Abstract: The aims of this study were to investigate the frequency of pulmonary hemorrhage (PH) in mice unable to produce functional B and T lymphocytes and to explore the effect of an inducible nitric oxide synthase gene (Inos) knockout (KO) on the frequency/severity of interstitial nephritis in vivo. We studied the outcome of infection by the virulent Leptospira interrogans serovar Copenhageni strain Cop. The animals used were Inos KO mice, recombination activating gene 1 (Rag1) KO mice, CB17 severe combined immunodef… Show more

“…Intraperitoneal infectious doses ranging from 10 3 to 10 9 have been used in mice aged 3 to 14 weeks (27)(28)(29)(30). In our study, we determined that ϳ10 8 cells of L. interrogans serovar Copenhageni were needed to establish infection consistently in 100% of the C3H/HeJ mice at 10 weeks of age.…”

“…The use of gerbils [10] and marmoset monkeys [11] in experimental leptospirosis have also been reported. Although mice are considered resistant to Leptospira infection, it has been reported that tlr4 mutant mice (C3H/HeJ) [12,13] and cyclophosphamide-treated mice [14] are susceptible to this infection but frequency and severity of nephritis is lower in Inos knockout mice [15]. However, these mouse models of infection do not mimic the natural susceptibility among rodents.…”

“…μMT mice lack functional B cells. Infection with L. interrogans of Rag −/− , SCID mice, μMT, or nude mice treated with cyclophosphamide results in lethal infection (Adler and Faine 1977;Bandeira et al 2011;Chassin et al 2009;Nally et al 2005). Adler and Faine (1977) clearly demonstrated the importance antibody (and therefore B cells) by injecting cyclophosphamide-treated nude mice with antibody to Leptospira resulting in protection against lethal Leptospira infection.…”
