The mouse disease model has the advantage of a broad array of immunological and genetic tools available for basic research. Some studies on transgenic and/or mutant mouse strains as models for experimental leptospirosis have been reported; however, the wider use of such models is hampered by a poor understanding of the outcome of experimental leptospiral infection among the different mouse strains available. Here, the outcome of infection by a virulent strain of Leptospira interrogans serogroup Icterohaemorrhagiae strain Cop was studied in four commonly used wildtype mouse strains: A, CBA, BALB/c and C57BL/6. The end points evaluated in this study were survival, presence of kidney lesions, leptospiral load in kidney samples, microscopic agglutination test titre and anti-leptospiral IgG antibody levels. As expected, none of the mouse strains were susceptible to lethal leptospirosis. However, these strains developed specific pathologies associated with sublethal leptospirosis. The A and C57BL/6 strains exhibited a high leptospiral load in kidney samples and the CBA and C57BL/6 strains developed severe inflammatory lesions, whilst the BALB/c strain proved to be the most resistant to subclinical leptospirosis.
The aims of this study were to investigate the frequency of pulmonary hemorrhage (PH) in mice unable to produce functional B and T lymphocytes and to explore the effect of an inducible nitric oxide synthase gene (Inos) knockout (KO) on the frequency/severity of interstitial nephritis in vivo. We studied the outcome of infection by the virulent Leptospira interrogans serovar Copenhageni strain Cop. The animals used were Inos KO mice, recombination activating gene 1 (Rag1) KO mice, CB17 severe combined immunodeficiency (SCID) mice, and the respective wild-type (WT) C57BL/6 and BALB/c controls. The Inos KO and WT mice survived with no clinical symptoms of leptospirosis. The frequency and severity of nephritis was significantly lower in the Inos KO mice. All of the Rag1 KO and SCID animals died of acute leptospirosis, whereas all of the WT mice survived. PH was observed in 57 and 94% of Rag1 KO mice and in 83 and 100% of SCID mice, using inoculum doses of 10 7 and 10 6 leptospires, respectively. There was no evidence of PH in the WT controls. In conclusion, the loss of the Inos gene had a negligible effect on the outcome of leptospiral infection, although we observed a reduced susceptibility for interstitial nephritis in this group. Of note, the absence of functional B-and T-cell lymphocytes did not preclude the occurrence of PH. These data provide evidence that PH in leptospirosis may not be related only to autoimmune mechanisms.
Additionally mailed in biopsy or necropsy skin samples from 31 goats and 45 sheep were studied histologically. Skin diseases were diagnosed in 35 goats (5.33% of all cases in this species) and 45 sheep (13.88% of the cases in this species). The most frequent skin disease was myiasis (10 cases in goats and 7 in sheep), followed by contagious echtyma (8 cases in goats and 2 in sheep), squamous cell carcinoma (4 cases in goats and 5 in sheep), dermatophylosis (8 cases in sheep), and allergic dermatitis (1 case in goats and 1 in sheep). Two cases of pythiosis in sheep, two cases of epidermolysis bullosa in goats, one case of poisoning by Brachiaria brizantha (photosensitization) and another by Leucaena leucocephala (alopecia) in sheep, and one case of papillomatosis, one of pemphigus foliaceous, one of protothecosis and one of rhabdomyosarcoma in goats were also diagnosed. In 4 cases in sheep and 19 in goats the etiologic diagnosis was not obtained. With the information of the occurrence and epidemiology of the skin diseases is possible to determine efficient control measures.
Abstract. A case of protothecosis is reported in an adult goat with inspiratory dyspnea and stertor. Dermatitis with prominent ulcerated nodules up to 3 cm in diameter was observed in the muzzle at the mucocutaneous junctions of nasal and lip skin, and in the border of the pinna. Histologic lesions were necrotizing pyogranulomatous dermatitis and rhinitis with myriads of walled sporangia, characteristic of Prototheca wickerhamii. This seems to be the first report of protothecosis in a goat.
