Resumo -O objetivo deste trabalho foi avaliar a influência da polinização artificial na produtividade e qualidade de frutos de cultivares de maracujazeiro-amarelo. Foram utilizadas sete cultivares comerciais de maracujazeiro-amarelo, em dois experimentos realizados na região de Tangará da Serra, MT, em delineamento de blocos ao acaso, com cinco repetições e parcelas constituídas por duas linhas de quatro plantas. No primeiro experimento, a polinização foi natural; no segundo, foi realizada a polinização artificial, duas vezes por semana. O plantio foi feito no dia 11 de janeiro de 2010 e as colheitas no período de julho de 2010 à março de 2011. A polinização artificial aumentou a produtividade de todas as cultivares, mas estas apresentaram diferentes sensibilidades à técnica. Quando a polinização artificial foi utilizada, as cultivares FB 100, FB 200 e BRS Ouro Vermelho tiveram maior rendimento do que IAC 275, IAC 277, BRS Sol do Cerrado e BRS Gigante Amarelo. A polinização artificial aumenta a produtividade, a massa de fruto, o diâmetro e comprimento de fruto e a percentagem de polpa, e reduz a espessura de casca. Há indicação de forte interação genótipo por ambiente, quanto à produtividade das cultivares avaliadas.Termos para indexação: Passiflora edulis, Xylocopa, florescimento, produção, variedades. Productivity and fruit quality of yellow passion fruit cultivars with or without artificial pollinationAbstract -The objective of this work was to evaluate the influence of artificial pollination on productivity and fruit quality of yellow passion fruit cultivars. Seven commercial cultivars of yellow passion fruit were evaluated in two experiments carried out in Tangará da Serra, MT, Brazil, in a randomized block design with five replicates and plots consisting of two rows of four plants. In the first experiment, pollination was natural, whereas in the second it was artificial, performed twice a week. Planting was done on January 11, 2010, and harvests occured from July 2010 to March 2011. Artificial pollination increase the productivity of all cultivars, but their response to the technique varied. When artificial pollination was used, the cultivars FB 100, FB 200 and BRS Ouro Vermelho had a higher productivity than IAC 275, IAC 277, BRS Sol do Cerrado e BRS Gigante Amarelo. Artificial pollination increases productivity, fruit mass, diameter and length of fruit, and pulp percentage, and it reduces peel thickness. Strong genotype by environment interaction may be occurring, as for the productivity of the evaluated cultivars.
The propagation of many passion fruit species (Passiflora spp.) is done by seeds or sexual method, although asexual propagation, such stem cuttings, grafting or tissue culture is also possible and very useful in many cases. In this work, it was presented information on scientific and technological advances in sexual and asexual propagation of passion fruit, considering the different propagation methods and their applications. New demands for research and development activities are also related, given the wide range of passion fruit species and its potential for food, ornamental and medicinal use.
Resumo -O objetivo deste trabalho foi estimar a divergência genética entre acessos de mandioca açucarados e não açucarados, por meio de marcadores moleculares, caracteres quantitativos e qualitativos, bem como determinar a correlação entre essas estimativas. Foram utilizados quatro acessos de mandioca açucarados e quatro não açucarados, com duas variedades locais e duas comerciais. Os acessos foram avaliados em campo, em laboratório, com marcadores RAPD, quanto a 12 caracteres quantitativos e 33 morfológicos. Foram estimadas as matrizes de dissimilaridade/distância genética entre os acessos, por meio dos caracteres qualitativos e quantitativos e da significância da correlação entre as matrizes. A divergência genética entre os acessos foi elevada e os acessos açucarados foram diferenciados das variedades não açucaradas locais e comerciais. As distâncias estimadas, por meio de marcadores moleculares e caracteres qualitativos, evidenciaram elevada associação entre si e associação moderada com a estimada por meio de caracteres quantitativos.Termos para indexação: Manihot esculenta, melhoramento genético, recursos genéticos, variabilidade genética. Genetic divergence among sugary and nonsugary cassava accessionsAbstract -The objective of this work was to estimate the genetic divergence among sugary and nonsugary cassava accessions, through molecular markers, and quantitative and qualitative characters, as well as to determine the correlation among these estimates. Four sugary cassava accessions and four nonsugary ones were used, including two landraces and two nonsugary improved varieties. The accessions were evaluated in field conditions and in laboratories, with RAPD markers, in 12 quantitative and 33 qualitative characters. Matrixes of genetic dissimilarity/distance among accessions were estimated, based on qualitative and quantitative characters, molecular markers, and on the correlation significance among matrixes. High genetic divergence among the evaluated accessions was observed, the sugary accessions were differentiated from the landraces and improved nonsugary varieties. The distances estimated through molecular markers and qualitative characters showed the high association among each other and moderate association with the distance estimated through quantitative characters.
