Salt stress is one of several major abiotic stresses that affect plant growth and development, and there are many evidences that silicon can ameliorate the injuries caused by high salinity. This study presents the results of an assay concerning: (1) the effect of in vitro NaCl-induced salt stress in cape gooseberry plants and (2) the possible mitigating effect of silicon in saline conditions. For that, nodal segments were inoculated in Murashige and Skoog (MS) medium under salinity (0.5 and 1.0% NaCl) with different silicic acid concentrations (0, 0.5 and 1.0g L-1). Phytotechnical characteristics, photosynthetic pigments content, and leaf anatomy were evaluated after 30 days. Shoot length, root length, number of leaves and buds, fresh and dry weight, pigment content, stomatal density and leaf blade thickness were drastically reduced by increased salt level. The supply of silicon (1.0g L-1) has successfully mitigated the effect of salinity at 0.5% NaCl for chlorophyll, carotenoids, stomatal density and leaf blade thickness. When salt stress was about 1.0%, Si was not effective anymore. In conclusion, we affirmed that, in in vitro conditions, salt stress is harmful for cape gooseberry plants and the addition of silicon showed effective in mitigating the saline effects of some features.
Foi multiplicado in vitro, o abacaxi ornamental Ananas comosus var. erectifolius, induzindo o estiolamento de segmentos nodais com posterior regeneração de brotações. Os eixos caulinares foram inoculados nos meios de cultura: MS sem regulador de crescimento; MS + 10 µM de ácido indolbutírico (AIB); MS + 10 µM de ácido naftalenoacético (ANA); MS + 10 µM de ácido indolacético (AIA), e mantidos no escuro, a 25±2ºC. Aos 60 dias após a inoculação, avaliaram-se número de brotos/explante, número de nós/broto, comprimento de brotos, distância entre os nós e número total de nós/explante. O meio MS + 10 µM de ANA promoveu os maiores valores para número de brotos e número total de nós/explante. Para a regeneração de brotos, foram utilizados segmentos nodais, oriundos de eixo caulinar (explante) estiolados in vitro, contendo dois nós, nos meios: MS sem regulador de crescimento; MS + 4,44 µM de 6-benzilaminopurina (BAP); MS + 8,88 µM de BAP e MS + 13,32 µM de BAP. As culturas foram incubadas sob fotoperíodo de 16 horas, a 25±2ºC. Aos 30 dias, não houve diferença no número de brotos regenerados por nó entre os meios testados, enquanto aos 45 e 60 dias de cultivo, a adição de BAP teve efeito positivo sobre o número de brotos regenerados por nó, quando comparado com o meio sem a adição desta citocinina.
ABSTRACT. The objective of this study was to determine the silicon concentration that would provide good growth in passion fruit plants. Passion fruit seeds were sown in polystyrene. After 60 days, when they were approximately 15 cm tall, the plants were transplanted into polyethylene pots containing 1.1 kg Tropstrato® substrate. Treatments consisted of four concentrations (0, 0.28, 0.55, and 0.83 g pot -1 ) of silicon applied as a silicic acid solution 1%. This solution was applied around the stem of the plants (drenched), with the first application being administered 15 days after transplanting. In total, three applications were made at intervals of 15 days. After the last application, the plants were subjected to chemical analysis to determine the silicon concentration and to X-ray microanalysis and flow cytometry. Phytotechnical analyses were performed during the applications. The use of silicon in concentrations of 0.28 and 0.55 g pot -1 provides better growth of the passion fruit, and the absorption and deposition of the silicon in the passion fruit leaves are proportional to the availability of this element in the plant. The roots of the passion fruit plant are silicon accumulators, and the DNA stability and amount are preserved in the silicon-treated passion fruit plants. ) de silício, na forma de solução de ácido silícico a 1%. RESUMOAs espécies tropicais apresentam grande importância no mercado de flores, mas informações sobre colheita e procedimentos pós-colheita ainda são limitados. Para a maioria das espécies tropicais, existem poucas informações sobre soluções a serem utilizadas para condicionamento ou manutenção após a colheita. Assim, objetivou-se avaliar a durabilidade e qualidade pós-colheita de hastes florais de bastão-do-imperador em concentrações de sacarose na solução de condicionamento e diferentes estádios de abertura floral. Hastes florais foram colhidas no início da manhã e tratadas com solução de condicionamento com sacarose nas concentrações 0 (controle), 10, 20, 30% por 24h, nos três estádios de abertura