Photosensitization is common in the Brazilian semiarid, affecting mainly horses, but also donkeys, mules, sheep and cattle. The dermatitis affects mainly non pigmented skin, and the animals recover after being withdrawn from the pastures. To demonstrate the etiology of the disease, Froelichia humboldtiana (Roem. et Schult.) Seub., collected in the field one or two times a week and kept in the refrigerator for 1-4 days, was administered for 30 or more days as the only food ad libitum to 2 donkeys and one white sheep, and as the only forage ad libitum, to one white horse. No clinical signs were observed in those animals, and serum activities of aspartate-aminotransferase (AST), alanine-aminotransferase (AST), and gamma-glutamyltransferase (GGT) were within normal ranges. In another experiment, one sheep was tied by a rope, during the day, in a pasture of exclusively F. humboldtiana for a 26 days period. Skin lesions characteristic of photosensitization appeared 4-5 days after the start of the experiment and became severe until day 26. After the withdrawal of the animal from the pasture on day 26 it fully recovered within 30 days. In another experiment, 4 sheep were tied by ropes in the same pasture, during the day, for a 29 days period. Another 4 control sheep were maintained in a neighboring pasture without F. humboldtiana. Skin lesions characteristic of photosensitization were observed after 11-25 days of F. humboldtiana grazing. Serum activities of AST and GGT, and serum levels of bilirubin were within normal ranges. At the end of the 29 days grazing F. humboldtiana, 2 sheep were euthanized, and 2 were withdrawn from the pastures. No gross or histologic liver lesions were observed on the 2 sheep euthanized; the other 2 sheep recovered within 17 and 20 days after the end of grazing. One mare and its foal grazed in the same pasture during 44 days. The mare that had a pigmented skin had no dermatitis, but the foal showed dermatitis on the white areas of the skin after 25 days of grazing. The two horses were withdrawn from the pastures after 44 days grazing, and the foal fully recovered within 15 days. These experiments indicate that F. humboldtiana causes primary photosensitization in domestic animals. The absence of lesions in animals ingesting the plant collected in the field and kept refrigerated suggests that F. humboldtiana lost toxicity after collection. The absence of lesions in the eyes of affected animal, which are characteristic of primary photosensitization caused by furocoumarins containing plants, suggests that F. humboldtiana contains a naphthodianthrone derivate, similar to those found in Fagopyrum esculentum and Hypericum perforatum which do not cause eye lesions.
É relatada a ocorrência de febre catarral maligna (FCM) em bovinos no estado da Paraíba. Descrevem-se seis casos esporádicos da doença, diagnosticados no período de 2000 a 2005, e um surto ocorrido em 1986-1987 no município de Taperoá. Em um caso foi feito o diagnóstico clínico e nos demais casos o diagnóstico foi realizado pela presença de vasculite difusa com degeneração fibrinóide e infiltrado mononuclear perivascular em diversos órgãos. Em 5 casos adoeceu apenas um bovino por rebanho, e em outro adoeceram 2 animais de um rebanho de 6 bovinos. Em 5 dos 6 casos a doença ocorreu no período de maio-agosto coincidindo com o fim da estação chuvosa e época de parição das ovelhas. Os sinais clínicos e achados de necropsia foram característicos da doença. Quatro dos seis animais morreram após um curso clínico de 5-8 dias e um foi eutanasiado após um curso de 40 dias. O surto descrito no município de Taperoá ocorreu de agosto de 1986 a dezembro de 1987, com morbidade de 8,22% e letalidade de 100%. A doença ocorreu após a introdução de um carneiro no rebanho ovino que convivia com os bovinos. O surto foi controlado somente após a eliminação de todos os ovinos da propriedade. Esses resultados demonstram que a febre catarral maligna é endêmica em bovinos na Paraíba, podendo também ocorrer surtos com alta morbidade e letalidade. Além disso, sugerem que, como descrito no Rio Grande do Sul e em outros países, alguns animais podem se recuperar. Os diagnósticos de FCM corresponderam a 3,1% dos 190 casos diagnosticados no Hospital Veterinário no período de 2000-2005, estimando-se que a doença afeta, anualmente, 0,125% dos bovinos no Estado.
Cases of congenital defects (CDs) in cattle diagnosed at the Laboratory of Veterinary Pathology of the Federal University of Santa Maria from 1964 to 2010 were reviewed. During the studied period tissues collected from the necropsy of 7,132 cattle were examined and 31 calves (0.4%) with CDs were found. These CDs were classified into 34 different types and ascribed to the body system primarily affected. [2], and ventricular septal defect [1]); four (7.5%) affected the lymphatic system and consisted of inherited hypoplasia or aplasia of lymphatic vessels and lymph nodes associated with lymphedema. Two cases (3.0%), of hypotrichosis were observed affecting the integument; one case (1.9%) of tracheal stenosis was found in the respiratory system and one case of goiter (1.9%) involved the endocrine system. The results of this survey inidicate that the majority o CDs in cattle in the central Rio Grande do Sul are sporadic; nevertheless their continued study is important for determining the etiology and control.
