Penelitian ini bertujuan untuk mengetahui ektoparasit yang terdapat pada ikan koi yang dijual di Jalan Bintang Medan. Ektoprarasit diketahui dapat menyebabkan penyakit pada ikan. Hal ini akan berdampak pada ikan lainnya. Kerusakan yang diakibatkan oleh ektoparasit ini akan meningkatkan intensitas penyakit pada ikan. Luka yang ditimbulkan oleh adanya ektoparasit pada ikan menjadi jalan masuk bagi kuman lain untuk menyerang ikan. Sampel yang digunakan ialah tiga ekor ikan koi yang mana ektoparasit yang menginfeksi ikan di amati lendir pada tubuh dan sirip ikan mas koki. Identifikasi ektoparasit dilakukan dengan menggunakan literatur. Ektoparasit yang teridentifikasi ialah Ichthyophthirius sp., Oodinium sp., Costia Sp. Dengan jumlah parasit terbanyak ialah spesies Oodinium sp.Kata Kunci : Ektoparasit, ikan koi, identifikasi.
Essential oil is a volatile substance and has a distinctive scent that can be found on plants. Lime peel oil is one part of the plant which can be processed to make an essential oil so that it can be used as a natural fragrance in soap making. The purpose of this study is to produce essential oil from lime peel, which can be used as a substitute for synthetic fragrance ingredients in soap making. The other purpose is to obtain the optimum method, type of solvent, and the optimum extraction time to produce essential oil. The raw material used in this study is lime peel powder sized 50 mesh. The water content of raw material on lime peel powder is 76,5 %. The extraction method used in the research is maceration with stirring equal to 150 rpm and Soxhlet with extraction time for 6, 9, and 12 hours. In this study, the ratio of raw material to the solvent is 1:10, and the type of solvent used is hexane, ethanol, and distilled water. The result in this study shows that Soxhlet is the optimum method, which obtains yield equal to 6,15% for 12 using hexane as solvent.
The resistance of pathogenic bacteria to antibiotics is increasing due to antibiotics with incorrect doses, wrong diagnostics, and the wrong target. Bacteria that have been resistant to several antibiotics are called multi-drug resistant organisms (MDRO) bacteria. Bacterial resistance to some antibiotics requires alternative herbal treatments, one of which is the Chromolaena odorata L. Research must therefore be conducted on the antibacterial activity of the ethanol extract fraction of C. odorata L. leaves for MDRO bacteria, such as Staphylococcus lugdunensis methicillin-resistant Staphylococcus aureus (MRSA), Pseudomonas aeruginosa extended-spectrum beta-lactamase (ESBL), and Klebsiella pneumoniae ESBL. This study aims to determine the antibacterial activity of the ethanol extract of Kirinyuh leaves (C. odorata L.) and the antibacterial activity of the fractionation against MDRO bacteria. Test results of kirinyuh leaf ethanol extract for S. lugdunensis MRSA, P. aeruginosa ESBL, and K. pneumoniae ESBL bacteria each resulted in an inhibition zone with an average diameter of 11.6 mm (strong), 11.5 mm (strong), and 11.13 mm (strong), respectively. Testing the antibacterial activity of the ethanol fraction against MDRO bacteria can show antibacterial activity against all tested bacteria, namely Fraction 5. The results of the antibacterial activity of fraction 5 against K. pneumoniae ESBL, P. aeruginosa ESBL, and S. lugdunensis MRSA bacteria with the formation of inhibition zones formed of 10.2 mm (strong), 8.8 mm (moderate), and 7.9 mm (moderate), respectively. The results of thin-layer chromatography showed that the secondary metabolites contained in the fifth fraction were terpenoids, steroids, and flavonoids.
This research purpose is to show the inhibition of the extract of ethanol and ethyl acetate stem bark and leaf of Raru according to the causes of bacterial E-coli (ATCC 25922) and St Aureus (ATCC 25923) with variation concentration 10%, 5%, 2.5% and 1.25%. Methods used were diffusion with disk blank paper and disk cloramphenicol as a control. The highest result observed on 10% concentration among the variation. Penelitian ini bertujuan untuk mengetahui jenis-jenis parasit yang menginfeksi pada insang ikan mas koki (Carassius auratus). Penelitian ini dilaksanakan pada bulan Mei 2017, di Laboratorium Biologi Universitas Negeri Medan. Pemeriksaaan ektoparasit dilakukan dengan menggunakan metode pengapungan, dimana sampel insang direndam didalam larutan garam fisiologis yang kemudian dibiarkan selama 5 menit. Dari hasil penelitian ditemukan 2 jenis parasit yang menginfeksi insang ikan mas koki (Carassius auratus), yaitu Unitubulotestis sardae dan Dactylogyrus sp. ABSTRAK Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri ekstrak metanol daun kirinyuh (Chromolaena odorata) terhadap bakteri MDR dengan metode KLT-bioautografi. Bakteri uji yang digunakan adalah bakteri MDR S.lugdunensis MRSA, K.pneumoniae ESBL dan P.aeruginosa ESBL. Metode ekstraksi dilakukan dengan cara maserasi menggunakan pelarut metanol p.a.Untuk pengujian KLT-bioautografi, ekstrak