The objective of the present paper was to verify the presence of aerolysin, hidrolipase, elastase and lipase virulence genes through the polymerase chain reaction (PCR) in A. hydrophila isolates obtained from ϐish of the São Francisco River Valley, and to evaluate virulence according to the presence of these genes in Nile tilapia ϐingerlings. One hundred and fourteen isolates from the bacteria were used. DNA was heat extracted and PCR undertaken using speciϐic primers described in the literature. For in vivo tests Nile tilapia ϐingerlings were used. From the PCR tests, negative isolates for all genes tested were selected, positive isolates for two genes (aerolysin and elastase) and positive for the four genes tested. These were inoculated at a concentration of 10 8 UFC/ml into the tilapias, considered as treatments; another group of animals was used as control (with inoculation of saline solution). In all, 12 distinct standards regarding the presence of virulence factors in isolates from A. hydrophila, were observed. Of the 114 isolates analyzed, 100 (87.72%) presented at least one of the virulence factors under study. The virulence factors were widely distributed among the A. hydrophila isolates. Aerolysin was the most frequent virulence factor present in the isolates analyzed. A. hydrophila led to the mortality of the Nile tilapia ϐingerlings, regardless of the absence or quantity of virulence genes tested.INDEX TERMS: Aeromonas hydrophila, virulence factors, aerolysin, elastase, hidrolipase, lipase, Oreochromis niloticus.
Mastitis is the principal disease affecting dairy herds worldwide. The aim of the present study was to characterize phenotypic and genotypic features associated with resistance to antimicrobials in Staphylococcus spp. isolated from 2064 milk samples of 525 lactating cows in the Northeast of Brazil. Of the 218 isolates analyzed, 57.8% were characterized as Staphylococcus aureus, 28% as coagulase-positive staphylococci other than S. aureus (oCPS), and 14.2% as coagulase-negative staphylococci (CNS). The test for susceptibility to antimicrobials showed amoxicillin (32.6%) to be the less effective drug in vitro, and the multi-drug resistance (MDR) rate for beta-lactams varied from 0 to 0.75. The genotypic characterization showed that 93.1% of the samples were tested positive for the blaZ gene, while none amplified mecA. The antibiotic efflux mechanism was observed in 0.9% of isolates. The biofilm formation was found in 3.7 and 96.3% of samples, respectively, on Congo red agar and on the microplate adhesion test, while the icaD gene was present in 92.2% of Staphylococcus spp. The high frequency of blaZ gene observed in this study was associated with the resistance of most Staphylococcus spp. to one or more of the beta-lactams tested, which are routinely used in Brazilian herds for mastitis treatment. The biofilm formation was also detected in the isolates analyzed being an important characteristic for pathogenicity and antimicrobial resistance of bacteria.
Pythiosis is a devastating infectious disease caused by an aquatic oomycete, Pythium insidioum, and affects animals and humans that inhabit wetlands. The disease is characterized mainly by granulomatous lesions in the hosts. The purpose of this study was to report the occurrence of pythiosis in sheep in the states of Pernambuco (PE) and Bahia (BA), Northeastern Brazil, as well as to evaluate the efficacy of an immunotherapic against ovine pythiosis. Blood samples were collected from 53 sheep, 49 from flocks in counties located in PE and four from BA. Seven sheep showed clinical signs of ovine pythiosis; one of them was submitted to euthanasia and its head and submandibular lymph node was collected and sent for histopathologic and mycological analyses. Other six sheep were treated with an immunotherapic. During the treatment the animals were kept in the Sheep Industry Sector facilities at Univasf/Petrolina-PE. ELISA, fungal culture and polymerase chain reaction (PCR) methods were used to confirm the diagnosis of clinical ovine pythiosis in the sheep flock. At microscopic examination of the material collected from the nasal cavity of a sheep euthanized was observed a focally extensive area of necrosis with presence of diffuse infiltration of intact and degenerated neutrophils bordering the cartilage. Only one sheep showed clinical cure, indicating efficiency in the pythiosis treatment of 16.7% (1/6). Ovine pythiosis has been increasing in several municipalities of PE and BA. In this context, the immunotherapy may be an alternative to be searched. Therefore, further studies are needed to investigate the effect of immunotherapy on ovine pythiosis. mação de lesões com aspecto granulomatoso nos hospedeiros. The development of new strategies for production of low‐cost deoxyribonucleic acid (DNA) adsorbents based on electrospun fibers of block copolymers [Eudragit E‐100/poly(ethylene oxide) (PEO)] is a promising way for fast retrieval of free of degradation double‐stranded DNA chains from aqueous solution. The adsorption takes place through electrostatic interaction between DNA and oppositely charged electrospun fibers that provide high surface area for DNA adsorption. The adsorption capacity observed for electrospun fibers of Eudragit E100/PEO (considering as target molecules the salmon sperm DNA) was in order of 44.31 mg g−1, following the Langmuir model in a typical adsorption at solid/liquid interface (pseudo‐second‐order model), that characterizes the electrospun fibers as promising templates for removal of biologic components and further use in molecular biology techniques, as DNA amplification by polymerase chain reaction. To verify the occurrence of caseous lymphadenitis in sheep and goats on farms of Pernambuco, Brazil, and in animals slaughtered in two Brazilian cities (Petrolina/PE and Juazeiro/BA), and to characterize the susceptibility profile of Corynebacterium pseudotuberculosis to disinfectants and antimicrobials, and its relationship with biofilm production were the objectives of this study. 398 samples were tested for sensitivity to antimicrobial drugs, disinfectants, and biofilm production. Among the 108 samples collected on the properties, 75% were positive for C. pseudotuberculosis. Slaughterhouse samples indicated an occurrence of caseous lymphadenitis in 15.66% and 6.31% for animals slaughtered in Petrolina and Juazeiro respectively. With respect to antimicrobials, the sensitivity obtained was 100% for florfenicol and tetracycline; 99.25% for enrofloxacin, ciprofloxacin and lincomycin; 98.99% for cephalothin; 98.74% for norfloxacin and sulfazotrim; 97.74% for gentamicin; 94.22% for ampicillin; 91.71% for amoxicillin; 91.21% for penicillin G; 89.19% for neomycin and 0% for novobiocin. In analyzes with disinfectants, the efficiency for chlorhexidine was 100%, 97.20% for quaternary ammonium, 87.40% for chlorine and 84.40% for iodine. 75% of the isolates were weak or non-biofilm producers. For the consolidated biofilm, found that iodine decreased biofilm formation in 13 isolates and quaternary ammonia in 11 isolates. The reduction of the biofilm formation was observed for iodine and quaternary ammonium in consolidated biofilm formation in 33% and 28% of the isolates, respectively. The results of this study highlight the importance of establishing measures to prevent and control the disease.