floral: inflorescências com as brácteas 1/3, 2/3 ou totalmente abertas. O delineamento experimental foi inteiramente casualizado, em esquema fatorial 4x3x6, com 3 repetições em parcela subdividida no tempo. As avaliações foram realizadas em intervalos de 3 dias, determinando-se a massa fresca das hastes florais e avaliação de qualidade das inflorescências por meio de notas, em escala de 0 a 4. Observou-se que a durabilidade comercial das hastes florais é maior (10 dias) quando as inflorescências são colhidas com 1/3 das brácteas abertas. Recomenda-se a realização de condicionamento em hastes florais de bastão-do-imperador com concentração de 20% de sacarose por 24h para inflorescências colhidas com 1/3 e 2/3 das brácteas abertas. ABSTRACT Developmental stage and pulsing in inflorescences of torch gingerTropical species are very important in the flower market, but information in harvest and postharvest procedures are not enough. There is not enough information on pulsing or holding solutions for most tropical species. The objective was to evaluate the durability and quality of postharvest torch ginger flower stems (Etlingera elatior) at sucrose concentrations in pulsing solution and different flower opening stages. Flower stems were harvested early morning at three flower opening stages: inflorescences with 1/3, 2/3 and completely open bracts, withal the stalks were pulsed with sucrose solution at concentrations of 0 (control), 10, 20, and 30% for 24h. Experimental design was completely randomized in a 4x3x6 factorial with 3 replications in a split plot in time. Evaluations were performed every 3 days, determining the fresh weight of flowers and assessing inflorescence quality through scores on a 0-4 point scale. Commercial vase life of stalks is longer (10 days) when inflorescences are harvested with 1/3 bracts opened. It is recommended to carry out pulsing in torch ginger flower stems with concentration of 20% sucrose for 24h for inflorescences harvested with 1/3 or 2/3 of the bracts opened.
<p>Na micropropagação comercial de antúrio as mudas são produzidas via organogênese indireta, podendo gerar variantes somaclonais. Visando reduzir este problema, o objetivo do trabalho foi obter um protocolo de multiplicação <em>in vitro</em>, por meio da indução ao estiolamento e posteriormente regeneração de plantas, em <em>A. andraeanum</em> var. IAC Eidibel. No estiolamento, segmentos caulinares desfolhados, com 3 a 5 gemas, foram inoculados em tubos de ensaio contendo os meios MS e Pierik, sem auxinas ou acrescido de AIA, AIB ou ANA na concentração de 10 µM. As culturas foram mantidas em câmara de crescimento, a 25 ± 1ºC, no escuro, por 60 dias. Na regeneração de plantas, brotos estiolados seccionados em segmentos contendo dois nós, foram inoculados horizontalmente em tubos de ensaio, contendo os mesmos meios, com diferentes concentrações de BAP (0; 2,22; 4,44; 6,66 µM). As culturas foram mantidas em câmara de crescimento com intensidade luminosa de 30 ?mol.m-2s-1, sob fotoperíodo de 16 horas, temperatura a 25 ± 1ºC, por 60 dias. Os dados foram submetidos à análise de variância, e as médias comparadas pelo teste de Tukey, a 5% de probabilidade. Para o estiolamento, o meio Pierik foi superior para as características: comprimento do broto, número de nós/broto, número total de nós/explante e distância entre os nós. Os meios sem a adição de auxina diferiram daqueles contendo 10 µM de ANA, para todas as variáveis. Na regeneração das mudas, as menores taxas de multiplicação foram registradas nos meios sem BAP. Recomenda-se meio Pierik sem auxina para o estiolamento; e, para regeneração das mudas, o mesmo meio acrescido de 2,22 µM de BAP. </p>

This study aims to induce callus and embryogenic potential and determine the genetic stability in pitaya. The study was divided into four steps: 1ª) Callus induction using doses of 2,4-D (2.0 and 4.0 mg L -1 ) and with doses of glutamine (100 and 200 mg L -1 ); 2ª) callus growth curve for various time periods (0, 7, 14, 21 , 28, 35, 42 , 49, 56, 63 , 70, 77, 84 , 91, 98, 105 and 112 days); 3ª) Cytochemical analysis to assess the potential of embryogenic callus (samples consisting of 100 mg of callus) and 4ª) Analysis of flow cytometry to assess the genetic stability of the callus (compared with a seedling in vitro -control). The conclusion is that the 4.0 mg L -1 2,4-D is the best concentration for callus induction in explants shoots of pitaya; the glutamine does not influence the callus formation. The callus has higher embryogenic potential at 49 days, therefore must be spiked in this period. By flow cytometric was detected endoreduplication in the material analyzed.
