Introducción: En condiciones naturales, las raíces del arbusto, Phyllanthus acuminatus, producen bajas concentraciones de metabolitos secundarios de interés medicinal. Esto abre una oportunidad para el cultivo in vitro, para aumentar la concentración de metabolitos. Objetivo: Determinar las condiciones óptimas de cultivo líquido para raíces pilosas de P. acuminatus. Métodos: Se utilizó la evaluación del crecimiento de la biomasa según porcentaje de inóculo inicial (0.50 y 0.10 %), porcentaje de nutrientes de los medios (100, 50 y 25 %) y tasa de agitación (90, 100 y 110 min-1) (N= 15 repeticiones). Resultados: Las mejores condiciones de cultivo líquido fueron: 0.10 % de inóculo inicial, nutrientes al 25 % y 90 min-1 para la tasa de agitación. Hay diferencias entre las raíces pilosas y las raíces no transformadas. Conclusiones: es factible producir raíces pilosas de P. acuminatus a gran escala, aplicando e implementando las condiciones evaluadas de porcentaje de inóculo, nutrientes en el medio y tasas de agitación utilizadas en este estudio.
<p class="p1"><em>Stevia rebaudiana </em>es una planta herbácea perenne que pertenece a la familia Asteraceae. Es originaria del norte de Paraguay y crece en climas tropicales y subtropicales. Debido a la dulzura natural de sus hojas, se le conoce como edulcorante natural, sin calorías y con muchas aplicaciones en la industria alimentaria. Por lo tanto, resulta de gran importancia comercial contar con técnicas que permitan determinar la calidad y pureza de la materia prima procesada en polvo para la producción de edulcorantes. Se ha recurrido a la implementación de técnicas moleculares mediante marcadores ISSR, que permiten amplificar las secuencias que se encuentran entre los microsatélites, con lo que se obtiene un patrón de bandas característico de cada organismo. </p><p class="p1">En este estudio se analizó la amplificación de los ISSR S3, S4, S5, S10, S11, S12 y S15 a partir de muestras de ADN de <em>S. rebaudiana</em>, provenientes de plantas cultivadas en invernadero. Las muestras de ADN se obtuvieron mediante tres diferentes protocolos de extracción a partir de material seco y fresco de la planta. De acuerdo con los resultados obtenidos, los ISSR S3, S4, S5 y S11 representan una buena opción para la detección de polimorfismos en esta especie, y por tanto para la identificación molecular de la especie. Se debe comprobar si los ISSR S10, S12 y S15 son capaces de amplificar segmentos de ADN de <em>Stevia</em>, ya que bajo las condiciones de trabajo de esta investigación no se logró la amplificación de ADN con estos imprimadores. Se considera que los ISSR son marcadores universales y se podría identificar individualmente cada cultivar de <em>Stevia </em>que vaya a destinarse a la producción comercial de esteviósidos, pero es necesario repetir los protocolos descritos con diferentes variedades e incluso se puede trabajar con diferentes especies del género <em>Stevia</em>, para obtener una caracterización general. </p>
<p>Los beneficios para la salud registrados a partir del uso de metabolitos secundarios de la planta llamada uña de gato (<em>Uncaria tomentosa</em>) han generado una fuerte demanda comercial, así como la extracción intensiva de esta especie en los países en los cuales se distribuye, con el consecuente deterioro de este recurso genético en su hábitat natural.</p> <p>Es por eso que resulta necesario implementar protocolos de cultivo de células y tejidos de esta especie, con el fin de lograr la síntesis de los compuestos en forma controlada. La corteza de las raíces es uno de los tejidos en los que se concentra la producción de estos compuestos, razón por la cual la producción de raíces de cabellera (<em>hairy roots</em>) resulta ser una técnica alternativa para la producción a escala de los metabolitos de interés.</p> <p>En este proyecto se implementó un protocolo de agroinfección de microestacas de <em>U. tomentosa </em>utilizando cepas silvestres de <em>Agrobacterium rizhogenes </em>(AR1500 y A4RS), así como el mantenimiento en medio líquido de las raíces pilosas obtenidas. En colaboración con el Laboratorio de Biología Molecular del programa PIPRA (UC Davis), se determinó la eficacia del protocolo de agroinfección, así como el uso de otras herramientas moleculares para la detección de expresión génica, las cuales mostraron resultados satisfactorios en los ensayos de agroinfección, bajo las metodologías establecidas en el proyecto.</p>
La biotecnología ha sido utilizada desde hace miles de años para obtener beneficios para el ser humano. Desde mucho antes los seres humanos han utilizado técnicas como la hibridación, mutagénesis y poliploidía, generando cambios a nivel genético para obtener características deseables Actualmente es posible utilizar la ingeniería genética para modificar o transferir genes de un organismo a otro con el fin de obtener productos con ciertas ventajas respecto a su contraparte convencional. Las técnicas de ingeniería genética se iniciaron desde hace 30 años a nivel de investigación y hace ya 20 años se comercializan a gran escala productos mediante estas las mismas. A pesar que se han generado diversas observaciones en cuanto a su inocuidad y bioseguridad por ciertos sectores de la sociedad a la fecha múltiples estudios con gran respaldo científico en diversas partes del mundo concluyen de manera general, que a nivel ambiental y de salud humana, no existen diferencias entre el mejoramiento convencional y el mejoramiento por ingeniería genética. El desarrollo de alimentos mejorados mediante biotecnología está acompañado por normas y marcos regulatorios de bioseguridad, los cuales involucran análisis de riesgo con pruebas de larga duración para asegurar la inocuidad del alimento.