Background: Milk quality is considered unsatisfactory in Brazil due to factors of a social, economic, cultural, and climatic nature. Mastitis is the main disease that affects dairy herds. Microorganisms of the genus Staphylococcus are the most frequently isolated pathogens in cases of mastitis in bovines. Staphylococcus aureus requires more attention because they have the ability to develop resistance to antimicrobials used in the treatment of mastitis. Thus, the aim of the present study was to evaluate virulence factors in isolates of S. aureus as well as analyze the hygienic-sanitary quality of raw milk produced on dairy farms in a semiarid region of northeastern Brazil.Material, Methods & Results: Samples were taken from milk containers at 44 properties. Fifty-eight isolates of Staphylococcus spp. were analyzed and genotypically identified as S. aureus. In the phenotypic characterization, 56.8% (33/58) of the isolates demonstrated moderate biofilm production. In the genotypic characterization (icaA, icaD and bap genes), icaA was the most representative among the isolates. No resistance to cephalothin or oxacillin was found, but 62% (36/58) of the isolates exhibited resistance to ampicillin, amoxicillin, and penicillin. In the genotypic evaluation in response to β-lactam antibiotics, 50% (29/58) of the isolates exhibited the blaZ gene. Total bacterial counts were determined based on the standard plate count and flow cytometry, for which a significant positive correlation was found (r = 0.61, P < 0.01). The somatic cell count was also performed using flow cytometry and demonstrated that 6.8% (3/44) of the properties had values above one million cells per mL. The analysis of Staphylococcus spp. revealed that 20% (9/44) of the properties had significant counts for the production of staphylococcal enterotoxins. Regarding total coliforms, 56.8% (25/44) of the milk analyzed had counts above 1.0 x 103 CFUs/mL. There were no cases of the isolation of Pseudomonas spp. Streptococcus spp. was isolated on 6.8% (3/44) of the properties. The results of the sequencing revealed one isolate to be Lactococcus lactis and two to be Enterococcus spp.Discussion: None of the 58 isolates of S. aureus was classified as negative in the phenotypic characterization of biofilmforming capacity, which suggests the presence of genes involved in this process. Studies have shown that the molecular techniques used to identify ica genes encoding biofilm synthesis are very important for the identification of virulent strains. The isolates demonstrated resistance to β-lactam antibiotics of the class of penicillins, which are the most widely employed antimicrobial agents for the treatment of diseases in dairy herds. The presence of the blaZ gene in 50% of the isolates is in agreement with data reported in other studies developed in northeastern Brazil. In 50% (29/58) of the isolates, the presence of the blaZ gene was not observed. Of these, (8/29) were resistant to ampicillin, amoxicillin and penicillin. This is due to the existence of other bacterial mechanisms. Research has shown that some isolates possessed three mechanisms (blaZ, mecA and msrA) of antimicrobial resistance simultaneously. The results of the quality analyses demonstrate that some properties produce milk of low quality, which underscores the need for management strategies directed at reducing the contamination of the product.
Canine Degenerative Myelopathy is a late onset recessive autosomal disease characterized by a progressive ascending degeneration of the spinal cord. Two causal mutations are associated with this disease: a transition (c.118G>A) in exon 2 of the SOD1 that was described in several breeds and a transversion (c.52A>T) in exon 1 of the same gene described in Bernese Mountain dogs. The aim of this study was to understand the impact of the SOD1:c.118G > A mutation by genotyping a population of German Shepherd dogs in Brazil. A PCR-RFLP approach was used to genotype 97 healthy individuals belonging from the Northeast (Bahia and Pernambuco states) and South (Santa Catarina state) regions of Brazil. A total of 95 individuals were successfully genotyped resulting in an observed genotype frequency (with 95% confidence interval) of: 0.758 (0.672–0.844), 0.242 (0.156–0.328) and 0.000 (0.000–0.000) for “GG”, “AG” and “AA” genotypes, respectively. To our knowledge, this is the first attempt to describe the presence of the “A” allele associated with CDM (SOD1:c.118G > A) in German Shepherd dogs in Brazil and, as such, these results contribute toward important epidemiological data in this country and to the knowledge of the distribution of the aforementioned mutation worldwide.
