Bras. 32(8) The study aimed to determine the antimicrobial resistance patterns and to identify molecular resistance markers in Staphylococcus spp. (n=210) isolated from small ruminant mastitis in Brazil. The antimicrobial resistance patterns were evaluated by the disk diffusion test and by detection of the presence of mecA, blaZ, ermA, ermB, ermC and msrA genes by PCR. The efϐlux pump test was performed using ethidium bromide and bioϐilm production was determined by Congo red agar test along with PCR for detection of the icaD gene. The isolates were most resistant to amoxicillin (50.0%), streptomycin (42.8%), tetracycline (40.4%), lincomycin (39.0%) and erythromycin (33.8%). Pan-susceptibility to all tested drugs was observed in 71 (33.8%) isolates and 41 Staphylococcus isolates were positive for the efϐlux pump. Although phenotypic resistance to oxacillin was observed in 12.8% of the isolates, none harbored the mecA gene. However, 45.7% of the isolates harbored blaZ indicating that beta-lactamase production was the main mechanism associated with staphylococci resistance to beta-lactams in the present study. The other determinants of resistance to antimicrobial agents ermA, ermB, ermC, and msrA were observed in 1.4%, 10.4%, 16.2%, and 0.9% of the isolates, respectively. In addition, the icaD gen was detected in 32.9% of the isolates. Seventy three isolates (54 from goats and 19 from sheep) were negative for all resistance genes tested and 69 isolates presented two or more resistance genes. Association among blaZ, ermA, ermB, ermC and efϐlux pump were observed in 17 isolates, 14 of which originated from goats and three from sheep. The data obtained in this study show the resistance of the isolates to beta-lactamics, which may be associated with the use of antimicrobial drugs without veterinary control.
Mastitis is an inflammation of mammary gland, that are important in milking breed as well in meat ones. It is associated with serious reduction in milk production and quality, lambs weight gain reduction and mortality The goal of this work was determine the major etiologic agents of goat and sheep mastitis, as well as antimicrobial drug-resistance patterns and the agreement between two different diagnostic tools. We visit 25 goat, sheep, and goat and sheep farms in Pernambuco and Bahia State, and a total of 439 goats and 76 sheep milk samples were collected. To diagnose of small ruminant mastitis were compared two tests: Milk culture and California Mastitis Test (CMT). The bacterial drug-resistance pattern was determined by Kirby Bauer test. Staphylococcus spp. was the most frequent bacteria isolated from goat and sheep mastitis cases. Streptococcus spp., Corynebacterium spp. and gram-negative bacilli were isolated. It was possible to observe the high sensitivity to antimicrobial drugs in all tested bacteria, being the lower sensitivity percentage determined to nalidixic acid. Considering caprine mastitis diagnostic the comparative analysis between microbiologic culture and shown a concordance degree of K=0,17, although to ovine species these value was K=0,22. The use of CMT to subclinical mastitis diagnostic in goat and ewes must be associated to milk bacterial culture.INDEX TERMS: Pathogens, antimicrobial sensitivity, diagnostic, mastitis.RESUMO.-A mastite é a inflamação da glândula mamária que acomete raças de aptidão leiteira como também aquelas voltadas para produção de carne. O presente estudo teve por objetivo conhecer os principais agentes causadores de mastite em ovinos e caprinos, bem como a sua susceptibilidade aos agentes antimicrobianos, além de avaliar o grau de concordância entre testes diagnósticos. To verify the occurrence of caseous lymphadenitis in sheep and goats on farms of Pernambuco, Brazil, and in animals slaughtered in two Brazilian cities (Petrolina/PE and Juazeiro/BA), and to characterize the susceptibility profile of Corynebacterium pseudotuberculosis to disinfectants and antimicrobials, and its relationship with biofilm production were the objectives of this study. 398 samples were tested for sensitivity to antimicrobial drugs, disinfectants, and biofilm production. Among the 108 samples collected on the properties, 75% were positive for C. pseudotuberculosis. Slaughterhouse samples indicated an occurrence of caseous lymphadenitis in 15.66% and 6.31% for animals slaughtered in Petrolina and Juazeiro respectively. With respect to antimicrobials, the sensitivity obtained was 100% for florfenicol and tetracycline; 99.25% for enrofloxacin, ciprofloxacin and lincomycin; 98.99% for cephalothin; 98.74% for norfloxacin and sulfazotrim; 97.74% for gentamicin; 94.22% for ampicillin; 91.71% for amoxicillin; 91.21% for penicillin G; 89.19% for neomycin and 0% for novobiocin. In analyzes with disinfectants, the efficiency for chlorhexidine was 100%, 97.20% for quaternary ammonium, 87.40% for chlorine and 84.40% for iodine. 75% of the isolates were weak or non-biofilm producers. For the consolidated biofilm, found that iodine decreased biofilm formation in 13 isolates and quaternary ammonia in 11 isolates. The reduction of the biofilm formation was observed for iodine and quaternary ammonium in consolidated biofilm formation in 33% and 28% of the isolates, respectively. The results of this study highlight the importance of establishing measures to prevent and control the disease.
