Background: Leptospirosis is considered the most widespread zoonosis worldwide, occurring more frequently in tropical and developing regions. The aim of the present study was to detect the presence of Leptospira spp. in different primate tissues, using immunohistochemical (IHC) assays, taking advantage of the considerable number of necropsies compatible with a diagnosis of leptospirosis in neotropical primates at the Animal Pathology Laboratory (APL) of the University of Passo Fundo (UPF) in the northern region of Rio Grande do Sul. Materials, Methods & Results: Paraffin-embedded primate tissue samples were selected from necropsy examinations and subjected to IHC. The streptavidin-biotin-peroxidase method was used with diaminobenzidine chromogen (DAB) to verify immunostaining. Of the101 primates tested for Leptospira spp., 51.48% were positive; taining was distributed between lung (76.92%), liver (44.23%), and kidney (32.69%) tissue. Analysis of the combined anatomopathological verification data of the studied organs revealed a high frequency of lesions commonly observed in the tissues of animals exposed to the pathogen. For complementary diagnosis, an anti-Leptospira spp. antibody test was performed in primates at the UPF-Zoo, from which a population of the necropsied animals originated. The microscopic agglutination test (MAT) was utilized, which demonstrated 90.47% positivity in 21 individuals; sejroe and panama were the most frequent serovars. Discussion: Different intensities of tissue immunostaining were observed. Areas of fragmented or diffuse staining were considered to indicate equal positivity to that indicated by areas of staining with preserved morphology. Of 52 Leptospirapositive primates, most presented some degree of staining in lung samples, which shows a high level of involvement for this organ in primate leptospirosis. Conventional pathological diagnostic methods do not allow fort issue antigen recognition, thus making the IHC technique important to facilitate conclusive antigen sample verification. In the liver, leptospires were detected mostly between the sinusoids, hepatocytes, and Kupffer cells. In kidney tissues, staining indicated small agglomerates in the tubular lumen, interstitium, and glomeruli. All these forms of presentation have been previously reported. Considering that we detected the highest number of positive samples in lung tissue, followed by those from liver and kidney tissue, we argue that the IHC technique, when applied to samples of these three tissues, decreases the chance of false negatives. Anatomopathological studies of primate leptospirosis are scarce. In dogs, renal lesions are characterized by the necrosis and degeneration of tubular epithelium, cellular debris, and hyaline cylinders. RESUMO O objetivo do presente trabalho foi determinar a presença de anticorpos para Leishmania infantum, Neospora caninum e Toxoplasma gondii, por meio da reação de imunofluorescência indireta (RIFI), em cães (n=78) provenientes da região central do Rio Grande do Sul, necropsiados no Hospital Veterinário da Universidade Federal de Santa Maria (UFSM), bem como avaliar os dados epidemiológicos, sazonais e anátomo-histopatológicos. Do total de animais avaliados, 67,9% (53/78) apresentaram soropositividade para ao menos um agente. A ocorrência de anticorpos para L. infantum, N. caninum e T. gondii foi de 33,3 (26/78), 37,1 (29/78) e 43,5% (34/78), respectivamente. Detectaram-se monoinfecções em 9,4% (5/53) para L. infantum, 18,8% (10/53) para N. caninum e 20,7% (11/53) para T. gondii. As coinfecções foram observadas em 27/53 (50,9%) dos animais. As infecções ocorreram independentemente de idade, sexo, procedência ou raça (P>0,05). Não se verificaram lesões anatomo-histopatológicas relacionadas aos agentes pesquisados, caracterizando-os como animais assintomáticos. Os resultados confirmaram a exposição de cães a esses protozoários na região central do RS e, em especial, demonstraram a circulação do agente causador da leishmaniose em uma área considerada indene para a enfermidade.
