation A1 within 6 months, a reduction of total petroleum hydrocarbons (TPH) (66.03%), BTEX (80.08%) and PAHs (38.86%) was achieved and its toxicity was reduced. Inoculation of AB soil with the biopreparation A1 with the addition of γ-PGA reduced the concentration of TPH, BTEX and PAHs by 79.21%, 90.19%, and 51.18%, respectively, and reduced its toxicity. The conducted research has shown that the addition of γ-PGA affects the efficiency of the biodegradation process of petroleum pollutants, increasing the degree of TPH biodegradation by 13.18%, BTEX by 10.11% and PAHs by 12.32% compared to pure biopreparation A1.
Biodegradation is one of the most effective and profitable methods for the elimination of toxic polychlorinated biphenyls (PCBs) and total petroleum hydrocarbons (TPH) from the environment. In this study, aerobic degradation of the mentioned pollutants by bacterial strains Mycolicibacterium frederiksbergense IN53, Rhodococcus erythropolis IN129, and Rhodococcus sp. IN306 and mixed culture M1 developed based on those strains at 1:1:1 ratio was analyzed. The effectiveness of individual strains and of the mixed culture was assessed based on carried out respirometric tests and chromatographic analyses. The Rhodococcus sp. IN306 turned out most effective in terms of 18 PCB congeners biodegradation (54.4%). The biodegradation index was decreasing with an increasing number of chlorine atoms in a molecule. Instead, the Mycolicobacterium frederiksbergense IN53 was the best TPH degrader (37.2%). In a sterile soil, contaminated with PCBs and TPH, the highest biodegradation effectiveness was obtained using inoculation with mixed culture M1, which allowed to reduce both the PCBs (51.8%) and TPH (34.6%) content. The PCBs and TPH biodegradation capacity of the defined mixed culture M1 was verified ex-situ with prism method in a non-sterile soil polluted with aged petroleum hydrocarbons (TPH) and spent transformer oil (PCBs). After inoculation with mixed culture M1, the PCBs were reduced during 6 months by 84.5% and TPH by 70.8% as well as soil toxicity was decreased.
The article presents the issues related to the determination of colored fluorescent tracers such as fluorescein, eosin yellowish, rhodamine B and uranine in reservoir waters by spectrophotometric method. For this purpose, the influence of the pH of the solution on the absorption spectra of the tested tracers was checked. Test results show that fluorescein, rhodamine B and uranine are sensitive to changes in the buffer pH, therefore it is advisable to use stable tracer solutions as well as to control and possibly correct pH in further tests. As part of the study, calibration curves of fluorescein, eosin yellowish, rhodamine B and uranine in distilled water, reservoir water A4 and highly sulfated reservoir waters A5 and A6 were plotted and the analytical methods were validated. Analytical validation included determination of linearity, standard deviation and relative standard deviation of the tested tracers solutions. High values of the regression parameters (0.9927–0.9998) of the analyzed tracers prove a good linear fit, while low values of standard deviation and relative standard deviation prove its repeatability and precision. Particular attention was paid to testing the stability of colored fluorescent tracers in highly sulfated reservoir waters. For this purpose, solutions of the tested tracers were prepared at concentrations of 10 mg/dm3 in distilled water, A4 reservoir water and highly sulfated A5 and A6 reservoir waters. Measurements of the tested tracers in the prepared solutions were performed every 2 days over the period of 1 month. The test results show that fluorescein, eosin yellowish, rhodamine B and uranine solutions are stable in the distilled water and A4 reservoir water, while they degrade in the A5 and A6 reservoir waters. Fluorescein and uranine turned out to be the most sensitive, as they degraded completely in the A6 reservoir water after 20 (fluorescein) and 22 (uranine) days. Yellowish eosin and rhodamine B turned out to be slightly more stable in highly sulfated reservoir waters, as they degraded completely in the A6 reservoir water after 24 days.