Caseous lymphadenits (CL) is a chronic and subclinical disease that affects goats and sheep and, consequently, causes economic losses, especially to small producers. The purpose of this study, through use of Polymerase Chain Reaction (PCR), was to verify the presence of virulence genes of phospholipase D (PLD), integral membrane protein (FagA), iron enterobactin transporter (FagB), ATP binding cytoplasmic membrane protein (FagC) and iron siderophore binding protein (FagD) in 168 isolates of C. pseudotuberculosis obtained from cases of caseous lymphadenitis in goats and sheep. FagA, FagB and PLD genes were detected in all 145 strains isolated from abscesses in superficial lymph nodes and in 23 strains isolated from viscera. The FagC gene was positive in 167 (99.40%) isolates. The FagD gene was detected in 160 (95.23%) isolates. All virulence factors analyzed were found more frequently among isolates collected in the viscera of animals with CL, indicating a multifactorial nature, as well as variations, in the invasive potential of C. pseudotuberculosis strains.
Hybanthus calceolaria, also known as "papaconha" or "ipepacuanha," is a herbaceous plant found in northeastern Brazil, which is often implicated by farmers as the cause of neurological signs in livestock grazing. Several poisoning outbreaks associated with the ingestion of this plant were observed in cattle in the municipalities of Colônia de Gurguéia in the state of Piauí and Sirinhaém in the state of Pernambuco, Brazil. The main clinical signs were ataxia, recumbency, and myokymia. No significant lesions were observed during necropsy or on histological examination. The disease was experimentally reproduced by the administration of 2 daily doses of 40 g/kg/body weight of the fresh green plant containing fruits. The plants without fruits were nontoxic, which is in accordance with the farmers' information, as it was stated that the poisoning only occurs when the plant is fruiting.
The semiarid northeast of Brazil contains a unique biome known as caatinga, with a maximum temperature of 40 ºC and a relativity humidity of 56%. The caatinga is characterized by a variety of plants, including Cereus jamacaru Dc (mandacaru), Poincianella microphylla Mart. ex G. Don (catingueira), Pilosocereus gounellei FAC Weber (xique-xique) and Mimosa tenuiflora (Willd.) Poir (jurema preta). Sheep and goat industries are economically strong in that region, despite the fact that caseous lymphadenitis is highly prevalent. The aim of the present study was to assess the survival and biofilm production of Corynebacterium pseudotuberculosis isolates in the environment and under controlled temperatures (28°C, 37°C and 42°C) under different surfaces (plants, soil, wood, wire and thorns). In addition, we investigated the effects of applying the disinfectants chlorhexidine, hypochlorite and quaternary ammonia in soil, tiles, wood and vegetation cover. Four strains of C. pseudotuberculosis were selected (two from goats and two from sheep) for inoculation according to their in vitro biofilm production. Adherence to microplates was used to assess the biofilm-forming ability of the bacteria. Lower survival rates were observed when isolates of C. pseudotuberculosis were subjected to a temperature of 42°C. In terms of caatinga biome plants, contamination of jurema-preta plants resulted in the lowest survival rates. The disinfectant quaternary ammonia promoted a lower inoculum survival in all surfaces. The disinfectants and the higher temperature contributed to the reduction of biofilm production in isolates of C. pseudotuberculosis. knowledge of these patterns is important for the establishment of disease control measures, given the questionable efficacy of the treatment and the immuno-prophylaxis of caseous lymphadenitis.
