O bioma Cerrado é fonte de moléculas químicas com grande potencial medicinal. As substâncias extraídas do pequi, fruto do Caryocar brasiliense, têm propriedades antiparasitárias, antioxidantes e antineoplásicas. O osteossarcoma canino é um tumor ósseo altamente agressivo e metastático, pouco responsivo às modalidades atuais de quimioterapia. Este estudo teve como objetivo mostrar os efeitos do extrato etanólico da casca de pequi (EEPP) sobre as células do osteossarcoma canino, além de sugerir uma rota metabólica que explique a ação do extrato. As células D-17 foram semeadas e expostas a EEPP nas concentrações de 0, 10 e 100 µg / mL. Após o tratamento, os núcleos celulares foram marcados com DAPI e quantificados por microscopia de fluorescência. A expressão das proteínas p53, Ki-67, Bcl-2, Akt, AMPK e mTOR foi analisada por imunocitoquímica. Ao usar o DAPI, encontramos uma redução no número de núcleos quantificados, tempo e dose-dependente. A marcação de anticorpos p53, Ki-67 e Bcl-2 diminuiu nos grupos expostos a EEPP. O oposto foi observado com os anticorpos Akt, AMPK e mTOR, onde a proteína não foi encontrada no grupo controle, mas foi expressa nos grupos expostos à EEPP. Nós sugerimos uma possível rota metabólica onde EEPP promove a morte celular e o controle do ciclo celular em células D-17.
Introdução: Em doenças vasculares, a interrupção do fluxo sanguíneo locale subsequente reperfusão de oxigênio pode causar efeitos deletérios e danos irreparáveis às células. Curcuma (Curcuma longa L.) neutraliza radicais livres além de apresentar efeitos preventivos e terapêuticos. Objetivo: Caracterizar os compostos bioativos e a capacidade antioxidante do extrato etanólico de cúrcuma (EEC); avaliar seu efeito nas células endoteliais da veia umbilical humana, e analisar a expressão de vias de sinalização celular. Métodos: As células foram expostas a diferentes concentrações de EEC por 24, 48 e 72 horas. Utilizamos o teste de Folin-Ciocalteau, análise por HPLC-Fluorescência e método DPPH para determinar os compostos fenólicos, conteúdo de curcumina e ação antioxidante, respectivamente; o método de redução de tetrazólio para viabilidade celular, a citotoxicidade e a concentração que inibe 50% da viabilidade celular; e a técnica de imunocitoquímica para analisar a expressão de caspase3, SIRT1 e mTOR. Resultados: Observou-se presença de polifenóis nas classes de ácidos fenólicos e curcuminóides no EEC, com teor de curcumina de 16,7%. A quantidade de antioxidante necessária para reduzir a concentração inicial de DPPH em 50% foi de 18,1 μmol/g. O extrato mitigou o dano celular na dosagem de 100 μg/ml, diminuiu a imunoexpressão da caspase3 e promoveu a sinalização das vias de sobrevivência SIRT1 e mTOR. Conclusão: O EEC teve efeito protetor nas células endoteliais de veia umbilical humana, submetidas ao estresse oxidativo, com diminuição da apoptose (caspase3) em concentrações mais baixas, citoproteção pela manutenção das funções celulares essenciais (mTOR) e sinalização da via de sobrevivência (SIRT1).
O osteossarcoma canino é o tumor ósseo mais comum em cães. Ele apresenta intensa capacidade metastásica e a sobrevida do paciente é baixa nessa doença. A curcumina, o composto mais importante derivado da planta Curcuma longa L., tem sido amplamente estudada e mostrou efeitos antineoplásicos consideráveis contra vários tumores. Este estudo tem por objetivo identificar a ativação de proteínas específicas de vias de apoptose, sobrevivência tumoral e mau prognóstico dessa doença em células OSC da linhagem D-17. Para isso, as células foram cultivadas e tratadas com a curcumina nas concentrações de 20μM, 50 μM e 100 μM em lâminas, que foram preparadas e fixadas. Posteriormente, foi realizada a técnica de imucitoquímica, com os anticorpos anti-caspase3, anti-JNK, anti-AMPK, anti-p53, anti-AKT e anti-mTOR. Foi observado que a curcumina ativou em células de osteossarcoma canino in vitro as proteínas de morte celular caspase-3, JNK e AMPK, reduziu a expressão da proteína p53 mutada e não alterou as proteínas AKT e mTOR. Assim, verificou-se que a curcumina promove apoptose extrínseca mediada por caspase, JNK e cAMP/AMPK em células de osteossarcoma canino. Além disso, tem o potencial de melhorar o prognóstico tumoral dessa doença por inativação da p53 mutada. No entanto, ela não interfere na expressão de AKT/mTOR, relacionado a proliferação e sobrevivência tumoral. Tais resultados servirão de base para estudos futuros que analisem o efeito da curcumina in vivo nessa doença.
O desenvolvimento de novos quimioterápicos é premissa da comunidade científica nos últimos anos, principalmente por meio do uso de plantas e outras fontes naturais. Nesse contexto, análises de citotoxicidade e efeito residual têm se tornado essenciais para o estabelecimento de novas terapias. Portanto, este estudo tem como objetivo avaliar os efeitos citotóxicos e residuais do extrato etanólico da casca do pequi (EEPP) sobre as células do osteossarcoma canino. Para isso, foram utilizadas análises de viabilidade celular, citotoxicidade, IC50 e sobrevida celular após terapia nos tratamentos de 24, 48 e 72 horas. Utilizou-se o delineamento de blocos ao acaso, utilizando-se a Análise de Variância com teste posthoc de Tukey para comparação dos tratamentos, considerando significância de 5%. O extrato não mostrou nenhum efeito citotóxico em 24 horas de tratamento, mas um efeito protetor, ou possivelmente um efeito indutor de mitose nas células de osteossarcoma. Porém, em 48 e 72 horas, encontramos citotoxicidade gradativa favorecendo o controle efetivo das células neoplásicas, obtendo-se, em média, 55% de citotoxicidades nas concentrações de 10 a 100 µg / mL. Os valores de IC50 foram 4,05 µg / mL para o grupo de 48 horas e 4,53 µg / mL para o grupo de 72 horas. O efeito residual do tratamento reduziu o crescimento celular após a exposição ao extrato. O fator de sobrevivência foi menor (18,81%) no grupo de 72 horas, com concentração de 10 µg / mL e maior (81,33%) no grupo de 48 horas, com concentração de 100 µg / mL. Esses resultados sugerem que o extrato etanólico da casca do pequi promove aumento da viabilidade celular em 24 horas, ação citotóxica em exposições acima de 48 horas e efeito residual dependente do tempo nas células do osteossarcoma canino.