Obecność różnorodnych zanieczyszczeń w odpadach przemysłowych oraz możliwość zanieczyszczenia wód powierzchniowych i podziemnych oraz gleby substancjami toksycznymi, które nie są wymieniane w obowiązujących wykazach środków toksycznych, stwarza konieczność przeprowadzania badań własności toksycznych gleby, wód i odpadów z wykorzystaniem testów toksykologicznych bezpośredniego kontaktu. Omówiono badania zmian toksyczności zachodzących podczas procesów bioremediacji zastarzałych odpadów wiertniczych z dołu urobkowego. Przedstawiono także wyniki badań toksyczności płynów do hydraulicznego szczelinowania i uzyskiwanych płynów pozabiegowych. Przeprowadzone próby zastosowania nowoczesnych testów toksykologicznych (Microtox, MARA, test Amesa, testy typu toxkit) w przemyśle naftowym dowodzą, że uzyskane wyniki mogą stanowić podstawę do oceny zagrożeń stwarzanych dla środowiska oraz być pomocne przy wyborze metod zapobiegania skażeniom oraz zagospodarowania różnorodnych materiałów i odpadów generowanych przez przemysł. Application of toxicological tests in oil industry The presence of various pollutants in industrial waste and the possibility of surface and underground waters, and soil contamination with unknown substances (not listed as toxic substances) creates the necessity to examine soil, water and waste toxic properties with the use of toxicological tests of direct contact. Described are studies of soil toxicity changes during bioremediation of old drilling wastes. The data of toxicity tests of hydraulic fracturing fluids and flowback waters is also presented. Practical application of innovative toxicological tests (Microtox, MARA, AMES, and toxkit-type tests) carried out in the oil industry, have proven, that the data can be used to assess the environmental threat. They can also be helpful in the selection of methods for preventing contamination and management of different materials and waste generated by oil industry.
W artykule opisano proces opracowania metodyki jakościowego i ilościowego oznaczania węglowodorów monoaromatycznych, do których należą benzen, toluen, etylobenzen i izomery ksylenu (BTEX), w próbkach gleby. Optymalizacja metodyki polegała na wykonaniu szeregu analiz wzorcowych i rzeczywistych próbek gleb skażonych jednopierścieniowymi węglowodorami aromatycznymi. Do oznaczania węglowodorów z grupy BTEX zastosowano metodę chromatografii gazowej z ekstrakcją do fazy nadpowierzchniowej, która pozwala w szybki i prosty sposób uzyskać informację o zawartości BTEX w próbkach środowiskowych, z pominięciem długotrwałego etapu przygotowania próbki. Opracowana metodyka została przetestowana na rzeczywistych próbkach gleb pobranych z terenów kopalń ropy naftowej, na których występowały doły urobkowe zawierające zastarzały odpad wiertniczy zanieczyszczony substancjami ropopochodnymi. Opracowaną metodykę wykorzystano również w kontroli przebiegu procesu bioremediacji substancji ropopochodnych w glebie (ex situ).
STRESZCZENIE: W artykule przedstawione zostały zagadnienia związane z opracowaniem metodyki chromatograficznego oznaczania polichlorowanych bifenyli w próbkach gleb. Do tego celu stosowano chromatograf gazowy wyposażony w detektor wychwytu elektronów (GC-ECD) firmy Perkin Elmer. W ramach prowadzonych badań określono optymalne parametry pracy chromatografu, takie jak: temperatura inżektora, temperatura detektora ECD, przebieg temperaturowy pieca w trakcie analizy chromatograficznej, czas analizy oraz dobrano kolumnę chromatograficzną umożliwiającą selektywny rozdział analitów. Następnie przeprowadzono kalibrację układu chromatograficznego i walidację metody analitycznej oznaczania polichlorowanych bifenyli, stosując roztwory wzorcowe firmy Supelco i Sigma-Aldrich o numerach odpowiednio 47330-U oraz 36989. Sporządzono krzywe kalibracyjne dla każdego z badanych kongenerów PCB (PCB-10, PCB-28, PCB-53, PCB-101, PCB-138, PCB-153, PCB-180, PCB-209) w dwóch zakresach stężeń. Walidacja metody analitycznej obejmowała wyznaczenie liniowości, odchylenia standardowego, względnego