This paper investigated the effect of cryoprotectant substances on Prochilodus lineatus embryos in an experimental incubator. The prospective study applied combinations of polyvinyl alcohol, hydroxyethyl cellulose, gelatin and fetal bovine serum with dimethyl sulfoxide and ethylene glycol in a new experimental incubator. The morphology of embryos, larval viability and the efficiency of experimental incubators in maintaining the quality of embryos were evaluated. This study demonstrates the efficient association between hydroxyethylcellulose and dimethyl sulfoxide as greater viability (p<0.05) was found for embryos (72.9 ± 23.9%). It should also be noted the permeation of cryoprotectants in embryos through the changes found in chorion diameter, embryo diameter and embryo volume comparing the treatments versus control group (water) (p<0.05), this results can help in future cryopreservation protocols. Although the temperature and oxygenation differed between the usual and experimental incubators (p<0.05), the results showed a high fertilization rate (79.6 ± 13.2%) for experimental incubators (p<0.05) which is sufficient for the maintenance of embryos in a cryoprotective environment and effectively allows experimentation for long periods with cryoprotectant substances. Cryopreservation of fish embryos has not been accomplished yet and new approaches are required for understanding the permeability of teleost embryos, especially in Brazilian native species.Este trabalho avaliou o efeito de substâncias crioprotetoras sobre embriões de Prochilodus lineatus em uma incubadora experimental. O estudo aplicou combinações de álcool polivinílico, hidroxietilcelulose, gelatina e soro fetal bovino com dimetilosulfóxido e etilenoglicol em uma nova incubadora experimental. Foram avaliadas a morfologia dos embriões, a viabilidade larval e a eficiência das incubadoras experimentais na manutenção da qualidade dos embriões. Este estudo demonstra a associação eficiente entre hidroxietilcelulose e dimetilsulfóxido pela maior viabilidade (p<0,05) encontrada para os embriões (72,9 ± 23,9%). Deve-se notar também a permeação dos crioprotetores nos embriões através das alterações encontradas no diâmetro córion, diâmetro do embrião e no volume do embrião comparando os tratamentos ao grupo controle (água) (p<0,05), estes resultados podem ajudar em futuros protocolos de criopreservação. Embora a temperatura e a oxigenação diferiram entre as incubadoras comuns e as experimentais (p<0,05), os resultados mostraram elevada taxa de fertilização (79,6 ± 13,2%) para incubadoras experimentais (p<0,05), o que é suficiente para a manutenção de embriões em ambiente crioprotetor e permite efetivamente a experimentação por longos períodos com substâncias crioprotetoras. A criopreservação de embriões de peixes ainda não foi realizada e novas abordagens são necessárias para a compreensão da permeabilidade dos embriões de teleósteos, especialmente em espécies nativas brasileiras.
ABSTRACT. The present study examined the effect of adding the antioxidants vitamins C and E on the quality of semen cryopreserved of curimba (Prochilodus lineatus). Semen samples from nine breeding males were collected for analysis of rate (%) and duration (s) of sperm motility. The sperm pool was diluted into three cryoprotective solutions: Solution A: 5 gr. Beltsville Thawing Solution (BTS) added with 5 mL dimethyl sulfoxide (DMSO) and distilled water to 100 mL; Solution B: Solution A + 0.0001 mg vitamin E; Solution C: Solution A + 0.0001 mg vitamin C. The vitamins C and E were not toxic to the semen of curimba. The sperm motility did not present any significant difference. However, the semen cryopreserved with vitamin C had a longer duration of the motility after thawing. Therefore, the vitamin C is recommended for the cryopreservation of the semen of curimba.Keywords: sperm motility, fish, reproduction, cryoprotective solution. Fish damage in hydroelectric stations has been extensively reported, but further investigations focusing on Brazilian migratory species, especially large fish, are needed. The present study describes swimbladder abnormalities in piapara Leporinus obtusidens, a migratory physoclistous species, collected downstream from the Funil Hydroelectric station dam, Minas Gerais, Brazil. The 117 specimens collected were dissected for swimbladder evaluation in the coelomic cavity. The abnormalities detected in 14 of the fish (12%) were swimbladder rupture with exteriorization (N = 12) and formation of sac-like projections (N = 2). The specimens that had swimbladder abnormalities weighed over 1300 g for both sexes. These abnormalities may be caused by bladder infections, however, inflammation process were not detected in these fish. The presence of anomalies in swimbladder suggests that the individuals were subjected to sudden depressurization, possibly related to passage through the turbines. Further comparative studies are needed to corroborate this hypothesis.
