Bovine tuberculosis is an important infectious disease caused by Mycobacterium bovis, which is responsible for considerable economic losses. This disease constitutes a serious public health problem. Control programs in most countries, including Brazil, are based on the identification and slaughter of infected animals, as defined by the skin tuberculin test, which has its constraints. In the present study, the recombinant proteins CFP-10, ESAT-6, Mb0143, MPB83, PE5, PE13, TB10.4, TB15.3 and a chimera of ESAT-6/MPB70/MPB83 (fusion protein) were tested as ELISA antigens for the diagnosis of bovine tuberculosis. The proteins were produced in Escherichia coli, purified and tested in ELISAs with sera from 126 cattle having tested negative in the comparative intradermal tuberculin test (CITT) and 107 sera from cattle having tested positive in the CITT. Also, 236 sera from two BTB-free beef cattle herds were tested. Among the proteins tested, only the ESAT-6/MPB70/MPB83 chimera demonstrated satisfactory agreement with the CITT (kappa index: 0.688), reflecting in 83.2% sensitivity and 86.5% specificity. The ELISA absorbances of the cattle sera from BTB-free herds showed similar levels to those of CITT positive cattle, probably as the result of successive skin tuberculinizations to define the BTB-free status of the herds. However, the ELISA with the ESAT-6/MPB70/MPB83 chimera was useful to discriminate BTB positive and negative cattle in herds prior to the tuberculin skin test.
Interest in edible oil extraction processes is growing interest because the final nutritional quality of the extracted oil depends on the procedure used to obtain ir. In this context, a domestic cold oil press machine is a valuable tool that avoids the use of chemicals during oil extraction, in an environmentally friendly way. Although babassu (Attalea speciosa) oil is economically important in several Brazilian regions due to its nutritional and healthy features, few studies have been conducted on the chemical composition and stability of babassu oils extracted by cold pressing. Babassu oil’s major constituents are saturated fatty acids (~86.42%), with the most prevalent fatty acids being lauric (~47.40%), myristic (15.64%), and oleic (~11.28%) acids, respectively, within the recommended range by Codex Alimentarius, presenting atherogenicity and thrombogenicity indexes favorable for human consumption. Peroxide value, Rancimat, and TGA/DSC results indicated that babassu oil is stable to oxidation. Also, macro- (Na, K, Ca, Mg, P) and micro-elements (Fe, Mn, Cr, Se, Al, and Zn) of babassu oil were determined, revealing levels below the tolerable upper intake level (ULs) for adults. These findings demonstrated that cold-press extraction using a domestic machine yielded a high-quality oil that kept oil chemical composition stable to oxidation with natural antioxidants.
and Pb by inductively coupled plasma-optical emission spectrometry (ICP OES) was performed on dry matter and decoctions of the medicinal plants Cordia salicifolia, Chiococca alba (L.) Hitchc., and Echites peltata used as an appetite suppressant and diuretic in Brazil. e accuracy of the measurements was analyzed by the spike recovery test. Results showed that the concentration of these seven metals (Cd, Co, Cr, Cu, Fe, Na, and Zn) in dry plant samples is below the oral concentration of elemental impurities established by the United States Pharmacopoeia Convention (USP). However, there are no concentration limits for Fe, Na, and Zn established by the USP in drug substances and excipients. Levels higher than the recommended value by the USP were observed for Pb and the lowest for Cd, Co, Cr, and Cu, both in dried plant samples and their decoctions. In the decoctions prepared from these plants were found elements such as Cd, Co, Cr, Cu, Fe, Na, Zn, and Pb. In the decoction prepared from 40 g C. salicifolia leaves and 40 g C. alba wood, the content of Cd is above the oral daily exposure value set by the USP. Hazard index (HI) for decoctions prepared from these plants exceeded the threshold (1). Given the uncertainties associated with the estimates of toxicity values and exposure factors, futures researches should address the possible toxicity in humans. Uncontrolled selling and long-term ingestion of medicinal plants can cause toxicity and interfere with the effect of drugs. Limited knowledge on the interaction potential of medicinal plants poses a challenge and public health problem in Brazil and other countries.
Antioxidants have drawn the attention of the scientific community due to being related to the prevention of various degenerative diseases. The antioxidant capacity has been extensively studied in vitro, and different methods have been used to assess its activity. However, the main issues related to studying natural antioxidants are evaluating whether these antioxidants demonstrate a key role in the biological system and assessing their bioavailability in the organism. The majority of outcomes in the literature are controversial due to a lack of method standardization and their proper application. Therefore, this study aims to compile the main issues concerning the natural antioxidant field of study, comparing the most common in vitro methods to evaluate the antioxidant activity of natural compounds, demonstrating the antioxidant activity in biological systems and the role of the main antioxidant enzymes of redox cellular signaling and explaining how the bioavailability of bioactive compounds is evaluated in animal models and human clinical trials.
Reprodutive system of Anadenanthera peregrina and Vochysia haenkiana in a fragment of Cerrado forest from Chapada dos Guimarães -MT, Brazil RESUMO O presente estudo objetivou avaliar o sistema reprodutivo das espécies Anadenanthera peregrina (L.) Speg. e Vochysia haenkiana (Spreng.) Mart. Foram estudados aspectos do sistema de cruzamento, complementados pelas observações de maturação dos frutos e a germinação das sementes em função dos tipos de cruzamentos testados. As síndromes de dispersão das sementes foram também avaliadas através das características morfológicas dos frutos e sementes. O estudo foi realizado em um fragmento de cerrado com aproximadamente 32 ha, no município de Chapada dos Guimarães, MT, a 15 0 10 S, 55 0 35 W, e à altitude de 450m. O delineamento experimental utilizado na determinação do sistema de cruzamento para cada espécie foi o de blocos ao acaso, com seis tratamentos para a espécie Vochysia haenkiana e cinco tratamentos para Anadenanthera peregrina, ambas com três repetições. O sistema de cruzamento detectado para as duas espécies foi o preferencialmente alogâmico, porém, com a possibilidade, não desprezível, de produção de frutos/ sementes por autofertilização, sugerindo um aumento gradual de autocompatibilidade genética como estratégia de geração de descendência no ambiente florestal fragmentado. A polinização cruzada em Anadenanthera peregrina é favorecida tanto pelo alto grau de auto-incompatibilidade genética quanto pela protandria detectada. O padrão de maturação dos frutos e sementes e o período de dispersão das sementes foram distintos em ambas as espécies, ocorrendo, entretanto, durante ou no final da estação seca, facilitando a distribuição dos propágulos anemocóricos. Bredemeyera floribunda Willd ( B. floribunda ) is a medicinal plant used by the Brazilian rural population to treat snakebites, but there are no data on the elemental composition and stability of the emulsion of extract of this plant. In this article, we present for the first time data on contents obtained the roots of the medicinal plant B. floribunda by using inductively coupled plasma optical spectrometry (ICP OES), as well as the data on preliminary stability tests of emulsions formulated with aqueous extract of this plant. The first set of data shows a total of 13 elements (Al, Ca, Cr, Fe, K, Mg, Mn, Na, Ni, P, Cu, Se and Zn) detected in the roots of the plant. The second set of data shows that the extract data of the B. floribunda roots presented an emulsifying potential. In addition, the article provides information on the heating program for microwave oven digestion and ICP OES operating conditions. The data presented make way for nutritional and toxicological studies involving the roots of B. floribunda .