odchylenia standardowego, granicy wykrywalności (LOD) i granicy oznaczalności (LOQ) wyżej wymienionych kongenerów PCB. W tym celu opracowania metodyki przygotowania próbek gleb do analizy chromatograficznej przetestowano układ pod kątem doboru metody ekstrakcji (ekstrakcja rozpuszczalnikiem wspomagana wytrząsaniem, ekstrakcja z wykorzystaniem ultradźwięków i ekstrakcja w aparacie Soxhleta), doboru rozpuszczalnika/układu rozpuszczalników (n-heksan, mieszanina n-heksanu i dichlorometanu w stosunku 1:1) oraz doboru materiału sorpcyjnego w etapie ekstrakcji do fazy stałej SPE (kolumienki do zadań specjalnych oraz kolumienki z wypełnieniem Florisil). Do badań modelowych wykorzystano próbki gleb zanieczyszczone roztworem wzorcowym firmy Supelco o numerze 47330-U. Dla wszystkich kombinacji wyżej wymienionych warunków przygotowania próbki do analizy chromatograficznej obliczono stopień odzysku poszczególnych analitów. Na podstawie analiz chromatograficznych i stopni odzysku wytypowano najbardziej optymalną metodykę przygotowania próbek gleb do analizy chromatograficznej pod kątem oznaczania polichlorowanych bifenyli. Opracowaną metodykę oznaczania polichlorowanych bifenyli w próbkach gleb przetestowano na rzeczywistych próbkach gleb skażonych zestarzałym olejem transformatorowym pobranych z terenów południowo-wschodniej Polski. Procedura analityczna oznaczania PCB w próbkach gleby obejmowała: wstępne przygotowanie próbek (suszenie, mielenie i homogenizacja), izolację analitów z matrycy próbki (ekstrakcja rozpuszczalnikiem wspomagana wytrząsaniem z zastosowaniem mieszaniny rozpuszczalników n-heksan:dichlorometan w stosunku 1:1), oczyszczanie próbki z substancji przeszkadzających (zestaw dwóch kolumienek do ekstrakcji SPE: Silica gel nr 7086-03 i PCB-A nr 7511-04 firmy Bakerbond), analizę chromatograficzną. W artykule przedstawiono ocenę efektywności biodegradacji zanieczyszczeń ropopochodnych w trakcie etapowego oczyszczania zestarzałego odpadu, który zdeponowano w dole urobkowym G-70. Odpad z dołu urobkowego G-70 był silnie zanieczyszczony substancjami ropopochodnymi (TPH) (14 258÷120 380 mg/kg s.m.) i BTEX (68,9÷92,7 mg/kg s.m). Etapowa technologia oczyszczania odpadów wiertniczych metodą in situ obejmuje: wstępną remediację, modyfikację struktury odpadu, bioremediację podstawową stymulowaną przez wapnowanie i nawożenie (wzbogacanie środowiska odpadu w składniki biogenne) oraz inokulację biopreparatami opracowanymi na bazie autochtonicznych niepatogennych gatunków bakterii, grzybów i drożdży. Rozbudowany monitoring procesów bioremediacyjnych oparty na chromatograficznych analizach zanieczyszczeń ropopochodnych i badaniach parametrów fizyczno-chemicznych oraz mikrobiologicznych (całkowita liczba mikroorganizmów, liczba mikroorganizmów rozkładających węglowodory ropopochodne, liczba grzybów, identyfikacja rodzajowa i gatunkowa bakterii i grzybów oraz aktywność dehydrogenazowa) w pełni pozwala na optymalizację ich przebiegu oraz ocenę efektywności opracowanych biopreparatów. Po przeprowadzeniu w roku dwóch serii inokulacji biopreparatem na bazie autochtonicznych niepatogennych gatunków bakterii (G-70-1) najszybciej zachodziła biodegradacja węglowodorów alifatycznych o długości łańcucha nC 9-nC 20 w zakresie: 47,8÷69,6%, zaś w przypadku węglowodorów o długości łańcucha nC 21-nC 36 w zakresie: 25,7÷51,4%. Uzyskano także obniżenie zawartości BTEX o 53,9%. Wzbogacenie biopreparatu (G-70-1) o autochtoniczne niepatogenne gatunki grzybów i drożdży (G-70-2) oraz przeprowadzenie w II roku oczyszczania trzech serii inokulacji przyczyniły się do wzrostu stopnia biodegradacji węglowodorów z zakresu nC 9-nC 22 o 51,3÷79,5%, węglowodorów cięższych nC 23-nC 36 o 39,6÷54,4% oraz BTEX o 79,8%. Po zakończeniu oczyszczania sumaryczna zawartość węglowodorów ropopochodnych w odpadzie wynosiła poniżej 1000 mg TPH/kg s.m., co pozwoliło na przywrócenie aktywności biologicznej na tym terenie. The waste was strongly contaminated with hydrocarbons-TPH (14 258÷120 380 mg/kg dry mass) and BTEX (68.9÷92.7 mg/kg dry mass).