metanol daun kirinyuh ditotolkan pada plat KLT kemudian dielusi dengan eluen metanol:kloroform (9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7, 2:8 dan 1:9), kemudian plat KLT ditempelkan pada media MHA yang telah diinokulasikan ke 3 bakteri MDR dan diinkubasi selama 24 jam. Zona hambat yang terbentuk dilakukan identifikasi senyawa menggunakan pereaksi semprot FeCl3, dragendorff, uap amoniak, SbCl3 dan Lieberman bouchardat. Hasil uji KLT-bioautografi menunjukkan bahwa ekstrak metanol daun kirinyuh dapat menghambat pertumbuhan bakteri yang ditandai terbentuknya zona bening pada EMDKF3, EMDKF7b, EMDKF9b dengan masing-masing nilai Rf 0,47 cm (fenol), 0,87 cm (flavonoid), 0,92 cm (flavonoid) pada bakteri S.lugdunensis MRSA dan EMDKF2, EMDKF7a, EMDKF7c dengan masing-masing nilai Rf 0,6 cm (flavonoid), 0,57 cm (alkaloid), 0,96 cm (flavonoid) pada bakteri K.pneumoniae ESBL, sedangkan pada bakteri P.aeruginosa ESBL ekstrak metanol daun kirinyuh tidak memiliki aktivitas antibakteri yang ditunjukkan dengan tidak ada zona bening yang terbentuk. Senyawa yang berperan sebagai antibakteri terhadap bakteri S.lugdunensis MRSA dan K.pneumoniae ESBL adalah senyawa golongan alkaloid, flavonoid dan fenol. Kata Kunci: Chromolaena odorata, Bakteri MDR, Aktivitas Antibakteri, KLT Bioautografi ABSTRACT This study aims to determine the antibacterial activity of methanol extract of kirinyuh leaves (Chromolaena odorata) against MDR bacteria by TLC-bioautography method. The test bacteria used were MDR S.lugdunensis MRSA, K.pneumoniae ESBL and P.aeruginosa ESBL. The extraction method is carried out by maceration using methanol solvent p.a. for TLC-bioautographic testing, the methanol extract of the kirinyuh leaves was poured on the TLC plate then eluted with methanol:chloroform eluent (9:1, 8:2, 7:3, 6:4, 5:5, 4:6, 3:7, 2:8 and 1:9), then the TLC plate was attached to the MHA media which had been inoculated to 3 MDR bacteria and incubated for 24 hours. Inhibition zones formed were identified using compound spray reagents FeCl3, dragendorff, ammonia vapor, SbCl3 and Lieberman bouchardat. The TLC-bioautographic test results showed that the methanolic extract of kirinyuh leaves can inhibit bacterial growth marked by the formation of clear zones in EMDKF3, EMDKF7b, EMDKF9b with each Rf value of 0,47 cm (phenol), 0,87 cm (flavonoids), 0,92 cm (flavonoids) in S.lugdunensis MRSA and EMDKF2, EMDKF7a, EMDKF7c bacteria with Rf values of 0,6 cm (flavonoids), 0,57 cm (alkaloids), 0,96 cm (flavonoids) in K.pneumoniae ESBL, whereas in P.aeruginosa ESBL bacteria the methanolic extract of kirinyuh leaves had no antibacterial activity as indicated by no clear zone formed. The compounds that act as antibacterial against the bacteria S.lugdunensis MRSA and K.pneumoniae ESBL are alkaloid, flavonoid and phenol compounds.Keywords: Chromolaena odorata, MDR Bacteria, Antibacterial Activity, TLC Bioautography
Menghadapi tantangan abad 21, siswa diharapkan menguasai keterampilan berpikir kritis. Pembelajaran berbasis masalah mampu melatih keterampilan berpikir. Salah satu perangkat pembelajaran yang mampu meningkatkan keterampilan berpikir adalah LKPD berbasis PBL. Penelitian ini merupakan penelitian R&D dengan mengembangkan LKPD. Model pengembangan yang digunakan adalah 4-D dengan tahapan Define, Design, Develop, dan Disseminate. Instrumen penelitian adalah lembar validasi ahli materi, pembelajaran, dan desain, angket respons guru, angket respon siswa, serta soal pre-test dan post-test. Produk di validasi oleh ahli materi dengan persentase 90,63 % (sangat layak), pembelajaran dengan persentase 95% (sangat layak), dan desain dengan persentase 88,64% (sangat layak). Hasil respon siswa dengan persentase 90% (sangat layak) pada uji perorangan dan 86% (sangat layak) pada uji kelompok terbatas, hasil respon guru dengan persentase 92,65% (sangat layak). LKPD mampu meningkatkan keterampilan berpikir kritis dengan rata-rata skor pada pretest sebesar 40,26 (rendah) dan posttest sebesar 77,06 (tinggi) dengan N-Gain sebesar 0,57 (sedang).
Tujuan dari penelitian ini adalah untuk mengetahui pengaruh frekuensi penggantian sekam terhadap kehadiran ektoparasit pada mencit yang dilakukan di Laboratorium Biologi Universitas Negeri Medan. Metode yang digunakan dengan memakai mencit dengan frekuensi penggantian sekam sekali dua minggu dengan 3 kali pengulangan dan menggunakan mencit peliharaan dengan frekuensi penggantian sekam sekali dua hari sebagai pembanding kehadiran ektoparasit pada mencit. Badan Mencit di usap dengan kapas yang sudah ditetesi alkohol dan disisir ke atas alkohol 70% sebagai koleksi ektoparasit kemudian di preservasi untuk di identifikasi. Hasil yang diperoleh terdapat ektoparasit jenis tungau dan pinjal untuk jenis tungau berasal dari anggota Laelapidae dan Xenopsylla cheopis pada mencit dengan frekuensi penggantian sekam sekali dua minggu, untuk itu dapat diperoleh kesimpulan bahwa kehadiran ektoparasit berpengaruh dengan frekuensi penggantian sekam.