Molecular markers (RAPD, AFLP and microsatellites) were used to generate a linkage map and to identify QTLs associated to witches' broom (Crinipellis perniciosa) resistance in cacao (Theobroma cacao), using 82 individuals of an F 2 population derived from the clones ICS-1 (susceptible) and Scavina-6 (resistant). Fifteen evaluations of the number of brooms have been carried out in six years (1997)(1998)(1999)(2000)(2001)(2002). In order to increase the precision and accuracy in the measures of resistance, each F 2 plant was cloned in three replications in a randomized block design with singletree plots and evaluated over 2 years. Three hundred and forty-two markers were obtained, being 33 microsatellites, 77 AFLPs and 232 RAPDs. The distribution of the number of brooms in the F 2 population was skewed to resistance, suggesting the involvement of major genes controlling resistance and the repeatability estimated for resistance was 44%. A strong putative QTL was detected as being related to witches' broom resistance. Associated to this QTL, the microsatellite mTcCIR35 explained 35.5% of the phenotypic variation in resistance. This marker is being used for marker-assisted selection in Scavina-6 progenies, including those selected in private plantations, as an auxiliary tool to the phenotypic selection.
3RESUMO -Apesar do grande potencial comercial da pitaia, ainda são escassos os estudos de caracterização físico-química de frutos da pitaia, principalmente considerando espécies nativas do Cerrado. Neste trabalho, objetivou-se analisar a caracterização físico-química, polifenóis e flavonoides amarelos totais de frutos de espécies de pitaia Hylocereus costaricensis, Hylocereus undatus, Selenicereus setaceus e Selenicereus megalanthus. Para as avaliações físico-químicas, foram realizadas as análises de sólidos solúveis, pH e acidez total titulável. Para a determinação dos compostos fenólicos, realizaram-se as análises de polifenóis extraíveis totais e flavonoides amarelos. Foram observadas diferenças significativas entre as espécies de pitaia e entre as partes basal, mediana e apical dos frutos, quanto às características físico-químicas e a concentração de compostos fenólicos. A espécie S. megalanthus apresentou maior quantidade de sólidos solúveis, apresentando, assim, a polpa mais doce. Tal característica foi mais pronunciada na parte mediana do fruto de todas as espécies. Houve diferença significativa entre o pH, com valores variando de 4,84 a 5,67, classificando-se como alimentos pouco ácidos. A acidez variou de 0,10 % a 0,15 % de ácido cítrico. H. costaricensis merece destaque pela presença de maior quantidade de polifenóis totais e de flavonoides amarelos, diferenciando-se significativamente das demais espécies. PHYSICOCHEMICAL CHARACTERISTICS, POLYPHENOLS AND YELLOW FLAVONOIDS IN FRUITS OF COMMERCIAL AND WILD PITAYA SPECIES FROM THE BRAZILIAN SAVANNASABSTRACT -Although the high commercial potential of pitaya, there are yet few studies of physicochemical characterization of pitaya fruits, especially considering wild species from the Brazilian Savannas. This study was conducted with the objective to analyze the physico-chemical characterization, total polyphenols and yellow flavonoids of fruits of Hylocereus costaricensis, Hylocereus undatus, Selenicereus setaceus and Selenicereus megalanthus pitaya species. For the physico-chemical analyzes the percentage of soluble solids, pH value and total acidity. For the phenolic compounds quantification, total flavonoids and yellow polyphenols extractable were analyzed. Significant differences of chemical characteristics and the amount concentration of phenolic compounds were observed among pitaya species and basal, middle and apical portions of fruit. The species S. megalanthus had higher amounts of soluble solids. This characteristic is more pronounced in the middle part of the fruit of all species. There was significant difference of pH, with values ranging from 4.84 to 5.67. The pitaya fruits are classified as low acid fruits. The acid content ranged from 0.10% to 0.15% of citric acid. H. costaricensis presented higher amount of total polyphenols and yellow flavonoids, differing significantly from the other species.