Os danos que as usinas hidrelétricas causam na fauna aquática tem sido relatados, mas novos estudos focando as espécies nativas, especialmente os peixes de grande porte são necessários. O presente estudo descreve as anomalias de bexiga natatória em piapara Leporinus obtusidens, uma espécie de peixe migratório, coletados a jusante da barragem da estação Hidrelétrica do Funil, Minas Gerais, Brasil. Os 117 exemplares coletados foram dissecados para avaliação da bexiga natatória na cavidade celomática. As anormalidades detectadas em 14 dos peixes capturados (12%) foram ruptura do tecido com exteriorização da bexiga natatória (N = 12) e formação de projeções da bexiga (N = 2). Os exemplares que apresentavam anormalidades de bexiga, possuíam peso superior a 1300 g em ambos os sexos. Este tipo de anomalia pode ser causado por processo infeccioso da bexiga, no entanto, não foram detectados processos inflamatórios nesses peixes. O quadro de anomalias em bexiga natatória em piapara sugere que os indivíduos foram submetidos a uma brusca despressurização, possivelmente relacionada à passagem pelas turbinas. Outros estudos comparativos são necessários para corroborar esta hipótese
This study determined the age and its correlation with biometric variables and reproductive indices of piapara Leporinus obtusidens caught at the Funil Reservoir. We caught 133 specimens between September 2006 and August 2007. Weight, total length, standard length, depth, height, head length and radius of the scales were measured. The sex was determined by macroscopic examination of gonads. The age of each fish was determined by analyzing the growth rings on the scales. Seasonal differences in biometric variables were tested using the NK test at 5%. The correlation of age and radius of the scales with the biometric variables and reproductive indices were analyzed using the SAEG software. Fish individuals ranging from 3 to 14 years of age were caught. In the spring and summer, smaller fish were captured compared to those caught during the fall and winter. Females tended to have higher weight and morphometric values when compared to males. Age and radius of the scales were correlated with biometric variables in L. obtusidens. It can be concluded that the biometric variables of this species can be used as an indicator of age.
RESUMO:A endoscopia pode facilitar o manejo em piscicultura e possibilita a realização da sexagem de forma precisa, mesmo que seu uso ainda seja restrito. A curimba é uma espécie de peixe reofílica, utilizada em experimentos como espécie modelo para peixes migradores. Este trabalho teve como objetivo viabilizar a utilização do endoscópio rígido na sexagem de reprodutores de curimba e determinar se a técnica altera os índices reprodutivos. Foram examinados 16 curimbas, nos quais foi identificado o sexo através da introdução do endoscópio pelo poro urogenital para visualização das gônadas. O endoscópio rígido utilizado (modelo Multi-Purpose Rigid™, Karl Storz, Germany) apresentava ângulo de 30º, com 2,7 mm de diâmetro e 18 cm de comprimento. Foi realizada a indução hormonal 20 dias após a aplicação da técnica, as fêmeas receberam duas doses de extrato bruto de hipófise de carpa (EBHC) de 0,4 e 4,0 mg/kg de peixe, com intervalo de 12 horas e os machos receberam uma única dose de 4,0 mg/kg de peixe de EBHC, no momento da segunda aplicação nas fêmeas. Após 12 horas, procedeu-se a extrusão dos gametas, sendo retirada uma amostra de sêmen para avaliação da morfologia, taxa (%) e duração (segundos) da motilidade e concentração espermática. Para as fêmeas realizou-se a avaliação do peso da desova e coleta de uma amostra dos ovócitos para avaliar a posição da vesícula germinativa e o diâmetro dos ovócitos. Pode-se observar que 100% dos machos espermiaram e 62% das fêmeas desovaram. Além disso, todos os índices reprodutivos foram considerados dentro dos padrões esperados para a espécie. Conclui-se que a endoscopia pode ser utilizada de maneira eficiente na sexagem e na avaliação do grau de maturação gonadal, antes da piracema, sem alterar a eficiência reprodutiva dos peixes. The curimba is a rheophilic species of fish, used in experiments as a model species for migratory fish. This study aimed to enable the use of the endoscope in sexing breeding curimba and determine whether the technique alters the reproductive rates. Curimbas 16 were examined, in which sex was identified by the introduction of the pore urogenital endoscope for viewing the gonads.