Background: Skin tumors are the most common neoplasia in veterinary medicine and squamous cell carcinoma (SCC) is the most frequent. In cattle, it is the main tumor besides linfosarcoma and normally affects taurine cattle, but is rare in Nelore cattle. SCC mainly affects mucocutaneous junctions such as the eyelids, sclera, vulva, and perineum, and has already been described in vagina, eye, and periocular tissues, in addition to the superior digestive tract. The purpose of this paper is to describe a vaginal squamous cell carcinoma in a Nelore cow. Case: A 10-year-old Nelore cow, used as an embryo donor, was submitted to a necropsy procedure. Clinically, the animal had prolonged recumbency, anorexia, weight loss, and vaginal discharge. There was a history of recurrent vaginal prolapses and an intraluminal vaginal mass that had grown over two years. At necropsy, there was diffuse fibrinous peritonitis with marked intestinal adhesions; the vagina was markedly expanded and measured 40 × 20 cm due to the presence of an intraluminal mass occupying the vestibulum and extending up to the cervix. In the vaginal lumen, there was large amount of fibrinonecrotizing malodorous material; the vaginal wall was thickened by an unencapsulated and poorly delimited, firm and white mass composed of sheets and nests of epithelial cells from the squamous layer surrounded by abundant fibrovascular stroma. The neoplastic cells were polygonal with marked pleomorphism, anisokaryosis, and anisocytosis; abundant eosinophilic and well-defined cytoplasmic borders. Sometimes it was possible to see desmosomes. The nuclei were rounded to oval with sparse chromatin and one or more nucleoli were seen. Mitotic figures were frequent (at least 3 per higher power field) and there were also a few multinucleated cells. Frequently, there was the formation of distinct keratin "pearls". Discussion: SCC frequently occurs in the skin, eyes, and vulva in cattle, since these are sites that lack pigment within the epidermis and have prolonged exposed to ultraviolet light. Vaginal SCC has already been described in one cow with recurrent vaginal prolapses that was submitted to follicular aspiration. In a survey of cattle tumors, most genital tract neoplasias affected the uterus, and vaginal tumors normally originated from connective tissue -fibromas, fibropapillomas, and fibrosarcoma -or muscular tissue -leiomyoma and leiomyosarcoma. Irreversible changes in cellular DNA can occur due to physical or chemical carcinogens, and also chronic inflammation. The cow in this paper was frequently submitted to follicular aspiration and this can cause chronic inflammation in the vaginal mucosa after the puncture and, when it's chronic, can represent one of the main initiating factors for tumor development. This cow's death was caused by diffuse and marked peritonitis. The same cause was previously described in cows submitted to follicular aspiration and, in one of these animals, there was a rectal perforation caused by mistakes during the procedure. RESUMO: Apesar da prevalência da peritonite infecciosa felina (PIF) ser alta em praticamente o mundo todo, estudos anatomopatológicos recentes acerca dessa doença são escassos. Não obstante, as características microscópicas da medula óssea de gatos com PIF estão ausentes da literatura consultada. O objetivo deste artigo é descrever alterações medulares ósseas vistas em casos espontâneos de PIF. As medulas ósseas colhidas sistematicamente da região diafisária dos fêmures de 16 gatos necropsiados no Laboratório de Patologia Veterinária (LPV) da Universidade Federal de Santa Maria (UFSM), Rio Grande do Sul, entre janeiro de 2000 e junho de 2017, e que tiveram diagnóstico definitivo de PIF, foram avaliadas fenotípica (histopatologia - hematoxilina e eosina e histoquímica - reação de Perls) e imunofenotipicamente (utilizando marcadores mieloides (anti-MAC387) e de linfócitos (anti-CD79αcy e anti-CD3). Os resultados permitem afirmar que, independentemente da apresentação clinicopatológica da doença ocorrem as seguintes alterações: 1) hiperplasia mieloide; 2) hipoplasia eritroide, 3) displasia megacariocítica (dismegacariocitopoiese) e 4) plasmocitose medular. Exclusivamente nos casos de PIF seca há hemossiderose medular óssea e hepática. Essas alterações permitem estabelecer que gatos com PIF desenvolvem mielodisplasia, uma lesão mieloproliferativa muito semelhante àquela relatada em humanos infectados pelo HIV. Sugere-se que a partir dos achados aqui descritos, a mielodisplasia seja considerada a principal responsável pelas alterações hematológicas observadas na PIF, especialmente pela anemia e trombocitopenia arregenerativas frequentemente desenvolvidas pelos pacientes com essa doença.
RESUMO: Linfomas foliculares são uma rara forma de distúrbio linfoproliferativo descrita em medicina veterinária. Juntamente com a não reconhecida ocorrência dos linfomas de Hodgkin em cães, essa é a maior diferença acerca de linfoma entre humanos e cães. O objetivo deste artigo é descrever os achados epidemiológicos, clínicos e anatomopatológicos vistos em cinco cães com linfoma folicular. Destes, dois eram machos (40%) e três eram fêmeas (60%). A idade dos cães afetados variou de 11 a 13 anos. Quatro dos cinco (80%) cães eram de raça pura e um (20%) não tinha raça definida. Todos os cães apresentaram linfadenomegalia generalizada e esplenomegalia, o que incluiu os casos como linfoma multicêntrico. Na necropsia, os linfonodos e o baço demonstraram um padrão nodular à superfície de corte, caracterizado por dezenas a centenas de nódulos brancos, multifocais ou coalescentes e de tamanhos variáveis. Na superfície natural do baço, frequentemente (4/5, 80%), havia miríades de pontos brancos, multifocais ou coalescentes, de tamanhos variáveis. Na histopatologia, os tumores foram confirmados como linfomas foliculares. Todos os casos eram Grau III, sendo dois (40%) incluídos como IIIa e outros três (60%) como IIIb. Em um caso (1/5, 20%), o linfoma folicular foi considerado como IIIb variante de pequenos centroblastos semelhantes aos linfócitos neoplásicos vistos no linfoma de Burkitt. Os linfomas foram validados como tendo origem em células B através da imuno-histoquímica, utilizando anticorpos anti-CD20. Os casos de linfomas foliculares descritos comportaram-se de forma agressiva e levaram os pacientes à morte.
