© Сахарный диабет, 2015ольшая распространенность сахарного диа-бета (СД), а также ежегодный рост заболевае-мости во всем мире носит характер эпидемии, несмотря на значительные успехи фармакологии в об-ласти создания гипогликемизирующих средств. В РФ на январь 2013 г. зафиксировано 3,779 млн больных СД. Из них больше половины -это люди трудоспособного возраста. При этом реальное число больных СД в России может быть в 3-4 раза больше [1].Пропорционально росту заболеваемости диабетом со-ответственно растет число его осложнений, которые могут иметь тяжелые последствия. Почти пятая часть пациентов с СД (17,63%) имеет проблемы со зрением. Слепота на-ступает в 25 раз чаще у лиц с диабетом [2]. Диабетическая нефропатия является ведущей причиной терминальной почечной недостаточности и занимает 20-44% всех диа-лизных мест. Наличие СД 2 типа (СД2) повышает вероят-ность развития инсульта в 2-6 раз. Сосудистая патология головного мозга на фоне инсулинорезистентности и ги-пергликемии при СД2 являются одной из основных при-чин развития диабетической нейропатии [3,4].По приблизительным подсчетам, мировые затраты на борьбу с СД достигли 471 млрд долларов США [5].Имеющиеся гипогликемизирующие средства и фарма-котерапевтические стратегии не в полной мере решают проблемы лечения СД2 и его осложнений. Поэтому ак-туальность поиска новых мишеней и терапевтических стратегий лечения и профилактики СД не вызывает со-мнений. A new (aryloxyalkyl)adenine derivative Adeprophen (9-[2-(4-isopropylphenoxy)ethyl]adenine, VMA-99-82) has a strong antidepressant effect on the model of reserpine-induced depression in rats (single dose 4 mg/kg, intraperitoneally). This effect manifested in suppression of depression-like behavior in the Porsolt forced swimming test (shortening of immobility time and increase in immobility latency, number of jumping episodes, and time of active swimming) and sucrose consumption/preference test (increase in the consumption of 20% sucrose solution in g/100 g body weight and percentage of sucrose preference in relation to the total fluid preference). Adeprophen had a greater antidepressant effect than sertraline and fluoxetine, but was less potent than amitriptyline, imipramine, venlafaxine, and to a lesser extent to paroxetine.
GABA analogs containing phenyl (phenibut) or para-chlorophenyl (baclofen) substituents demonstrated nootropic activity in a dose of 20 mg/kg: they improved passive avoidance conditioning, decelerated its natural extinction, and exerted antiamnestic effect on the models of amnesia provoked by scopolamine or electroshock. Tolyl-containing GABA analog (tolibut, 20 mg/kg) exhibited antiamnestic activity only on the model of electroshock-induced amnesia. Baclofen and, to a lesser extent, tolibut alleviated seizures provoked by electroshock, i.e. both agents exerted anticonvulsant effect. All examined GABA aryl derivatives demonstrated neuroprotective properties on the maximum electroshock model: they shortened the duration of coma and shortened the period of spontaneous motor activity recovery. In addition, these agents decreased the severity of passive avoidance amnesia and behavioral deficit in the open field test in rats exposed to electroshock. The greatest neuroprotective properties were exhibited by phenyl-containing GABA analog phenibut.
Substitution of drinking water with 1.8 % NaCl solution in pregnant female rats from day 1 of gestation until parturitions was followed by the development of experimental gestosis. Gestosis manifested in an increase in BP by 18.2 %, protein concentration in the urine by 6.2 times, and edema severity in muscles, brain, and omentum in comparison with the initial level. The concentration of homocysteine in blood plasma of rats with complicated pregnancy 4.4-fold surpassed that in pregnant rats without gestosis, which can probably in a cause for gestosis development. GABA derivatives citrocard (50 mg/kg) and salifen (15 mg/kg), and the reference substance sulodexide (30 U/kg) reduced the severity of gestosis manifestations, which was seen from the absence of BP rise, decrease in urinary protein concentration by 1.9, 2.0, and 1.3 times and blood level of homocysteine by 1.7, 1.5, and 2.6 times, respectively, and a decrease in edema degree in comparison with female rats with experimental gestosis receiving physiological saline.
