OBJECTIVE: To assess the evolution of motor function in patients with Duchenne muscular dystrophy (DMD) treated with steroids (prednisolone or deflazacort) through the Motor Function Measure (MFM), which evaluates three dimensions of motor performance (D1, D2, D3). METHODS: Thirty-three patients with DMD (22 ambulant, 6 non-ambulant and 5 who lost the capacity to walk during the period of the study) were assessed using the MFM scale six times over a period of 18 months. RESULTS: All the motor functions remained stable for 14 months in all patients, except D1 for those who lost their walking ability. In ambulant patients, D2 (axial and proximal motor capacities) motor functions improved during six months; an improvement in D3 (distal motor capacity) was noted during the total follow-up. D1 (standing posture and transfers) and total score were useful to predict the loss of the ability to walk. CONCLUSIONS: The use of the MFM in DMD patients confirms the benefits of the steroid treatment for slowing the progression of the disease.
BackgroundDuchenne muscular dystrophy (DMD) is a sex-linked inherited muscle disease characterized by a progressive loss in muscle strength and respiratory muscle involvement. After 12 years of age, lung function declines at a rate of 6 % to 10.7 % per year in patients with DMD. Steroid therapy has been proposed to delay the loss of motor function and also the respiratory involvement.MethodIn 21 patients with DMD aged between seven and 16 years, the forced vital capacity (FVC) and the forced expiratory volume in one second (FEV1) were evaluated at three different times during a period of two years.ResultsWe observed in this period of evaluation the maintenance of the FVC and the FEV1 in this group of patients independently of chronological age, age at onset of steroid therapy, and walking capacity.ConclusionThe steroid therapy has the potential to stabilize or delay the loss of lung function in DMD patients even if they are non-ambulant or older than 10 years, and in those in whom the medication was started after 7 years of age.
Severe Acute Respiratory Syndrome of Coronavirus-2 (Sars-CoV-2) is the causative agent of the new Coronavirus Disease (COVID-19) responsible for the current pandemic that threatens global health. Although some anti-COVID-19 therapeutic agents are under investigation, there is still no evidence of antiviral action against Sars-CoV-2. Research in the literature describes the success of probiotics in the treatment of viral infections from their byproducts, known as postbiotics, such as exopolysaccharides, hydrogen peroxide, and different bacteriocins. Based on these reports, we describe the main postbiotics that present antiviral actions against different viruses with a view to suggesting their use as possible therapeutic agents for COVID-19. The revised data show promising effects for using postbiotics as efficient vehicles against various types of viruses. However, further investigation of the underlying mechanisms is required for their indication against Sars-CoV-2 and other Sars-CoV infections.
O probiótico está relacionado a um suplemento alimentar, enquanto que os produtos vivos bioterapêuticos - PVBs são considerados medicamentos biológicos, ambos contêm micro-organismos vivos com substâncias ativas e que beneficiam o hospedeiro. O Enterococcus faecium apresenta muitas propriedades interessantes e funcionais para saúde humana e animal, entre elas a capacidade antimicrobiana, que ajuda em doenças de intoxicação alimentar como a Campilobacteriose. As espécies Campylobacter jejuni e C. coli, têm sido descritas neste contexto. Portanto, pela necessidade de desenvolver alternativas que permitam garantir a segurança dos produtos consumidos pela população, este trabalho propõe uma estratégia complementar para reduzir ou eliminar o Campylobacter por meio do uso de Enterococcus faecium EF137V, isolado de queijo “Coalho” artesanal de Pernambuco-Brasil. As análises foram realizadas in vitro. Dadas as condições adversas, como pH ácido e presença de sais biliares, nas quais EF137V foi exposto, verificou-se que a bactéria apresentou resistência a condições gastrointestinais, aderência às células epiteliais, atividade antimicrobiana pelos seus compostos metabólicos contra cepas de referência de C. jejuni e C. coli, bem como atividade antagonista, o que sugere que ambas as espécies não serão capazes de crescer desordenadamente na microbiota intestinal do hospedeiro, podendo assim controlar a esses microrganismos na comunidade bacteriana em frangos de corte e evitando novas infecções em humanos. Pode-se concluir que o EF137V é um PVB e probiótico promissor, apresentando grande capacidade para aplicações de biotecnologia.
Objetivou-se com este estudo avaliar a eficácia das enterocinas produzidas pela Enterococcus faecium 141v e 137v, probióticos, oriundos do queijo de coalho artesanal de Pernambuco sobre o desenvolvimento do câncer colorretal em estudo in vivo. Como metodologia foram realizados testes de sensibilidade a protease com a finalidade de comprovar suas propriedades proteicas, além de sua atividade biológica como antimicrobiano. A ultrassonografia foi utilizada para avaliar o efeito das enterocinas sobre o câncer colorretal induzido por 1,2 Dimetilhidrazina (DMH ) em ratos Wistar. Os resultados obtidos in vitro demonstraram a natureza peptídica das enterocinas bem como sua eficácia quanto à atividade antimicrobiana. Quanto aos resultados in vivo avaliados na ultrassonografia, foi possível observar a diminuição na espessura da parede intestinal dos ratos que foram submetidos a indução do câncer colorretal e tratados com as enterocinas, podendo ser explicada pela possível estimulação apoptótica, visando a inibição de células cancerígenas. O presente estudo trouxe aspectos importantes a respeito da utilização das enterocinas para fins terapêuticos, sobretudo na prevenção e controle do câncer colorretal em modelo animal. Também destacou o uso da ultrassonografia para avaliar o efeito das enterocinas sobre o desenvolvimento da doença, mostrando ser um método que considera o bem-estar animal por ser menos invasivo e eficaz. Os resultados obtidos neste trabalho reforçam a necessidade de estudos que aprofundem o tema. Além disso, agregam conhecimentos em diversas áreas como biotecnologia, microbiologia, medicina, nutrição e farmacologia.