A 10-year-old male large mixed breed dog was presented with skin ulcers and fracture on the right hind limb caused by vehicle collision. Given required limb amputation, and as being a shelter senior dog, euthanasia was requested by the owner and a complete post-mortem examination was conducted immediately after death. Gross changes were consistent with marked bilateral nephromegaly. Histopathological examination of the kidneys revealed round cells filling blood vessels. Immunohistochemically, the round cells were positive for CD3 antibody. Based on these findings, in absence of involvement of the bone marrow and peripheral blood, and inexistence of primary extravascular masses, the tumor was classified as T-cell intravascular lymphoma. To the author’s knowledge, this is the first report describing intravascular lymphoma involving the kidneys alone in a dog.
RESUMO.-O objetivo deste estudo foi apresentar dados epidemiológicos, clínicos e anatomopatológicos de cinco cães com neuromiopatia isquêmica secundária a trombose ou tromboembolismo aórtico. Os cães eram fêmeas, com idade ≥11 anos, que apresentavam paraplegia (80%) ou monoplegia (20%) de membro pélvico. Em 40% dos casos (n=2) foi relatada claudicação previamente à disfunção motora. Os principais achados clínicos foram ausência de pulso femoral uni ou bilateral, extremidade de membros pélvicos frias, dor muscular acima da articulação do joelho e ausência de nocicepção (dor profunda) nos dígitos dos membros pélvicos.Quatro cães foram submetidos à eutanásia devido ao prognóstico desfavorável e um veio a óbito em menos de 24 horas de internação. Na necropsia foi encontrada alteração cardía-ca em três cães, neoplásica em um e, sem causa determinante em outro caso. Os trombos estavam localizados na aorta abdominal e se estendiam até as artérias ilíacas. Mesmo sendo baixa a incidência, a neuromiopatia isquêmica deve ser incluída no diagnóstico diferencial de cães com claudicação, paraparesia ou paraplegia uni ou bilateral, quando os sinais neurológicos são compatíveis com lesão de neurônio motor inferior (L4-S3) e de evolução aguda ou crônica. A avaliação do pulso femoral digital e/ou por doppler vascular deve ser empregada e pode auxiliar no diagnóstico da doença evitando que os animais iniciem tratamento para outras afecções. This study aimed to present epidemiological, clinical and anatomopathological data of five dogs with ischemic neuromyopathy secondary to aortic thrombosis or thromboembolism. The dogs were females, with eleven years of age or more, that had paraplegia (80%) or hind limb monoplegia (20%). Forty percent of the cases (n=2) had lameness previously the motor disfunction. The main clinical findings include absence of unilateral or bilateral femoral pulse, cool hind limbs extremities, muscle pain above the knee joint and absence of nociception (deep pain) on the hind limbs digits. Four dogs were euthanized due to the poor prognosis, and the last one died within 24 hours of hospital admission. The necropsy findings were cardiac lesions in three dogs, neoplastic disease in one, and unknown cause on the other. The thrombi were located on the abdominal aorta, extending to the iliac arteries. Even with low incidence, the ischemic neuromyopathy should be included in the differential diagnosis of dogs with lameness, paraparesis, or unilateral or bilateral paraplegia, when neurological signs are compatible with lower motor neuron lesion (L4-S3) and acute or chronic progression. The digital and/or vascular doppler evaluation of the femoral pulse must be used and can assist on the disease diagnosis, avoiding treatments for other illnesses.
RESUMO Reticulose pagetoide é a expressão utilizada para uma das mais raras formas de linfoma descritas em cães. Trata-se de um distúrbio linfoproliferativo cutâneo que emerge de linfócitos T intraepidérmicos. Descreve-se aqui um caso de reticulose pagetoide com apresentação localizada, muito semelhante à doença de Woringer-Kolopp dos humanos, que acometeu um cão, Boxer, fêmea de 10 anos de idade. O paciente foi atendido devido ao aparecimento de um nódulo na região nasolabial, com evolução clínica de cerca de 30 dias. Histologicamente o nódulo era composto por uma proliferação linfoide intraepidérmica e anexal. Os linfócitos neoplásicos, células T confirmadas por meio da imunomarcação positiva para CD3 e negativa para CD79, eram grandes células redondas e monomórficas. Apesar de rara, a reticulose pagetoide é um distúrbio linfoproliferativo de fácil suspeita com base apenas na morfologia celular. Esse neoplasma deverá ser lembrado sempre que um padrão linfoide intraepidérmico estiver presente.