ResumoO objetivo do trabalho foi caracterizar e estimar a variabilidade genética entre acessos de mandioca de indústria, com potencial de adaptação às condições do Cerrado do Brasil Central, por meio de caracteres quantitativos, qualitativos e marcadores moleculares de forma isolada e conjunta, bem como estimar a correlação entre as estimativas obtidas com base em cada grupo de caracteres. Dezesseis acessos de mandioca de indústria com potencial de adaptação às condições do Cerrado do Brasil Central foram avaliados a campo quanto a 11 caracteres quantitativos e 33 caracteres qualitativos, em experimento conduzido na Embrapa Cerrados. Os acessos foram também avaliados quanto a marcadores RAPD em laboratório. Posteriormente, foram estimadas as matrizes de dissimilaridade/distância genética entre os acessos por meio dos caracteres qualitativos, quantitativos e marcadores moleculares e pela análise conjunta dos dados. Além disso, foi estimada a correlação entre as matrizes. No grupo de acessos de indústria avaliados existe ampla variabilidade quanto aos caracteres quantitativos, qualitativos e moleculares aferidos. As únicas correlações significativas encontradas foram entre (i) a matriz de dissimilaridade estimada por meio da análise conjunta e a matriz de dissimilaridade estimada por meio de dados qualitativos (r = 0,52) e; (ii) a matriz de dissimilaridade estimada por meio da análise conjunta e a matriz de dissimilaridade estimada por meio de marcadores moleculares (r= 0,75). A fraca associação entre essas medidas indicou que a melhor estratégia para orientar ações de conservação e uso de germoplasma de mandioca de indústria é por meio de estudos de divergência genética com o emprego de marcadores moleculares, caracteres qualitativos e caracteres quantitativos de forma conjunta e complementar.
GENETIC VARIABILITY OF WILD AND COMMERCIAL PASSION FRUIT (Passiflora edulis Sims.) ACCESSIONS USING RAPD MARKERSABSTRACT -There are a great diversity of colors, sizes and aromas of fruits in wild accessions of P. edulis in Brazilian Savannah. These accessions are also important resistance sources against illness which can be incorpored in passionfruit breeding programs. In this work, the objetive was to evaluate the genetic variability in wild and commercial P. edulis accessions using RAPD markers. The genomic DNA of each accession was extracted and amplified using thirteen decamer primers (OPD-04, OPD-07, OPD-08, ) to obtain RAPD markers. These markers were transformed in binary matrix data to estimate genetic distances among accessions and to perform cluster and graphical dispersion analysis. A total of 187 markers were generated, and only 28 (14.97%) of them were monomorphic. The genetic distances among the 15 P. edulis accessions varied from 0.091 to 0.496. The molecular markers demonstrated the high genetic variability of the wild and commercial P. edulis accessions. The accessions with yellow fruits presented greater genetic distances in relation to the accessions with purple fruits. Lower genetic distances were verified among the accesses of the same geographical origin.
